1 NAUČNI RAD Original Paper EFEKT ČUREKOTOVOG (NIGELLA SATIVA) ULJA IZ HRANE NA LIPIDE SERUMA KOD BROJLERA DIETARY BLACK CUMIN (NIGELLA SATIVA) SEED OIL AFFECTS SERUM LIPIDS IN CHICKEN BROILERS Hodžić Aida, Hrković Porobija Amina, Gagić A., Hamamdžić M., Kavazović Aida, Pašić-Juhas Eva, Kustura Aida, Goletić T. 1 Abstract The research was performed on 40 chicken broilers of Cobb provenience to investigate the effect of dietary black cumin seed oil on some blood biochemical parameters in chicken broilers. 40 chicken broilers who were included in the 42 days lasting experiment were divided into two groups, each one of 20 individuals. Group K was the control with no added oil in feed mixture and the second group P was the experimental one chickens were fed with 0,025 g of p.o. administered black cumin seed oil. Feed and water supplies were ad libitum. Microclimate conditions (light, temperature and airflow) were maintained according to the technological procedure. Blood samples were taken from the wing vein of all animals from both groups at age of 25, 32 and 39 days. The following blood serum biochemical parameters were determined: total lipids, triglycerides, total cholesterol, albumen and calcium. Key words: black cumin seed oil, chicken broilers, blood serum biochemical parameters, fatty acid composition of oil Kratak sadržaj Provedeno je istraživanje na brojlerima provenijence Cobb 500 kako bi se ispitao utjecaj čurekotovog ulja na neke biohemijske parametre krvi brojlera u tovu. Pokus je obuhvatio 40 tovnih pilića, podijeljenih u dvije grupe, svaka po 20 jedinki. Grupa K je bila kontrolna - bez dodatka ulja u hrani, a grupa P je bila pokusna. Svakom P piletu je tokom tova svakodnevno peroralno (p.o.) aplicirano 0,025 g čurekotovog ulja. Hranjenje i napajanje pilića tokom pokusa su bili ad libitum. Mikroklimatski uslovi (svjetlost, temperatura i strujanje zraka) regulirani su prema poznatim tehnološkim parametrima. Uzorci krvi su uzimani iz krilne vene od svih jedinki iz obje grupe, u dobi od 25, 32 i 39 dana. U krvnom serumu su određivane koncentracije sljedećih biohemijskih parametara: ukupni lipidi, trigliceridi, ukupni holesterol, albumin i kalcij. Čurekotovo ulje aplicirano p.o. je statistički značajno (P < 0,05) utjecalo na vrijednosti lipidnih komponenata seruma, najviše ukupnih lipida, ali ne i albumina i kalcija kod tovnih pilića. Jedan od mogućih razloga ovakvog nalaza je masna komponenta hrane, kao i masno-kiselinska kompozicija dodatog čurekotovog ulja. Ključne riječi: čurekotovo ulje, brojleri, biohemijski parametri krvnog seruma, masno-kiselinska kompozicija ulja Uvod Čurekotovo ulje se već stoljećima koristi u medicinske svrhe u zemljama Bliskog istoka, Azije i Afrike (1). Moderna nauka je njegovu ljekovitost počela proučavati prije nekoliko godina i došla do zaključka kako biljni ekstrakti i njihova ulja imaju širok spektar djelovanja, uključujući inhibitorno djelovanje na patogene, utjecaj na fiziopatologiju, antiupalna i antidijaroična svojstva (21). Čurekotovo ulje direktno djeluje na stabiliziranje većeg broja sistema, a prvenstveno imunološkog i endokrinog. Istraživanja (23) su pokazala da korištenje 0,3% čurekotovog ulja u hrani utječe na poboljšanje razvoja imuniteta u pilića. Peradarska industrija nalazi se pod sve većim pritiskom za proizvodnju visokokvalitetnih proizvoda. Antibakterijski dodaci hrani, kao što su antibiotici, koji se koriste već godinama, imaju za svrhu povećanje profitabilnost u peradarskoj proizvodnji i pomažu u kontroli patogenih bakterija u sluznici crijeva, čime se pozitivno utječe na povećanje stope iskoristivosti hrane (2). Međutim, kada se koriste antibiotici u prehrani životinja, postoji potencijal razvoja rezistencije patogenih bakterija, što je dovelo do zabrane korištenja antibiotika u ishrani ili promicanju rasta u domaćih životinja (26). U novije vrijeme, sve je veći broj istraživanja koja su usmjerena ka 24

3 pronalaženju dodataka hrani biljnog porijekla, kao što su eterična ulja i ekstrakti aromatičnih biljaka, kao alternativa za tradicionalne antibakterijske dodatke. Zbog svog antimikrobnog, antihistaminskog, antihipertenzivnog i antiupalnog djelovanja, sjeme, odnosno ulje sjemena čurekota je predmet interesovanja liječnika alternativne medicine, imunologa, kao i farmaceutskih kompanija. Čurekotovo ulje sadržava 58% esencijalnih masnih kiselina, uključujući omega-6 i omega-3 masne kiseline, koje su potrebne za formiranje prostaglandina E1, koji balansira i jača imunološki sistem, dajući mu potporu za suzbijanje infekcije i kontrolu hroničnih bolesti. Sjeme i ulje sjemena čurekota sadržava alkaloide, isparljiva ulja i niz esencijalnih nutrijenata, što ih čini prihvatljivim aditivom u obrocima za perad. Pri dodatku čurekotovog ulja u obrok za perad, ustanovljen je pozitivan efekt na unos hrane i tjelesnu masu (2). Polazeći od svega prethodno navedenog, cilj ovog istaživanja je ocijeniti utjecaj čurekotovog ulja na određene biohemijske parametre (ukupni lipidi, trigliceridi, ukupni holesterol, albumin i kalcij) u serumu pilića u periodu tova, koji bi u perspektivi mogli pružiti vrijedne informacije, prvenstveno za unapređenje proizvodnje, ali i za diferencijalnu dijagnozu hranidbenih poremećaja. Biohemijski parametri u serumu pilića mogu biti pod utjecajem različitih faktora, uključujući bolesti peradi (19, 13, 4), aditive u hrani (18), hranjive tvari (17), uvjete smještaja (7) i temperaturu okoliša (10). Materijal i metode Istraživanje u trajanju od 42 dana provedeno je u laboratorijima Veterinarskog fakulteta u Sarajevu. Četrdeset pilića provenijence Cobb 500 bilo je podijeljeno u dvije grupe, svaka po 20 jedinki. Prva grupa (K) je bila kontrolna, gdje su pilići hranjeni bez dodatka ulja, a druga (P) je bila pokusna. P pilići su tokom cijelog perioda tova svakodnevno peroralno (p.o.) tretirani uljem čurekota (0,025 g/pile). U laboratoriju As Vitas (Norveška) izvršena je analiza sastava masnih kiselina u čurekotovom ulju (C14:0- C20:0) nakon ekstrakcije sadržane masti i metilacije masnih kiselina prema Ulberthu i Henningeru (25). Određen je sadržaj sljedećih masnih kiselina (g/100g FA): C14:0- miristinska, C16:0-palmitinska, C16:1-palmitoleinska, C18:0-stearinska,C18:1-oleinska, C18:2-linolna, C18:3n-3-linolenska, C20:0-arahidonska, C20:1n-9-eikosenoična i C20:2n-6 eikosadienoična. Tokom tova, pilići su hranjeni trima vrstama namjenskih krmnih smjesa (starter, grover i finišer) za tov hibridnih jedinki Cobb 500, proizvođača stočne hrane Brovis Visoko. Za pokusni tov pilića primijenjen je podni način držanja, u skladu sa preporukama selekcionara. Hranjenje i napajanje su bili ad libitum. Svakodnevno je vršena klinička kontrola zdravstvenog stanja pilića. Tokom pokusa, pilići iz obje grupe su vakcinirani protiv njukastlske (NB) (11. dan) i gamborske (GB) (18. dan) bolesti. Vakcine su aplicirane u vodi za piće. Uzorci krvi su uzimani iz krilne vene, od svih jedinki iz obje grupe, u dobi od 25, 32 i 39 dana (uzorkovanje I, II i III). Na osnovu prethodno postavljenog cilja, vršeno je određivanje koncentracije sljedećih biohemijskih Veterinaria 61 (1-2), 23-33, Sarajevo

4 parametara: ukupni lipidi (g/l), trigliceridi (mmol/l), holesterol (mmol/l), albumin (g/l) i kalcij (mmol/l). Koncentracije biohemijskih parametara određivane su spektrofotometrijski na Beckmann DU-64 UV/VIS analizatoru s komercijalnim kitovima, proizvođača Human (Njemačka) i Semikem (Sarajevo). Statistička obrada podataka vršena je korištenjem gotovog softverskog paketa Microsoft Excel. Statistička značajnost razlika je ispitivana na nivou statističke značajnosti p < 0,05 t-testom za dva uzorka jednake varijanse ANOVA. Rezultati Rezultati mjerenja biohemijskih parametara prikazani su na grafikonima 1 do 5, a masno-kiselinska kompozicija čurekotovog ulja prikazana je u tabeli 1. Grafikon 1. Utvrđene su statistički značajne razlike za ukupne lipide kako između grupa tako i između uzorkovanja unutar iste grupe, a naročito između I i III uzorkovanja (graf. 1). Lipidi su u cirkulaciji vezani s proteinima u obliku hidrofilnih lipoproteina, čija je funkcija da putem krvi prenesu svoje lipidne sastojke. Kao biohemijski parametar, ukupni lipidi pokazali su se kao parametar koji je najviše varirao kako sa dobi tako i sa hranidbenim tretmanom. Faktori koji mogu utjecati na koncentraciju lipida plazme uključuju količinu i tip dijetarnih lipida, vrijeme proteklo nakon konzumacije hrane, dob životinje, hormonalnu ravnotežu i energetske potrebe. Kontrolna grupa je imala niže vrijednosti triglicerida kod sva tri uzorkovanja u odnosu na pokusnu, što se može pripisati utjecaju čurekotovog ulja, ali sa statističkom značajnošću između grupa samo u prvom uzorkovanju (graf. 2). Prema kraju eksperimenta, kod obje grupe ustanovljena je tendencija stabilizacije ovog parametra. Koncentracija holesterola pokazivala je tendenciju smanjenja sa smanjenjem dobi pilića u kontrolnoj grupi, sa statistički značajno višom vrijednošću u prvom, u odnosu na druga dva uzorkovanja (graf. 3). Selvaraj i sar. (22) su u svojim istraživanjima potvrdili ovisnost pojedinih biohemijskih parametara o dobi jedinke. U našem istraživanju, kontrolna grupa je u odnosu na pokusnu pokazala razliku u I i III uzorkovanju, s tim da se holesterol pokusne grupe pokazao kao vrlo stabilan parametar (graf. 3). Holesterol je strukturna komponenta staničnih membrana čije mjesto i opseg sinteze ovise o vrsti, dobi životinje i hranidbenim faktorima. Na nivo ukupnog holesterola mogu utjecati omega-3 masne kiseline, kao i odnos omega-3 : omega-6 masnih kiselina. Promjene koncentracije holesterola mogu se pripisati i sezonskim utjecajima i Veterinaria 61 (1-2), 23-33, Sarajevo

8 varijacijama u brzini biosinteze holesterola. Masne tvari se u ptica sintetiziraju u jetri, te odlažu u jetrenim, masnim i jajnim stanicama (8). Istraživanja koncentracije proteina u serumu pilića uglavnom se odnose na ukupne vrijednosti i omjer albumina i globulina, kao i omjer pojedinih frakcija u određenim patološkim stanjima. Ispitivanjem koncentracije albumina ustanovljene su najniže vrijednosti kod obje grupe u II uzorkovanju i sa statističkom značajnošću u odnosu na I i III uzorkovanje, ali bez statističke značajnosti između grupa (graf. 4). Ispitivanjem koncentracije kalcija u serumu pilića nije utvrđena statistički značajna razlika između grupa, niti između uzorkovanja (graf. 5). Koncentracija serumskog kalcija utječe na imuni odgovor i titar antitjela kod NB (5). Qaisar i sar. (20) su utvrdili postojanost različitih koncentracija serumskog kalcija između različitih sojeva. Analizom korištenog čurekotovog ulja detektovano je ukupno 11 masnih kiselina čije je učešće prikazano u tabeli 1. S čurekotovim uljem u organizam dospijevaju korisne, višestruko nezasićene masne kiseline (linolna i linolenska). Obogaćivanje pilećeg mesa linolnom i linolenskom kiselinom je moguće dodavanjem navedenog ulja (16, 24, 15), mada povećanje nivoa ne rezultira obavezno i adekvatnom konverzijom u masne kiseline. Količina i vrsta masnih kiselina prisutnih u hrani ima značajan efekt na metabolizam sterola kod peradi. Analiza masno-kiselinske kompozicije korištenog čurekotovog ulja pokazala je da je najzastupljenija linolna kiselina, a zatim slijede oleinska, palmitinska i stearinska (tab.1). Prema ranijim istraživanjima, smatralo se da linolna kiselina snižava nivo ukupnog holesterola više nego oleinska ili ugljeni hidrati. Valsta i sar. (26) su ustanovili da linolna i oleinska kiselina imaju primarno identične efekte na nivo holesterola. Jiang i sar. (11) iznose tezu o hipoholesterolemičnom efektu linolne kiseline hrane, ali koji zavisi i od drugih faktora, kao što su ukupan sadržaj linolne kiseline, njen odnos prema drugim lipidnim faktorima hrane, odnosi zasićenih i polinezasićenih masnih kiselina. Palmitinska kiselina se smatra masnom kiselinom koja povećava holesterol, dok se oleinska označava kao neutralna, jer ona djeluje na ukupni holesterol, slično ugljenim hidratima. Istina, oleinska kiselina snižava nivo plazmatskog holesterola kada zamjenjuje palmitinsku kiselinu hrane (3). Palmitinska kiselina povećava nivo holesterola u sastavu lipoproteina niske gustoće, LDL-holesterola (engl. low density lipoprotein cholesterol), paralelno s koncentracijom ukupnog holesterola, kada ona mijenja ugljene hidrate ili mononezasićene masne kiseline hrane (12). Suprotno palmitinskoj, stearinska kiselina ne povećava nivo ukupnog holesterola u serumu, i u poređenju s palmitinskom, stearinska je hipoholesterolemična (6), ali u poređenju sa linolnom je hiperholesterolemična (14, 9). Zaključci Koncentracije lipidnih komponenata seruma (ukupni lipidi, holesterol, trigliceridi), uz oscilacije između grupa unutar istog uzorkovanja, varirale su i sa dobi, i sa hranidbenim tretmanom. Količina i tip dijetarnih lipida, vrijeme proteklo nakon 30

9 konzumiranja hrane, hormonalna ravnoteža i energetske potrebe su faktori koji su mogli utjecati na lipidnu komponentu seruma. Obzirom na mogući utjecaj količine i kvaliteta masti iz hrane na resorpciju kalcija u provedenom tretmanu pilića, odsustvo statističke značajnosti razlika između pokusne i kontrolne grupe može se smatrati pozitivnim nalazom s aspekta eventualne primjene čurekotovog ulja u ishrani pilića. Dobiveni rezultati za koncentraciju proteinske komponente (albumin) mogu se dovesti u vezu s ulogom metabolizma bjelančevina u brzini prirasta tovnih pilića i mogli bi poslužiti kao dio metaboličkog profila u procjeni proizvodne sposobnosti, kao i zdravstvenog stanja pilića u periodu tova. Ova preliminarna istraživanja o upotrebi čurekotovog ulja u hranidbi tovnih pilića i njen utjecaj na biohemijske parametre predstavljaju dobru osnovu za daljnja istraživanja, a sve s ciljem rasvjetljavanja utjecaja čurekotovog ulja, odnosno njegovih sastojaka na metaboličke puteve kod pilića, ali i eventualnog povećanja proizvodnje i profitabilnosti u industrijskoj proizvodnji.
