Pharmacy College of Ningxia Medical University Yinchuan Functional Experiment Center of Ningxia Medical University Yinchuan Abstract Objective To determine whether oxymatrine and matrine can against antigen - challenged vasoconstriction of isolated thoracic aorta in guinea pig if so to investigate whether the protective mechanism are related to the activation of the transient receptor potential vanilloid type 1 TRPV1. Methods The thoracic aorta rings of guinea pigs were used to study the inhibitory effect of oxymatrine and matrine on vasoconstriction induced by bovine serum albumin. Results The intense vasoconstrictor response induced by bovine serum albumin was significantly attenuated by diphenhydramine the H1 receptor antagonist. Oxymatrine mol L - 1 ~ mol L - 1 significantly decreased the antigen - challenged vasoconstriction on isolated aortic rings of guinea pigs and this action was abolished by capsazepine 10-5 mol L - 1 a selective TRPV1 antagonist. Matrine mol L mol L - 1 could also significantly suppress the vasoconstriction but its effect was not modified by capsazepine. Conclusion The protective effect of oxymatrine on vasoconstriction is induced by anaphylaxis via activation of TRPV1 whereas the vasodilative effect of matrine is not. Methods 5mm 4mm 2mm and 10mm 4mm 2mm fragments was prepared with fresh human fetal ovarian cortex from an abortion of 20 weeks and cryopreserved after two - step osmotic equilibrium added 10μg ml - 1 FSH. After thawing the ovary fragments were cultured in the medium added 10μg ml - 1 FSH for 4. 5 hours. The follicle diameter and follicle constituent ratio of thawed cultured ovary fragments were detected and compared with the fresh ovary fragments. Results 1The constituent ratio of normal follicles and follicular diameter were similar to that in vitrification and thawing groups as compared with fresh control group. 2The follicular diameter of cultivation in thawing groups were longer than that in thawing and culture groups P < Conclusion Adding FSH during vitrifiation and thawing process did not influence constituent ratio of follicles but some primordial follicles could develop to primary follicle after adding FSH in vitro and culturing for 4. 5 hours.
