Day 1 - Monday, 23 rd July 2018 Lab 307 Premise: Group One will use the chromatin and antibodies prepared in Cambridge and will involve a step by step guide to the process in the ChIP process, including a introduction etc. Group Two will be supplying their own chromatin and antibodies, so they will have a load of home-work prior to the practicals; this will also require at least some basic knowledge of the process to complete it. Notably group two will do the entire process, whilst certain aspects of the process will be gratuitous and be missed out for group one. 9.00-09:30 Welcome and Brief Introduction and Division into Two Groups Group 1: 09:30-10:00 Talk: Introduction to ChIP 10:00-11:00 Talk: Introduction to Practicals Group 2: 09:30-11:00 Open forum to discuss the home-work and review all quality control etc. It is also to gauge the quantity of samples each person wants to process etc. and to clear up exactly what they should be doing. It will be desired that each person in this group has an outline of their experimental design (ideally in excel) and should be happy for it to be modified nothing complicated but a template of the practical objective (no need for the hypothesis/question) 11:00-12.00 Lab practical (Groups 1+2): Chromatin Pre-clearing and Bead-Antibody Preparation 12.0-13.00 Lunch 13:00 15:00 Lab practical (Groups 1+2): Recover Chromatin and Wash Beads 15:00-17:00 Lab practical (Groups 1+2): Prepare Overnight Chromatin Immunoprecipitation Page 4 of 9

Day 3 - Wednesday, 25 July 2018 Lab 307 Group 1: Either come in late (e.g. 12:00 or we can have talks or open discussions) - the Library Prep kit being used is a straight forward and completely gratuitous process if the samples are not being sequenced; the method itself is guaranteed to work if manufacturer's instructions are followed.
