Training Course Program Level I - Ventricular Pressure-Volume Basics This session will introduce attendees to the background physiology, equipment functionality, and experimental design considerations required to fully optimize Ventricular Pressure-Volume systems in a laboratory setting. Learning Objectives The purpose of this session is to familiarize attendees with: Comparative Cardiac Physiology A review of basic cardiovascular physiology Pressure-Volume Loops Pressure-Volume Loops are introduced, focusing on important landmarks of the cardiac cycle Disease States Systolic Dysfunction, Diastolic Dysfunction and Valve Diseases will be discussed along with the corresponding changes in PV Loop morphology Heart Mechanics Important Pressure-Volume parameters are introduced with their application to cardiac function Pressure-Volume System Technology and Theory Introduction to conductance catheter theory and MPVS system operation Calibration and Data Analysis Requirements for proper cuvette calibration of PV data is discussed, along with parallel conductance calibration theory Experimental Design Proper experimental protocols and procedures are introduced Level II - Ventricular PV Experimental Techniques This session will provide comprehensive training for Pressure-Volume experimental techniques. Attendees will receive expert demonstration, as well as and hands-on experience operating Pressure-Volume hardware and performing surgical procedures. Blood Volume Cuvette Calibration Technique for catheter calibration and deriving blood resistivity Saline Calibration Proper technique for assessment of parallel conductance of the myocardium Real-Time Assessment Using LabChart s PV Loop module and other features to acquire relevant data in real time Level III - Leveraging Pressure-Volume Data The final session will take attendees from the acquisition of Pressure-Volume data to producing quality data, ready for publication using the PV Loop module in LabChart 7. Learning Objectives The purpose of this session is to familiarize attendees with: Applying Calibration Techniques Utilizing recorded blood cuvette data to calibrate conductance to volume units Saline Bolus Data Analysis Using the PV Loop module to adjust recorded volume data and derive parallel conductance of the myocardium PV Catheter Maintenance Best practices for prolonging the life of your MPVS systems and Pressure-Volume catheters PV Loop Data Analysis Leveraging recorded data using the PV Loop module and producing quality results Defining & Reporting Key Hemodynamic Parameters Exporting images and relevant cardiovascular hemodynamics for publication Advanced LabChart Techniques Harnessing the power of LabChart 7 to make recording and analyzing data as simple as possible

Training Course Instructors: Dr. Craig Plato CEO & Founder, Plato BioPharma, Inc. Dr. Plato has over 10 years experience in the pharmaceutical industry and served as Head of In Vivo Pharmacology at both Myogen and Gilead Colorado from 2004-2010. He established Plato BioPharma, Inc. in 2010 and has served small and large Biotech and Pharma in drug discovery. While directing the cardiovascular group at Myogen, Craig s team led several drug discovery pharmacology projects including Histone Deacetylase Inhibitors, Protein Kinase D Inhibitors, and a-myosin Hevay Chain activators for the Myogen-Novartis collaboration focusing on heart failure and pathological cardiac hypertrophy. Dr. Plato provided the first direct evidence and patented the use of HDAC inhibitors as combination therapeutics for the treatment of cardiac dysfunction. At Gilead Sciences, Dr. Plato led CKD drug discovery efforts as well as participated in development activities of the Endothelin Receptor Antagonist program and managed the team responsible for generating preclinical data requested by the FDA for approval of Ambrisentan Orphan Drug status for Idiopathic Pulmonary Fibrosis in 2009. Prior to these efforts, Craig was a Senior Scientist responsible for leading cardiorenal drug discovery teams at Abbott Laboratories (2001 2004) in the Chicago area where he focused on diabetic nephrology, electrophysiologic safety, and platelet and coagulation activation. Dr. Plato received his Ph.D. Degree in Physiology from the Medical College of Wisconsin (1996), and completed his Post-Doctoral training in Hypertension and Vascular Research at Henry Ford Hospital. During Craig s academic career, he was twice awarded the American Physiological Society (APS) Excellence in Renal Research Award and the American Heart Association Merck New Investigator Award. During Craig s industry career he has served on the APS Renal Section Steering Committee (2006 2009) as well as sat on, then Chaired, the APS Physiologists in Industry Committee. Jeff Harris Global Training & Support Manager, ADInstruments As a member of ADInstruments Corporate Training & Support Team, Jeff has extensive experience providing hands-on training and technical support to a large customer base of cardiovascular researchers focused on in vivo pressure-volume measurements in a wide range of animal models to include mice, rats, rabbits, dogs, pigs, sheep, and horses. Jeff s strong background in biomedical engineering and heart mechanics proved invaluable during the recent design and development of ADInstruments new PV Loop Analysis Module. Jeff is constantly designing, revising, and implementing newly structured training programs to help researchers master successful Pressure Volume hardware/software integration in their laboratory. Jeff received his B.S. Degree in Biomedical Engineering, with minors in Mathematics and Pre-Med, from Saint Louis University (2003.)

Brandon Bucher - Research Product and Support Manager, ADInstruments As Research Product Manager at ADInstruments, North America, Brandon has developed and delivered hands-on training and technical support to a wide range of cardiovascular physiologists, including hardware and application implementation and LabChart Software Training. Brandon s background in biomedical engineering and previous work designing specialized laboratory instrumentation delivers an exceptional understanding of the underlying theory, mechanics and operation of Ventricular Pressure-Volume Systems, with a strong grasp of the necessary data collection and analysis tools that are key to the experiment. Brandon received his B.S. in Bioengineering from Washington State University (2006). Kristin Joly Plato BioPharma, Inc. Kristin Joly has cardiovascular drug discovery experience at Myogen, Gilead Sciences and since its inception, at PBI. Kristin has been performing direct systemic and cardiac hemodynamic measurements since 2006. As part of that experience, Kristin contributed extensively to systolic and diastolic heart failure model development, validation and compound efficacy testing for the Myogen-Novartis heart Failure collaboration. Later, Kristin s expertise was expanded to developing and applying the Millar catheter and ADInstruments PowerLab technology to direct evaluation of pulmonary hemodynamics in the setting of Pulmonary Arterial Hypertension and Idiopathic Pulmonary Fibrosis. Kristin has contributed to several published efforts utilizing the Millar-ADInstruments platform to evaluate cardiac performance, and recently to a peer-reviewed paper (Liles, JT, KK Ida, KM Joly, J Chapo and CF Plato. Age exacerbates chronic catecholamine-induced impairments in contractile reserve in the rat. Am J Physiol Regul Integr Comp Physiol August 2011 301: R491-R499) evaluating cardiac contractile reserve in the setting of disease with mechanistically distinct inotropic agents. Since 2006, Kristin has performed over 2,000 P-V procedures as well as well over 1000 pulmonary arterial catheterizations. This expertise has been leveraged to both train PBI staff as well as serving as a technical expert at the ADInstruments-Millar Rodent PV workshops since 2010.
