Received: August, 1990 Accepted: 17 January, 1991 ABSTRACT: Serial extracts of the seeds of M.fragrans were investigated for their antidiarrhoeal potential in three experimental models. Though all the three extracts exhibited significant activity, the petroleum ether and ethanol extracts were found to be more effective above mg kg dose levels. Myristica frgrans Houtt. (Jayphal) is indigenously seen and its seeds or nutmegs are considered as a folk remedy for diarrhoea. It is also aromatic, stimulant, carminative and in large doses, narcotic. It contains a volatile oil (.%) which is stimulant and carminative 1. Nutmeg powder has been used to control diarrhoea in patients suffering from thyroid carcinoma where the prostaglandin (PG) E and F levels are high. Earlier attempts were also made to control PG-mediated diarrhoea with indomethacin, aspirin and nutmeg. It has also been reported to be of therapeutic value in diarrhoea associated with Crohn s disease. This effect is due to a fall in PGE1 levels. Experimental studies in albino rats using the pet. Ether and aqueous extracts of nutmeg showed a weak antidiarrhoeal activity. In view of these reports it was thought worthwhile to screen the serial extracts, pet. Ether, chloroform and ethanol for their antidiarrhoeal effects in experimental models. Adult male Holtzman rats weighing 0 to g, maintained at room temperature at the College animal house and fed on Hindlever feed, were used for the study. Animals were housed separately and had free access to water. More than animals in each group were used for each dose level. Good quality nutmegs obtained locally were powdered and successively extracted with pet. Ether (0 0 o ), chloroform and ethanol as per conventional methods. The extracts were administered orally, as solution in % propylene glycol. Antidiarrhoeal activity was assessed using the following three experimental models: (a) Measurement of faecal output in albino rats: Method of Bass et al 5 was followed and drug extracts were given in, and mg kg dose levels. Percentage of faecal output in treated rats and the percentage reduction in output was calculated.

(b) Castor oil diarrhoea in rats: In overnight-fasted male rats diarrhoea was induced by oral administration of castor oil (1 ml rat). Test extracts were given orally one hour prior to castor oil. Percentage of animals not showing diarrhoeal droppings at the end of th hour after castor oil was noted and compared. Pet. Ether extract in, and mg kg dose levels, chloroform etc. in mg kg and ethanol extract in and mg kg were used. (c) Enteropooling assay in rats: Drug extracts were given in overnight fasted rats and one hour later they all received the diarrhoeal agent Magnesium sulphate (% soln.) ml rats orally. All of them were killed 0 minutes later and entire small intestine collected, weighed and compared 7. Pet. Ether and ethanol extracts in and mg kg and chloroform extract in mg kg were used for the study. In all the three test patterns loperamide hydrochloride ( mg kg) was used as the standard drug for comparison. Statistical evaluation was carried out using the student s t test. Ethanol extracts as mg kg dose level reduced the faecal output by 7.% compared to 90.% of loperamide (Table I). In castor oil induced diarrhoea, the pet. Ether ( mg kg) and ethanol ( mg kg) extracts exhibited significant suppression of diarrhoea in rats (90%) compared to the % in loperamide treated group (Table 1). In enteropooling assay in rats, only the pet. Ether ( mg kg) and chloroform ( mg kg) extracts significantly reduced (P < 0.05) the weight of intestine (Table ). These observations were in support of the earlier findings of Shidore et al using the aqueous and pet. Ether extracts in castor oil diarrhoea in rats. Our earlier studies using the decoction or aqueous extract of the seeds also showed significant antidiarrhoeal effect in all the three test patterns. The castor oil diarrhoea model has been suggested for evaluation of PG biosynthesis inhibition. The propulsive activity of intestine (in vivo) is activated by the castor oil, which may well be associated with increased PG mediated diarrhoea is also well documented by various workers. In the present observations, pet. ether and ethanol extracts have shown significant suppression of castor oil induced diarrhoea, which may be due to the inhibition of PG biosynthesis. Similar effects were also seen when magnesium sulphate induced enteropooling in intestine was inhibited partially by the extracts. However, these experimental findings are in support of the usefulness of nutmeg as an antidiarrhoeal agent in folk medicine.
