1 IMPROVEMENT OF CHERRY PLANT GROWTH BY ENDOPHYTIC FUSARIUM ISOLATED FROM WEEDS Jelena Ilic, PhD Department of Plant Pathology Faculty of Agriculture in Osijek Croatia

2 experiment: investigation of the influence of five endophytic fungi belonging to genus Fusarium and isolated from symptomless weeds, on growth and development of cherry plants grown from tissue culture.

3 Endophytes: microorganisms, mostly fungi and bacteria live within plants intercellularly and/or intracellularly do not cause any visible manifestation of disease under normal circumstances (Bacon and White 2000).

4 During this association, none of the interacting partners is harmed, and the individual benefits depend on all organisms involved. Endophytic fungi that grow within their plant hosts without causing disease symptoms are relatively unexplored and unattended as compared with soil isolates and plant pathogens.

5 Biological diversity of endophytes is large and each plant species may be a host to a number of endophytes (Strobel 2003). According to Petrini (1986) endophytes have been found in all parts of all plants, including xylem and floem.

6 Endophytes play multiple physiological and ecological roles in the mutualistic association with their host plants: plant might provide nutrients to the microbe and microbe may produce metabolites that protect the host plant from attack by animals, insects or other microbes, improve its tolerance to environmental stress (drought tolerance, metal tolerance) and positively influence plant growth and development (Redman et al. 2001).

7 Previously identified isolates of Fusarium spp.isolated from symptomless weeds were used for additional analysis Fusarium spp. Fusarium oxysporum (61) Weed Abutilon theoprasti Med. Fusarium solani I (149) Sonchus arvensis L. Fusarium solani II (112) Chenopodium album L. Fusarium subglutinans (111) Chenopodium album L. Fusarium verticillioides (102) Chenopodium album L.

8 Pathogenicity of these isolates has been previously investigated in Ilic et al. (2012) and the above mentioned Fusarium isolates showed positive influence to wheat and maize development.

9 INOCULATION OF TISSUE CULTURE MEDIA WITH ENDOPHYTES (TREATMENT 1) The five selected species of Fusarium spp. were grown on PDA (4 Petri dishes for each isolate) for 14 days in growing chambers, at 22 C and 12 hour day/12 hour night regime. After 14 days mycelia were removed from the medium surface by scraping with sterile metal scalpel.

10 Since most of the isolates did not sporulate or produced only a small number of spores after incubation, the whole mycelia was blended with 160 ml of distilled water and refrigerated at 4 C.

11 20 ml of endophytes suspension was injected into a warm (approx. 40 Cº), still liquid introduction media trough injection with a filter Introduction media: Schenk & Hildebrandt Basal medium, Schenk and Hildebrandt,, 1972, Murashige Skoog medium (Murashige and Skoog 1962), BAP 6-Benzylaminopurine, NAA 1- Naphthaleneacetic acid, GA3 Gibberellic Acid, sucrose, Agar.

12 PREPARATION OF TISSUE CULTURE PLANTS As described by Pereira et al. (1999) apical meristems of cherry were obtained from vegetative buds of intact, two year old mother plants grown in the greenhouse. Surface of culture introduction buds were sterilized with Na-hypoclorite and 75% ethanol.

15 Meristems were aseptically excised and explants were placed into the introduction medium containing endophytic fungi. Light was provided with cool light fluorescent bulbs located 20 cm above the containers. Light intensity at the top of containers was 40 to 60 µmol m -2 s -1.

17 Micropropagated shoots were maintained in glass tubes, one per each tube, at 25 ºC, 16 hour fotoperiod and transfered biweekly.

18 When the plantlets were 2-4 cm tall, callus was removed and leaf surface reduced. Each plant was divided into at least two plants, depending on the development of the plantlet, and transfered to flasks containing multiplication media Multiplication media: macroelements, microelements and vitamins - Driver Kuniyuki medium (DKW) (Driver and Kuniyuki 1984), BAP, IBA Indole-3-butyric acid, agar, Myoinositol.

19 This procedure was repeated 6 times. After that, plantlets were transfered to rooting media Rooting media: macroelements, microelements and vitamins - MS medium, IBA, Sequestren 138 Fe 100 SG, agar.

20 50 shoots were placed into jars for root development and then cool stored in culture for 4 weeks to improve transplantation, according to results of Varshney et al. (2000).

22 Explants in each experiment were from the same subculture cycle. After 28 days in root development medium, rooted shoots were washed thoroughly to remove residual medium and re-inoculated with fungal suspension.

23 RE-INOCULATION OF CHERRY PLANTS WITH FUNGAL SUSPENSION (TREATMENT 2) In the transplanting stage, cherry explants were selected based on their uniformity in size and growth vigour. The selected 40 seedlings were artificially infected with fungal suspension. The five selected Fusarium isolates were prepared by the same procedure mentioned previously.

24 Dip inoculations were performed with modified method of Gera Hol et al. (2007) by simultaneously dipping the whole plants into a 500 ml beaker for one hour. Whole plants were dipped instead of only roots due to small plant size. Rooted plantlets were potted into plastic boxes with places for 40 plants and containig substrate with macroelements, microelements and perlit.

27 Plants in boxes were placed under a plastic tent on a bench in a fan and pad greenhouse, where high humidity was maintained (99% in the first week) using a humidifier. The temperature of the substrate was between 18 and 21 ºC and in the tent it was approx. 25ºC.

28 In vivo conditions were gradually introduced by reducing moisture and exposing plants to more light. Once a week plants were treated with fungicide Previcur Energy to prevent Pythium infection and with insecticide Dursban against mushroom fly Amanita muscaria (L.) Lam. Plants were fertilized once a week by root immersion on the tables with fertilizer Polyphid.

29 After 4 weeks of acclimatization, tents were removed and plants adjusted completely. Plants remained in controlled greenhouse for additional 30 days.

30 The following plant characteristics were measured: fresh weight, stem lenght (distance from pseudostem base to the point where the youngest leafe emerges from the pseudostem), number of functional leaves, width (at the widest point) and lenght of the largest leaf and root lenght were recorded. Dry weight was not recorded because the same plants were used for further research.

31 SECONDARY METABOLLITES IDENTIFICATION AND CHARACTERIZATION Fusarium were grown on PDA agar for two weeks under the same regime mentioned above. Four petri dishes were prepared for each of the five Fusarium species. After two weeks, 20 ml of methanol was added to each Petri dish, sealed with parafilm and left to rest for 24 hours. After 24 hours, methanol extracts were collected and methanol was evaporated. Extracts were analyzed at Fundación MEDINA center in Granada, Spain,

32 RESULTS Plants inoculated with F. solani 112 died during the tissue culture treatment. The remaining treatments showed visibly positive significant influence on growth parameters of cherry.

33 Fusarium solani 149 had the highest influence on most of the plant parameters. This isolate in treatment 1 improved leaf width by 166%, leaf lenght by 165%, stem lenght by 263% and fresh weight by 262%

35 The number of leaves showed only small statistical differences between some treatments but none of them was significant compared to control. During plant measurement we noticed that plant development was not correlated to number of leaves and that smaller plants tend to have more leaves.

37 In regard to leaf parameters, leaf lenght in six and leaf width in four out of eight treatments showed significant difference compared to control.

40 All treatments showed significant influence on stem lenght compared to control, where six values had very significant difference of p<0,0005.

42 Surprisingly, root lenght of the control was significantly higher compared to almost all treatments and the same can be said as for the number of leaves: root lenght is not correlated to plant lenght and weight. We noticed that small plants tend to have longer roots and vice versa.

44 Plant fresh weight showed four significant differences between treatment values and controls, and three of them were very significant (p<0,0005).

46 The largest number of high significant differences (p<0,005) compared to controls and all treatments had treatment 149.

47 For several of the variables, treatment 1 (media inoculation) had more significant influence on growth parameters than treatment 2 (media+plant inoculation). It is a surprising fact since treatment 2 included double infection with the same fungi and therefore would suggest a higher influence on plant growth.

50 Integracide A detected in F. solani 149 had a very high area percentage, and because isolate 149 showed the highest positive influence on growth parameters, we could think that Integracide A may be involved in plant growth improvement.

51 DISCUSSION Clonal plants grown under the same conditions should definitelly look alike with only small differences in their growth. And contrary to that, in our research differences among treatments were significant compared to controls. Difference in plant size was not so noticeable during in vitro conditions, but only after greenhouse growing.

52 Little research has been done so far to determine endophytic fungi living inside weeds. In regard to agricultural production, weeds growing inside or near agricultural fields are especially interesting, as wild weeds are more resistant to diseases than cultivated plants and might be an important bridge between pathogen and cultivated plants.

53 They might provide fitness enhancement to fungal species living inside them. In our previous research we have proven that endophytic fungi isolated from symptomless weed can act both as pathogens and beneficiaries towards cultivated plants (Ilic et al., 2012).

54 CONCLUSION In conclusion we can say that endophytic Fusarium species isolated from symptomless weed had significant influence on growth of cherry plants. Several secondary metabolites that were identified as major components of fungal extracts need to be investigated in order to determine their influence on plant growth promotion.
