Three common excitation/emission wavelength configurations are offered. Built-in memory saves data, which can be retrieved for data analysis by a USB interface. 2 Ammonia Ammonium 1. DNH to 1 mm.3 mm (.5 ppm) Urine Soil extracts and other environmental R 2 = Nominal (NH 3 /NH 4 +, mm) Important: prior to assay, bring the assay reagents (cat# DNH3-2) to room temperature. 1. Prepare 1mM Ammonia Standard by mixing 5 µl provided 2 mm MH 4 Cl and 95 µl H 2 O in an Eppendorf tube. per tube: 1 µl Assay Buffer, 4 µl Reagent A and 4 µl Reagent B. In separate mini glass tubes (cat#: MGLTB1), add 1 µl H 2 O (Blank), 1 µl 1 mm NH 4 Cl ( Std ), and 1 µl Sample. Then add 1 µl Working Reagent to each tube and mix. Incubate for 15 min at room temperature in the dark. Press left arrow on "<- Std -> ", until the window shows "1.". The ammonia/ammonium concentration (mm) will be displayed in the window. Prepare 1 µl 1, ng/ml DNA by mixing 1 µl provided 1 µg/ml DNA standard and 9 µl H 2 O in an Eppendorf tube. 2. In separate mini-glass tubes (cat#: MGLTB1), add 1 µl H 2 O (Blank), 1 µl 1, ng/ml DNA ( Std ), and 1 µl Sample. Then add 1 µl Reagent. Incubate for 5 min. Press left arrow on "<-Std -> ", until the window shows "1.". The DNA concentration (ng/ml) will be displayed in the window. Record the data, or press "Save" to save the data for later retrieval. 3 Formaldehyde Measured (µm) DFOR-1.9 to 1 µm.9 µm (27 ppb) Formaldehyde extracted from various. R 2 = Nominal (Formaldehyde,µM) Important: prior to assay, bring the assay reagents (cat# DFOR- 1) to room temperature. 1. First prepare a 35 mm Formaldehyde stock by diluting 4 µl of the provided Standard with 149 µl H2O. Next, dilute 4 µl of the 35 mm Formaldehyde with 1396 µl H2O to make a 1 µm Standard. per tube: 33 µl Reagent A and 22 µl Reagent B. In separate mini glass tubes (cat#: MGLTB1), add 5 µl H2O Blank, 5 µl 1 µm Standard ( Std ), and 5 µl Sample. Then add 5 µl Working Reagent mix by pipetting. Incubate for 3 min in the dark. Press left arrow on "<- Std -> ", until the window shows "1.". Place the Std tube into the Sample holder. Press "Measure". The reader shows "Calibrate Finished". The Reader is now calibrated. The Formaldehyde concentration (µm) will be displayed in the window. The assay is based on a kinetic reaction, it is important to keep the incubation times identical between different assay tubes. 3. Prior to assay, bring the assay reagents (cat# QFPR-2) to room temperature. 1. Prepare 1 µg/ml Standard by mixing 1 µl of the provided 1 mg/ml BSA standard with 9 µl H2O or sample buffer. 2. In separate mini-glass tubes (cat#: MGLTB1), add 1 µl H 2 O or Sample Buffer ("Blank"), 1 µl 1 µg/ml BSA Standard ("Std"), and 1 µl Sample. Then add 9 µl Reagent to each tube and mix. Incubate for 1 min in the dark. Press left arrow on "<-Std -> ", until the window shows "1.". The protein concentration (µg/ml) will be displayed in the window. Record the data, or press "Save" to save the data for later retrieval. 4 Cholesterol Measured (mg/dl) 3 E2CH to 3 mg/dl 1 mg/dl (26 µm, 1 ppm) <2% Serum Plasma Other biological R 2 = Nominal (Cholesterol, mg/dl) Important: prior to assay, bring the assay reagents (cat# E2CH- 1) to room temperature. Keep enzyme tubes cold during the assay. 1. Mix 5 µl provided cholesterol standard with 145 µl Assay Buffer. Dilute this mixture by mixing 5 µl with 495 µl Assay Buffer. Label this 3-fold diluted standard as 1 mg/dl. Dilute serum/plasma 1:1 in the Assay Buffer. per tube: 55 µl Assay Buffer, 1µL Enzyme Mix, 1µL Dye Reagent. In separate mini-glass tubes, add 5 µl Assay Buffer ("Blank"), 5µL 1mg/dL Standard ("Std"), and 5 µl Sample. Then add 5µL Working Reagent to each tube. Incubate for 3 min at room temperature in the dark. Press left arrow on "<- Std -> ", until the window shows "1.". The Cholesterol concentration (mg/dl) will be displayed in the window. Record the data, or press "Save" to save the data for Sample in Assay Buffer and repeat assay. Cholesterol Assay Kit (cat# E2CH-1): 2 ml Assay Buffer 12 µl Enzyme Mix 12 µl Dye Reagent Sufficient for 1 tests (cat# FL5359) one bag of 1 Tubes. Glycerol Measured (µm) 1 EGLY µm 2 µm (18 mg/dl,.18 ppm) <8% Serum Plasma Other R 2 = Nominal (Glycerol, µm) Important: prior to assay, bring the assay reagents (cat# EGLY- 2) to room temperature. Keep enzyme tubes cold during the assay. 1. Prepare 1 mm Standard by mixing 5 µl of the provided standard with 495 µl distilled H 2 O. Then mix 5 µl of the 1mM Standard with 45 µl H 2 O to obtain 1 µm Glycerol Standard. In separate mini-glass tubes (cat#: MGLTB1), add 1 µl H 2 O ("Blank"), 1 µl 1 µm Glycerol Standard ("Std"), and 1 µl Sample. 2. Prepare enough Working Reagent for all assay tubes, by mixing per tube: 1 µl Assay Buffer, 2 µl Enzyme Mix, 1 µl ATP and 1 µl Dye Reagent in a clean Eppendorf tube. Then add 1 µl Working Reagent to each tube and mix. Incubate for 2 min in the dark. Press left arrow on "<-Std -> ", until the window shows "1.". The Glycerol concentration (µm) will be displayed in the window. Record the data, or press "Save" to save the data for later retrieval. 5 Acetate (Acetic Acid) 1. EOAC to 1 mm.1 mm (.59 mg/dl, 5.9 ppm) Biological Food, drink & environment R 2 = Nominal (Acetic Acid, mm) Important: prior to assay, bring the assay reagents (cat# EOAC- 1) to room temperature. Add 65 µl Developer to Enzyme A and 12 µl Developer Enzyme B tubes. Mix well by pipetting and vortexing. Keep enzyme tubes cold during the assay. 1. Prepare 1 mm Acetate Standard by mix 5 µl provided Standard with 995 µl H 2 O. In separate mini-glass tubes, add 1 µl H 2 O ("Blank"), 1 µl 1 mm Standard ("Std"), and 1 µl Sample. per tube: 9 µl Assay Buffer, 6 µl Enzyme A, 1 µl Enzyme B, 1 µl ATP, 1 µl Dye Reagent. Add 9 µl Working Reagent to each tube. Incubate for 3 min at room temperature in the dark. Press left arrow on "<- Std -> ", until the window shows "1.". ELAC to 1 mm.4 mm (1.4 mg/dl, 14 ppm) Biological Food and drink R 2 = Nominal (Lactose, mm) Important: prior to assay, bring the assay reagents (cat# ELAC- 1) to room temperature. Add 12 µl dh 2 O to Enzyme Mix and 12 µl dh 2 O to Lactase tubes. Mix well by pipetting and vortexing. Keep enzyme tubes cold during the assay. 1. Prepare 1 mm Lactose Standard by mix 1 µl provided Standard with 19 µl H 2 O. In separate mini-glass tubes, add 1 µl H 2 O ("Blank"), 1 µl 1 mm Standard ("Std"), and 1 µl Sample. per tube: 9 µl Assay Buffer, 1 µl Enzyme Mix 1 µl Lactase and 1 µl Dye Reagent. Mix well. Add 9 µl Working Reagent to each tube. Incubate for 3 min at room temperature in the dark. Press left arrow on "<- Std -> ", until the window shows "1.".
