TOXICOLOGICAL INFORMATION Unless otherwise noted the following pertains to the active pharmaceutical ingredient. Oral Toxicity: LD 50 = 5,000 mg/kg (oral, rat) Acute Effects: Serious acute hypersensitivity reactions to cephalosporins and penicillin may require the administration of epinephrine and other emergency measures. Pseudomembranous colitis has been reported with nearly all antibacterial agents, including cephalexin, and may range from mild to life threatening. Therefore, it is important to consider this diagnosis in patients with diarrhea subsequent to the administration of antibacterial agents. Effects of Repeated Doses: Prolonged use of cephalexin may result in the overgrowth of nonsusceptible organisms. Careful observation of the patient is essential. If superinfection occurs during therapy, appropriate measures should be taken. Other effects include salivation and vomiting, nausea and diarrhea. Skin Irritation: Possible mild skin irritant. Reproductive and Developmental Effects: FDA Pregnancy Category B. (Animal studies have failed to show a risk to the fetus but there are no adequate studies in pregnant women: or animal studies have shown an adverse effect but human studies have not shown a risk to the fetus in the first trimester and there is no evidence of risk in later trimesters.) The safety of cephalexin during pregnancy in humans has not been determined. The daily oral administration of cephalexin to rats in doses of 250 or 500 mg/kg prior to and during pregnancy, or to rats and mice during the period of organogenesis only, had no adverse effect on fertility, fetal viability, fetal weight or litter size. Sensitization: The literature supports the conclusion that cephalexin is a potent sensitizer capable of inducing allergic reactions in those that are not known to be allergic to cephalosporins and penicillins as well as those that are. Severe allergic reactions have occurred in those predisposed by allergies of any kind. Hypersensitivity can develop through contact with dusts and liquids containing by all exposure routes.
