EFFICACY TESTS (EN 1276) VIRUSOLVE + AMITY UK LTD HOSPITAL INFECTION RESEARCH LABORATORY CITY HOSPITAL NHS TRUST DUDLEY ROAD BIRMINGHAM B18 7QH NOVEMBER 2002

MANUFACTURER Amity UK Ltd Friendship House Dodworth Business Park Dodworth Barnsley S75 3SP TEST PRODUCT Virusolve + Constituents unknown. Diluted one part product plus 9 parts sterile hard water for testing LOT NUMBER EXPIRY DATE Not stated Not stated STORAGE CONDITIONS Room temperature TEST ORGANISMS Staphylococcus aureus NCTC 10788 Pseudomonas aeruginosa NCTC 6749 Escherichia coli NCTC 10418 Enterococcus hirae NCTC 12367 Methicillin Resistant Staphylococcus aureus NCTC 12493 TEST METHOD AND VALIDATION EN 1276 Chemical disinfectants and antiseptics - Quantitative suspension test for the evaluation of bactericidal activity of chemical disinfectants and antiseptics used in food, industrial, domestic and institutional areas (Phase 2, step 1). Tests for disinfectants for medical establishments not yet ratified. Product diluent used during the test sterile hard water 300mg/kg CaCO 3 Product test concentration 1 part product plus 9 parts water 2 of 5

Appearance product dilution Contact times Test temperature Interfering substance Inhibition method Neutralizer Clear solution 1, 2 and 5 minutes 20 o C Bovine albumin 0.03% clean conditions 0.3% dirty conditions Dilution/neutralization Tween 80 30g/l, lecithin 3g/l, sodium di-octyl sulphosuccinate 20ml/l Tests were performed to establish the suitability of this neutralizer in neutralizing the activity of the disinfectant without being inhibitory to the test organisms. It was demonstrated to be suitable for the gram negative test organisms ie Ps. aeruginosa and Esch. coli but using the EN 1276 method for culture preparation was unsuitable for the gram positive test bacteria. Therefore, a higher starting count was used ie a combination of dilution and neutralizer. Initial tests were carried out with Tween 80 30g/l, lecithin 3g/l and histidine 1g/l but this proved unsatisfactory as a neutralizer. SUMMARY OF TEST METHOD EN 1276 Chemical disinfectants and antiseptics - Quantitative suspension test for the evaluation of bactericidal activity of chemical disinfectants and antiseptics used in food, industrial, domestic and institutional areas (Phase 2, step 1). Tests for disinfectants for medical establishments not yet ratified. Copies available from BSI, 389 Chiswick High Road, London W4 4AL. The test method involves mixing 1 ml of the test bacteria with 1 ml of soil (0.3 or 3% albumin and then adding 8 ml of disinfectant at in use concentration. After the required contact time, 1 ml is removed to 9 ml of recovery/neutralizer which is then plated to detect surviving test bacteria. 3 of 5

RESULTS BACTERICIDAL ACTIVITY OF VIRUSOLVE + USING PHASE 2 STEP 1 SUSPENSION TEST EN 1276 Log 10 counts/reductions achieved in 1, 2 and 5 minutes* (Tests carried out in duplicate) Log 10 reduction Test organism Log10 initial count (challenge) Clean conditions (0.03% albumin) Dirty conditions (0.3% albumin) 1 min 2 mins 5 mins 1 min 2 mins 5 mins Ps. aeruginosa 7.30 >6.30 >6.30 >6.30 >6.30 >6.30 >6.30 Esch. coli 7.00 >6.00 >6.00 >6.00 >6.00 >6.00 >6.00 Ent. hirae** 9.60 >6.60 >6.60 >6.60 >6.60 >6.60 >6.60 Staph. aureus** 9.20 >6.20 >6.20 >6.20 >6.20 >6.20 >6.20 Methicillin resistant Staph. aureus** 9.04 >6.04 >6.04 >6.04 >6.04 >6.04 >6.04 * Contact time of 5 min are stated in EN 1276. However, 1, 2 and 5 min contact times were commissioned by Amity UK Ltd as more relevant for hard surface disinfectant for use in health care premises. The tests carried out were therefore, more stringent. ** A higher starting count was used for the gram positive test bacteria in order to demonstrate a >5 log 10 reduction A >6.0 log 10 reduction is the limit of detection for this test. 4 of 5

CONCLUSION When tested in accordance with EN 1276 (1997), Viruslove + tested at 10% dilution possesses bactericidal activity at 20 o C. A >5 log 10 (99.999%) reduction was achieved with all test organisms i.e. Ps. aeruginosa, Staph. aureus Esch. coli and Ent. hirae in 1 min, 2 mins and 5 mins under clean (0.03% albumin) and dirty (0.3% albumin) conditions. To satisfy the requirements of the test, at least a 5 log 10 reduction in specified test organisms is required within 5 minutes when the disinfectant is tested at its intended use dilution(s). Performance under light (clean) and moderate to heavy (dirty) soiling was assessed.... LR Simpson Research Scientist... CR Bradley Laboratory Manager...
