You may answer the questions either in Bahasa Malaysia or in English. [Arahan: Jawab LIMA soalan. - 2 - Section A (This question is related to Lab and compulsory to answer). 1. Answer both parts of this question Describe detailed steps involve in sugar analysis and state the precautions required. (15 marks) What is the conversion factor used for calculating reducing sugar? How is this factor obtained? (5 marks) Section B 2. Answer all parts of this question Discuss the Karl Fischer method for the determination of moisture content in food products. You have been using the peroxide value as a measure of off-flavors due to rancidity in specialty whole-wheat flour samples. But then you begin to receive a large number of off-flavor complaints for a specific sample lot. However, upon checking, the results showed low peroxide values were measured. (i) What could have happened? (ii) How would you modify your current quality control system for detecting offflavors due to rancidity? 3. Briefly explain the differences between the following: Wet ashing vs. Dry ashing (6 marks) Karl Fisher titration vs. Distillation method for moisture determination Kjedahl method vs Lowry method for quantifying protein content (6 marks) (8 marks) 3/-

- 3-4. Your analysis of an oil sample gives the following results. What do each of these results tell you about the characteristics of the sample? Briefly describe the principles for each method used. Large saponification number Low iodine value High TBA (thiobarbituric acid) number High free fatty acid content (20 marks) 5. Answer both parts of this question AOAC gravimetric procedure should be use to measure the fibre content of products that contain resistant starch. Why is the resistant starch measured in this procedure and not in the Englyst-Cummings procedure? Give an example of a food product suitable for each of these procedures. What are the differences between Soxhlet and Goldfish fat extraction? What type of samples would each of these systems be used for? 6. Briefly explain one method that could be used for each of the following: To prevent hydrolysis of polysaccharides when free sugars are extracted from fruits with an alcohol extraction To remove pigments from a sugar-containing solution To remove proteins from solution for starch analysis To measure total carbohydrate (20 marks) 7. Answer all parts of this question Munson and Walker, Lane-Eynon, and Nelson-Somogyi methods are used for the determination of total reducing sugars. Make a comparison of these methods based on the principles and steps involved. (15 marks) Explain how physico-chemical changes in a sample can influence the analysis results. (5 marks) 4/-

Jawab kedua-dua bahagian soalan ini Terangkan langkah-langkah utama dalam analisis gula dan nyatakan langkah berjaga-jaga yang diperlukan. (15 markah) Apakah faktor pertukaran yang digunakan untuk pengiraan gula penurun? Bagaimanakah faktor ini diperoleh. (5 markah) Bahagian B 2. Jawab semua bahagian soalan ini Bincang kaedah Karl Fischer bagi penentuan kandungan lembapan dalam produk makanan. Anda telah menggunakan nilai peroksida bagi off-flavour disebabkan ketengikan dalam sampel tepung gandum. Tetapi anda menerima beberapa aduan terdapatnya off-flavour dalam suatu lot sampel. Walaubagaimana pun, apabila diperiksa keputusan menunjukkan nilai peroksida yang rendah (i) Apakah yang mungkin telah berlaku? (ii) Bagaimanakah anda boleh ubahsuai sistem kawalan mutu bagi mengesan off-flavour? 3. Berikan perbezaan antara kenyataan berikut. Jawab kedua-dua bahagian soalan ini. Kaedah gravimetrik AOAC patut diguna untuk mengukur gentian dalam produk yang mengandungi kanji rintang. Kenapa kanji rintang diukur dalam kaedah ini dan bukan dalam kaedah Englyst-Cummings? Berikan satu contoh produk makanan yang sesuai bagi setiap kaedah tersebut. Apakah perbezaan antara pengekstrakan lemak Soxhlet dan Goldfish? Apakah jenis sampel yang sesuai untuk diuji dengan sistem berikut? 6. Terangkan dengan ringkas satu kaedah yang boleh digunakan bagi setiap yang berikut: Untuk menghalang hidrolisis polisakarida apabila gula bebas diekstrak daripada buah dengan pengekstrakan alkohol Untuk menyingkirkan pigmen dalam larutan mengandungi gula Untuk menyingkir protein daripada larutan untuk analisis kanji Untuk mengukur karbohidrat total (20 markah) 7.
