Humoral components The complement system is the major humoral non-specific defense mechanism. Once activated complement can lead to increased vascular permeability, recruitment of phagocytic cells, and lysis and opsonization of bacteria. Cytokines : In response to microbes, dendritic cells, macrophages, and other cells secrete cytokines that mediate many of the cellular reactions of innate immunity. cytokines are soluble proteins that mediate inflammatory reactions and are responsible for communications between leukocytes and between leukocytes and other cells (TNFα, and IL-1) Dendritic cells and macrophages also produce IL-12 in response to LPS and other microbial molecules. C-reactive protein (CRP) binds to phosphorylcholine on microbes and coats the microbes for phagocytosis by macrophages, which express a receptor for CRP. Plasma mannose-binding lectin (MBL) is a protein that recognizes microbial carbohydrates and can coat microbes for phagocytosis or activate the complement cascade by the lectin pathway.

Cellular locations of receptors of the innate immune system. Some receptors, such as Toll-like receptors (TLRs), are located on cell surfaces; other TLRs are in endosomes (they may be resident in the endoplasmic reticulum and may be rapidly translocated to endosomes in response to microbe entry); and some receptors for viral RNA and for bacterial peptides are in the cytoplasm.

phagocytosis As the microbe is being bound by the phagocyte s receptors and ingested, the receptors deliver signals that activate several enzymes in the phagolysosomes. One of these enzymes, called phagocyte oxidase, converts molecular oxygen into superoxide anion and free radicals. These substances are called reactive oxygen species (ROS), and they are toxic to the ingested microbes. (oxygen dependent pathway). A second enzyme, called inducible nitric oxide synthase, catalyzes the conversion of arginin to nitric oxide (NO), also a microbicidal substance (Nitric oxide dependent ).
