You may answer the questions either in Bahasa Malaysia or in English. [Arahan: Jawab LIMA soalan. Answer ALL the following questions - 2-1. Answer all parts of this question pertaining to Kjeldahl method. Why is the Kjeldahl method only sufficient for estimation of protein content? Describe the chemical reactions and components involved in each step? (12 marks) (c) What does the 14 in the equation (Eq.1) represent? (d) Why is 6.25 normally used as a conversion factor for the estimation of proteins in food products? Why are different conversion factors used in protein analysis? 2. Answer all parts of this question a. Briefly describe the following terms: (4 marks) (i) Saponification (ii) Simple lipids (iii) Complex lipids (6 marks) b. Briefly discuss the steps of enzymic method for quantification of total cholesterol content in a sample. c. How does vitamin E prevent the production of peroxide inside the body? (4 marks) d. State the active forms of vitamins A, E, K and D. 3/-

- 3-3. Answer all parts of this question Briefly describe determinate and indeterminate errors. Mention the scientific factors that cause these errors. Potassium acid phthalate is a primary standard for titratable acidity. Briefly discuss this statement. (c) Explain how a sharp end point is obtained for organic acid titration. (d) In the determination of trace copper in a biological sample, five replicates were obtained and analyzed. The mean obtained was 10.8 ppm with a standard deviation of 0.7 ppm. The true value was 11.7. Calculate the test (t) value. 4. You are given a sample (frog). According to the hypothesis, the frog s skin is believed to possess proteins that have pharmaceutical properties. Prepare a schematic plan for the sample preparation of protein analysis. (5 marks) Provide the four major steps to achieve purity of sample. Elaborate on the considerations in each step. (15 marks) 5. Answer all parts of this question. What is a quality control system? Explain the terms warning limit and action limit of Levey-Jenning control chart. Describe the following terms (i) Detection limit (ii) Precision (iii) Accuracy (6 marks) 4/-

- 4 - (c) A 1.5 ppm standard coloured solution of tryptophan gives an absorbance reading of 0.072 with a spectrophotometric method, whereas a series of sequential baseline absorbance readings for blank are 0.004, 0.001, 0.005, 0.007 and 0.003. Calculate the detection limit of the experiment. (6 marks) 5/-

Bincangkan tindakbalas kimia dan komponen-komponen yang terlibat dalam setiap langkah. (12 markah) (c) Apakah 14 dalam persamaan (Eq.1) mewakili? (e) Mengapakah 6.25 biasanya digunakan sebagai faktor penukaran untuk penentuan kandungan protein dalam produk makanan? Mengapakah faktor penukaran yang berbeza-beza digunakan dalam analisis protein? Jelaskan secara ringkas tentang istilah-istilah berikut: (i) Penyabunan (ii) lipid ringkas (iii) lipid kompleks (6 markah) Bincangkan secara ringkas tentang langkah-langkah kaedah enzimatik dalam kuantifikasi kandungan kolesterol total dalam sesuatu sampel. (c) Bagaimanakah vitamin E dapat mengelakkan penghasilan peroksida di dalam badan? (4 markah) (d) Nyatakan bentuk-bentuk aktif vitamin A, E, K dan D. 6/-

- 6-3. Jawab semua bahagian soalan berikut. Jelaskan secara ringkas tentang istilah-istilah ralat tentuan dan ralat tak tentu. Nyatakan faktor-faktor saintifik yang menyebabkan ralat-ralat ini. Kalium asid ftalat ialah piawai primer untuk keasidan tertitratkan. Bincang secara ringkas tentang kenyataan ini. (c) Jelaskan bagaimana titik akhir yang tajam dapat diperolehi bagi penitratan asid organik. (d) Dalam penentuan kandungan kuprum dalam sampel biologi, lima replikat diperolehi dan dianalisa. Nilai min yang diperolehi ialah 10.8 ppm dan sisihan piawai ialah 0.7 ppm. Nilai benar ialah 11.7. Kirakan nilai ujian (t). 4. Anda diberikan satu sampel (katak). Berdasarkan hipotesis, kulit katak ini dipercayai mengandungi protein yang mempunyai sifat-sifat farmaseutikal. Sediakan satu rancangan berskema untuk penyediaan sampel dalam analisis protein. (5 markah) Berikan empat langkah utama untuk mencapai ketulenan sampel. Huraikan pertimbangan-pertimbangan dalam setiap langkah. (15 markah) 5. Jawab semua bahagian soalan berikut. Apakah sistem kawalan kualiti? Jelaskan istilah batas amaran dan batas tindakan dalam carta kawalan Levery-Jenning. Gambarkan istilah-istilah berikut: i. Batas pengesanan ii. Kepresisan iii.
