NEWCASTLE-BRONCHITIS VACCINE (B1 TYPE, B1 STRAIN; MASS. & CONN. TYPES) Merial Select Newcastle-Bronchitis Vaccine, B1 Type, B1 Strain, Mass. & Conn. Types, Live Virus U.S. Vet. Lic. No.: 279 Active Ingredient(s): These vaccines contain the B strain of the Newcastle disease virus and selected strains of infectious bronchitis virus of the Massachusetts and Connecticut types. The virus has been propagated in fertile eggs from specific pathogen free flocks. The immunizing capability of each vaccine has been proven by a master seed immunogenicity test. These vaccines offer proven immunity with mild reactions. Penicillin and streptomycin sulfate are added as bacteriostatic agents. Contains fungizone as a fungistatic agent. Notice: These vaccines have met the requirements of the USDA in regards to safety, purity, potency and the capability to immunize normally susceptible chickens. These vaccines have been tested by the master seed immunogenicity test for efficacy. Indications: These vaccines are recommended for administration to healthy chickens as an aid in the prevention of Newcastle disease and infectious bronchitis, Massachusetts and Connecticut serotypes. The frozen vaccine is recommended for use in healthy one day old chickens. The freeze-dried vaccine is recommended for the vaccination of healthy chickens at one day of age using the coarse spray method or for the vaccination of healthy chickens 14 days of age or older administered either in the drinking water or by aerosol spray. The use of an aerosol spray is recommended for revaccination only. Dosage and Administration: A. Frozen Vaccine: For healthy one (1) day old chickens. Preparation of the Vaccine for Use: Important: Sterilize the vaccinating equipment by autoclaving for a minimum of 15 minutes at 250 F (121 C) or by boiling in water for at least 20 minutes. Never allow chemical disinfectants to come into contact with the vaccinating equipment. 1. Remove only one (1) ampule of vaccine at a time from the liquid nitrogen container. Thaw and use immediately. Do not hold the ampule towards the face when removing it from a liquid nitrogen container. Never refreeze a vaccine ampule after thawing The contents of the ampule are thawed rapidly by immersing it in water at room temperature (15-25 C). Gently swirl the ampule to disperse the contents. Break the ampule at its neck and quickly proceed as described below. 3. Transfer the contents of the ampule into a sterile 2 ml or 5 ml syringe fitted with an 18 to 20 gauge needle to

2 cool, distilled, de-ionized water (10,000 doses to 700 ml of water). 4. Use the vaccine mixture immediately as described below. Method of Vaccination: 1. Use the coarse spray application method. 2. Attach the spray head to the container and set the nozzle at a coarse spray setting. 3. To each box of 100 chickens, administer the vaccine by spraying inches above the box. 4. Each box of 100 chickens should receive approximately 7 ml of vaccine. The exact amount can be determined by spraying into a calibrated tube or container to arrive at the number of applications needed to deliver 7 ml of vaccine. 5. Whatever the volume of vaccine used, take care to administer 1,000 doses to 1,000 chickens. B. Freeze-dried Vaccine (5,000 doses): The vaccines are recommended for the vaccination of healthy chickens at one (1) day of age using the coarse spray method or for the vaccination of healthy chickens 14 days of age or older administered either in the drinking water or by aerosol spray. The use of an aerosol spray is recommended for revaccination only. Directions for Coarse Spray Vaccination: 1. Be certain that all of the vaccinating equipment is clean before use. Never allow chemical disinfectants to come into contact with the vaccinating equipment. 2. Do not open and mix the vaccine until ready to use. 3. Remove the aluminum seal and rubber stopper from the vial. 4. Aseptically transfer 3-5 ml of cool, distilled water into the vaccine vial and mix gently. 5. Transfer the dissolved vaccine into cool, distilled water; 350 ml for 5,000 doses. Mix thoroughly. 6. Attach the spray head to the container and adjust the nozzle for coarse spray. 7. Administer the vaccine by spraying inches above each tray of chicks. 8. Each tray of 100 chickens should receive approximately 7 ml of vaccine. The exact amount can be determined by spraying into a calibrated tube to arrive at the number of applications necessary to deliver 7 ml of vaccine. 9. Whatever volume of vaccine is used in the spray, take care to administer 5,000 doses to 5,000 birds. 10. Since coarse spray application is of a large droplet size, aerosol-like mist of the vaccine is reduced. However, always wear goggles and a face mask while spraying a live virus vaccine. 11. Any equipment used for the application of a live virus vaccine should not be used for any other purpose. Directions for Drinking Water Vaccination: Do not use the drinking water method for vaccination of chickens younger than two (2) weeks of age or less than satisfactory results may be obtained. 1. Do not open or mix the vaccine until ready to use. 2. Remove all medications, sanitizers and disinfectants from the drinking water 72 hours prior to vaccination. Clean and rinse the waterers thoroughly. 3. Provide sufficient waterers so that all of the birds can drink at one time. 4. Withhold all water from the birds for two (2) to four (4) hours prior to vaccination to stimulate thirst. 5. Add nonfat, dry milk to the water at the rate of 1 oz. per gallon before mixing the vaccine. 6. Remove the aluminum seal and rubber stopper from the vial. 7. Fill the vaccine vial with 3-5 ml of clean, cool water and mix gently. 8. Mix the dissolved vaccine with water as shown below: Age of birds Water per 5,000 doses vaccine 2-4 weeks 12.5 gallons (49.5 L) 4-8 weeks 25 gallons (94.5 L)

3 8 weeks or older 50 gallons (189.5 L) 9. Distribute the vaccine solution among the waterers. Avoid direct sunlight. 10. Do not provide any other drinking water until all of the vaccine solution has been consumed. Directions for Aerosol Spray Vaccination: Do not use for initial vaccination. Use only for revaccination of healthy chickens two (2) weeks of age or older. Use a sprayer delivering an aerosol mist to disperse the rehydrated vaccine quickly and evenly throughout the chicken house. 1. Prior to spraying, reduce the air flow to a minimum. Keep the air flow reduced for 20 minutes following spray vaccination. 2. Remove the aluminum seal from a vial of the vaccine. 3. Remove the rubber stopper and fill the vial with 3-5 ml of cool, distilled water. 4. Pour the rehydrated vaccine into a clean container and add 500 ml of cool, distilled water per 5,000 doses of vaccine. Mix thoroughly. 5. For aerosol spray vaccination, apply at the rate of 500 ml per 5,000 chickens. 6. Place the vaccine solution in the sprayer canister, set the discharge control and walk through the house spraying at the rate of 1,000 chickens per minute. Direct the spray above the heads of the birds. 7. Whatever volume of vaccine solution is used, take care to administer 5,000 doses of vaccine per 5,000 birds. 8. Avoid direct contact with the vaccine solution. Wear goggles and a face mask while spraying a live virus vaccine. 9. Any equipment used for the application of a live virus vaccine should not be used for any other purpose. C. Freeze-dried Vaccine (15,000 doses): For healthy chickens 14 days of age or older. Drinking Water - Pouring Application: For use in houses containing between 10,000 and 15,000 chickens. 2. Remove all medications, sanitizers and disinfectants from the drinking water 72 hours (3 days) prior to vaccination. 3. Provide sufficient waterers so that all of the chickens can drink at one time. Shut off the water supply and allow the chickens to drink the troughs dry. 4. Raise the water troughs above the chicken's heads. Clean and rinse the waters thoroughly. 5. Withhold all of the water from the chickens for a minimum of two (2) hours in warm weather to four (4) hours in cool weather prior to vaccination to stimulate thirst. The water withdrawal times should be reduced if half-house brooding is in process. 6. Calculate to supply the vaccine solution at a rate of three (3) gallons (11 L) per 1,000 chickens in cool weather 10,000 chickens in cool weather x 3 gallons (11 L) = 30 gallons (114 L). 14,500 chickens in warm weather x 4 gallons (15 L) = 58 gallons (220 L). 7. Add one of the following to pouring water for vaccine distribution: a. One (1) ounce (30 ml) of nonfat, dry milk per gallon (4 L) of water, or b. Four (4) ounces (118 ml) of fresh skim milk per gallon (4 L) of water. 8. Prepare the vaccine concentrate as follows: a. Prepare a container of milk/water figuring one (1) ounce (30 ml) of concentrate for one (1) gallon (4 L) of water to be used. b. Add cool milk/water to the vial until the dried vaccine is dissolved. Add the vaccine to the container of milk/water to make the vaccine concentrate. Rinse the vaccine vial to remove all of the vaccine.

4 9. To make the vaccine solution, add one (1) ounce (30 ml) of concentrate for each gallon (4 L) of water needed. 10. Distribute the vaccine solution among the waterers. Avoid direct sunlight. 11. Lower the water troughs and allow the birds to drink freely. Add the remaining vaccine solution to the troughs as the birds drink. 12. Do not provide any additional drinking water until all of the vaccine solution is consumed. Drinking Water Vaccination - Proportioner Application: Several types of medicator/proportioners are commercially available. Set the proportioner to deliver one (1) ounce (30 ml) of vaccine concentrate per one (1) gallon (4 L) of water. 2. Remove all medications, sanitizers and disinfectants from the proportioner 72 hours (3 days) prior to vaccination. 3. Clean all containers, hoses, and waterers prior to vaccination. 4. Withhold all water from the chickens for a minimum of two (2) hours in warm weather to four (4) hours in cool weather prior to vaccination to stimulate thirst. 5. Calculate to supply the vaccine solution at a rate of three (3) gallons (11 L) per 1,000 chickens in cool weather 10,000 chickens in cool weather x 3 gallons (11 L) = 30 gallons (114 L). 14,500 chickens in warm weather x 4 gallons (15 L) = 58 gallons (220 L). 6. Continue until all of the concentrate has been consumed before changing the water supply to direct flow. 10. Do not medicate or use disinfectants for 24 hours after vaccination. Coarse Spray Application: For use in houses containing between 10,000 and 15,000 chickens. Use a sprayer delivering a coarse spray pattern to disperse the rehydrated vaccine quickly and evenly throughout a house of chickens. 1. Prior to spraying, reduce the air flow by raising curtains or stopping fans. Air movement should be limited for 15 minutes following vaccination. 2. Fill the vaccine container with cool, distilled, de-ionized water. 3. Pour the reconstituted vaccine into a clean container and add five (5) ounces (150 ml) of cool water for each 1,000 chickens to be vaccinated. 4. Place the vaccine solution into a spray canister and walk through the house spraying at the rate of 1,000 chickens per minute. Direct spray directly above the heads of the chickens. 5. Avoid direct contact with the vaccine solution. Wear goggles and a face mask while spraying. 6. Do not use vaccinating equipment for any other purposes. D. Freeze-dried Vaccine (25,000 doses): For healthy chickens 14 days of age or older. Drinking Water - Pouring Application: For use in houses containing between 15,000 and 25,000 chickens. 2. Storage Conditions for Frozen Vaccine: Ampules: Store in a liquid nitrogen container. Liquid nitrogen container: Carefully observe all liquid nitrogen precautions, including wearing eye protection and gloves. Store in a cool, well-ventilated area. Check the liquid nitrogen level once a day. Keep the container away from incubator intakes and chicken boxes. Liquid Nitrogen Precautions: The liquid nitrogen containers and vaccines should be handled by properly trained personnel only. These persons should be familiar with the Union Carbide publication Precautions and Safe Practices - Liquid Atmospheric Gases, form #9888. Liquid nitrogen is extremely cold. Accidental contact with the skin or eyes can cause serious frostbite. Protect the eyes with goggles or a face shield. Wear gloves and long sleeves when removing and handling frozen ampules or when adding liquid nitrogen to the container. Storage and handling of liquid nitrogen containers should be in a well-ventilated area. Excessive amounts of nitrogen reduces the concentration of oxygen in the air of an unventilated space and can cause asphyxiation. If drowsiness occurs, get fresh air quickly and ventilate the entire area. If a person becomes groggy or loses consciousness while working with liquid nitrogen, get the person to a well-ventilated area immediately. If breathing has stopped, begin artificial respiration. Call a physician immediately. B. Storage Conditions for Freeze-dried Vaccine: Store the freeze-dried vaccine at F (2-7 C). Do not freeze. Caution(s): A. Frozen Vaccine: Do not vaccinate diseased birds. Vaccinate all of the birds on the premises at one time. Administer a minimum of one dose for each bird. Avoid stress conditions during and following vaccination.

7 Do not place chickens in contaminated facilities. Exposure to disease must be minimized as much as possible. For veterinary use only. The capability of the vaccine to produce satisfactory results depends upon many factors, including, but not limited to, conditions of storage and handling by the user, administration of the vaccine, health and the responsiveness of individual animals and the degree of field exposure. Therefore, directions for use should be followed carefully. The use of the vaccine is subject to applicable state and federal laws and regulations. Use only in localities where permitted. Newcastle disease virus can cause inflammation of the eyelids of humans. Care should be taken to avoid contamination of the eyes, hands and clothing with the vaccine. Administer only as recommended. Use the entire contents when first opened. Burn the container and all unused contents. B. Freeze-dried Vaccine: Do not vaccinate diseased birds. Birds should be free of respiratory disease and parasite infestations. Vaccinate all of the birds on the premises at one time. Administer a minimum of one dose for each bird (ie. 5,000 doses to 5,000 chickens). Avoid stress conditions during and following vaccination. Do not place chickens in contaminated facilities. Exposure to disease must be minimized as much as possible. Do not start the vaccination program for replacement pullets after birds are 16 weeks old. For veterinary use only.
