METHOD 4010 SCREENING FOR PENTACHLOROPHENOL BY IMMUNOASSAY 1.0 SCOPE AND APPLICATION 1.1 Method 4010 is procedure for screening solids such s soils, sludges, nd queous medi such s wste wter nd lechtes for pentchlorophenol (PCP) (CAS Registry 87-86-5). 1.2 Method 4010 is recommended for screening smples to determine whether PCP is likely to be present t concentrtions bove 0.5 mg/kg for solids or 0.005 mg/l for queous smples. Method 4010 provides n estimte for the concentrtion of PCP by comprison with stndrd. 1.3 Using the test kits from which this method ws developed, 95 % of queous smples contining 2 ppb or less of PCPs will produce negtive result in the 5 ppb test configurtion. Also, 95 % of soil smples contining 125 ppb or less of PCBs will produce negtive result in the 500 ppb test configurtion. 1.4 In cses where the exct concentrtion of PCP is required, dditionl techniques (i.e., gs chromtogrphy (Method 8040) or gs chromtogrphy/mss spectrometry (Method 8270)) should be used. 2.0 SUMMARY OF METHOD 2.1 Test kits re commercilly vilble for this method. The mnufcturer's directions should be followed. In generl, the method is performed using wter smple or n extrct of soil smple. Smple nd n enzyme conjugte regent re dded to immobilized ntibody. The enzyme conjugte "competes" with PCP present in the smple for binding to immobilized nti-pcp ntibody. The test is interpreted by compring the response produced by testing smple to the response produced by testing stndrd(s) simultneously. 3.0 INTERFERENCES 3.1 Compounds tht re chemiclly similr my cuse positive test (flse positive) for PCP. The test kit used in preprtion of this method ws evluted for interferences. Tble 1 provides the concentrtion of compounds found to give flse positive test t the indicted concentrtion. 3.2 Other compounds hve been tested for cross rectivity with PCP, nd hve been demonstrted to not interfere with the specific kit tested. Consult the informtion provided by the mnufcturer of the kit used for dditionl informtion regrding cross rectivity with other compounds. 3.3 Storge nd use tempertures my modify the method performnce. 4.0 APPARATUS AND MATERIALS 4.1 PENTA RISc Test Kits (EnSys, Inc.), or equivlent. Ech commercilly vilble test kit will supply or specify the pprtus nd mterils necessry for successful completion of the test. 5.0 REAGENTS 5.1 Ech commercilly vilble test kit will supply or specify the regents necessry for successful completion of the test. 6.0 SAMPLE COLLECTION, PRESERVATION, AND HANDLING 6.1 See the introductory mteril to this chpter, Orgnic Anlytes, Section 4.1. 7.0 PROCEDURE 7.1 Follow the mnufcturer's instructions for the test kit being used. Those test kits used must meet or exceed the performnce indicted in Tbles 2-3. 8.0 QUALITY CONTROL 8.1 Follow the mnufcturer's instructions for the test kit being used for qulity control procedures specific to the test kit used. Additionlly, guidnce provided in Chpter One should be followed. 8.2 Use of replicte nlyses, prticulrly when results indicte concentrtions ner the ction level, is recommended to refine informtion gthered with the kit. 8.3 Do not use test kits pst their expirtion dte. 8.4 Do not use tubes or regents designted for use with other kits. 8.5 Use the test kits within their specified storge temperture nd operting temperture limits. 8.6 Method 4010 is intended for field or lbortory use. The pproprite level of qulity ssurnce should ccompny the ppliction of this method to document dt qulity. 9.0 METHOD PERFORMANCE 9.1 This method hs been pplied to series of groundwter, process wter, nd wstewter smples from industries which use PCP, nd the results compred with GC/MS determintion of PCP (Method 8270).
