1 Website: thermofisher.com/ebioscience Customer Service (US): thermofisher.com/contactus IL-4 Monoclonal Antibody (11B11), Functional Grade, ebioscience Catalog Number Details Size 50 µg Host/Isotope Class Type Clone Conjugate Form Concentration Purification Rat / IgG1, kappa Monoclonal Antibody 11B11 Functional Grade Liquid 1 mg/ml Affinity chromatography Storage buffer PBS, ph 7.2 Contains Storage Conditions no preservative 4 C Product data sheet Species Reactivity Tested species reactivity Mouse Published species reactivity Mouse Tested Applications Dilution * ELISA (ELISA) µg/ml Functional Assay (FN) Assay-Dependent Published Applications Flow Cytometry (Flow) See 6 publications below ELISA (ELISA) See 2 publications below Miscellaneous PubMed (MISC) See 2 publications below Immunofluorescence (IF) See 1 publications below Functional Assay (FN) See 2 publications below * Suggested working dilutions are given as a guide only. It is recommended that the user titrate the product for use in their own experiment using appropriate negative and positive controls. Product specific information Description: The 11B11 antibody reacts with mouse interleukin-4 (IL-4), a 14 kd cytokine. The 11B11 antibody is a neutralizing antibody. Mouse IL-4, also known as BCDF, BCGF-1 and BSF-1, is secreted by activated Th2 and natural killer T cells, and to a lesser extent by Th1 and mast cells. IL-4 is speciesspecific and promotes proliferation and differentiation of B cells. IL-4, in conjunction with other signals such as CD40, induces Ig isotype switching of B cells and the upregulation of surface MHC class II and CD23. Applications Reported: This 11B11 antibody has been reported for use in cytokine neutralization, ELISPOT, and ELISA cap. Applications Tested: The Functional Grade Purified 11B11 antibody has been tested by LAL assay to verify low endotoxin levels, by ELISA, and in bioassay for neutralization of mouse IL-4 bioactivity. The 11B11 antibody at µg/ml has been found to inhibit by 50% the biological effects of 1.0 ng/ml mouse IL-4 in a CTLL-2 cell proliferation assay. Please refer to Best Protocols for more information. The 11B11 antibody has been tested as the capture antibody in a sandwich ELISA for analysis of mouse interleukin-4 (IL-4) in combination with the biotinylated BVD6-24G2 (cat ) antibody for detection and recombinant mouse IL-4 (cat ) as the standard. A suitable range of concentrations of this antibody for ELISA capture is 1-4 µg/ml. A standard curve consisting of doubling dilutions of the recombinant standard over the range of 500 pg/ml - 4 pg/ml should be included in each ELISA plate. Endotoxin: Less than ng/ug antibody as determined by the LAL assay. Storage and handling: Use in a sterile environment. Filtration: 0.2 µm post-manufacturing filtered. Purity: Greater than 90%, as determined by SDS-PAGE. Endotoxin Level: Less than ng/µg antibody, as determined by LAL assay. Aggregation: Less than 10%, as determined by HPLC.

2 Background/Target Information The protein encoded by this gene is a pleiotropic cytokine produced by activated T cells. This cytokine is a ligand for interleukin 4 receptor. The interleukin 4 receptor also binds to IL13, which may contribute to many overlapping functions of this cytokine and IL13. STAT6, a signal transducer and activator of transcription, has been shown to play a central role in mediating the immune regulatory signal of this cytokine. This gene, IL3, IL5, IL13, and CSF2 form a cytokine gene cluster on chromosome 5q, with this gene particularly close to IL13. This gene, IL13 and IL5 are found to be regulated coordinately by several long-range regulatory elements in an over 120 kilobase range on the chromosome. Two alternatively spliced transcript variants of this gene encoding distinct isoforms have been reported.

3 Product Images For IL-4 Monoclonal Antibody (11B11), Functional Grade, ebioscience IL-4 Antibody ( ) in ELISA Splenocytes from 8 week old mice pre-immunized with OVA/IFA were recallactivated with OVA antigen for 36 hours in Mouse IL-4 ELISPOT assay. Control well is medium alone.
