Product specific information Description: The 11B11 antibody reacts with mouse interleukin-4 (IL-4), a 14 kd cytokine. The 11B11 antibody is a neutralizing antibody. Mouse IL-4, also known as BCDF, BCGF-1 and BSF-1, is secreted by activated Th2 and natural killer T cells, and to a lesser extent by Th1 and mast cells. IL-4 is speciesspecific and promotes proliferation and differentiation of B cells. IL-4, in conjunction with other signals such as CD40, induces Ig isotype switching of B cells and the upregulation of surface MHC class II and CD23. Applications Reported: This 11B11 antibody has been reported for use in cytokine neutralization, ELISPOT, and ELISA cap. Applications Tested: The Functional Grade Purified 11B11 antibody has been tested by LAL assay to verify low endotoxin levels, by ELISA, and in bioassay for neutralization of mouse IL-4 bioactivity. The 11B11 antibody at 0.125 µg/ml has been found to inhibit by 50% the biological effects of 1.0 ng/ml mouse IL-4 in a CTLL-2 cell proliferation assay. Please refer to Best Protocols for more information. The 11B11 antibody has been tested as the capture antibody in a sandwich ELISA for analysis of mouse interleukin-4 (IL-4) in combination with the biotinylated BVD6-24G2 (cat. 13-7042) antibody for detection and recombinant mouse IL-4 (cat. 14-8041) as the standard. A suitable range of concentrations of this antibody for ELISA capture is 1-4 µg/ml. A standard curve consisting of doubling dilutions of the recombinant standard over the range of 500 pg/ml - 4 pg/ml should be included in each ELISA plate. Endotoxin: Less than 0.001 ng/ug antibody as determined by the LAL assay. Storage and handling: Use in a sterile environment. Filtration: 0.2 µm post-manufacturing filtered. Background/Target Information The protein encoded by this gene is a pleiotropic cytokine produced by activated T cells. This cytokine is a ligand for interleukin 4 receptor. The interleukin 4 receptor also binds to IL13, which may contribute to many overlapping functions of this cytokine and IL13. STAT6, a signal transducer and activator of transcription, has been shown to play a central role in mediating the immune regulatory signal of this cytokine. This gene, IL3, IL5, IL13, and CSF2 form a cytokine gene cluster on chromosome 5q, with this gene particularly close to IL13. This gene, IL13 and IL5 are found to be regulated coordinately by several long-range regulatory elements in an over 120 kilobase range on the chromosome. Two alternatively spliced transcript variants of this gene encoding distinct isoforms have been reported.
