The CS Solution contains a mixture of Azur A and Methylene Blue specially formulated to provide optimized staining of cells and structures grown on Basement Matrix Extract (BME). Ideal staining contrast is achieved with minimum background since excess stain can be easily washed away using the wash buffer. The blue coloration allows optimal contrast between cells and the growth matrix. Storage Conditions: Store at room temperature. Applications: Staining of cells grown on Cultrex Basement Membrane Extract (BME) or other extracellular membrane protein extracts after application such as: Tube Assay: Basement Membrane Extract promotes differentiation of a mouse endothelial cell line derived from axillary lymph node (SVEC4-10), into capillary-like structures. Neurite outgrowth assay: Basement Membrane Extract promotes differentiation of neurite from chick dorsal root ganglion. Angiogenesis assay such as the Ring Assay: Basement Membrane Extract promotes differentiation of mouse aorta tissue to form capillary-like structures. Staining Procedure: Note: This procedure requires methanol and distilled or deionized water to dilute the 10X CS Wash Solution) 1- Remove culture media from well. 2- Add enough methanol to cover sample and incubate 1 minute at room temperature. 3- Remove methanol and wash 3 times with equal volumes of 1X CS Wash Solution. 4- Add enough CS Solution to cover samples in culture wells. 5- Leave stain on samples from 10 seconds to 1 minute. 6- Remove excess dye with pipette. 7- Wash samples 3 times with 1X CS Wash Solution or until desired destaining is achieved. 8- Discard excess and observe under microscope. 9- Samples can be stored for 24 hours at 4 C covered with 1X CS Wash Buffer. 2010 Trevigen, Inc.
