RESEARCH EXPERIENCE UTSW-Dallas, TX, Postdoctoral research fellow, Trained in Dr(s) Vega/Grundy labs to o execute experiments to study the characteristics of human adipocytes from different depots (abdominal and gluteofemoral) o perform experiments to measure non-esterified fatty acids (NEFA), β-hydroxybutyrate (BHP) and Triglycerides in human plasma samples. Developed and wrote a research protocol titled Adipose Tissue Cellularity in Polycystic Ovarian Syndrome (PCOS). This protocol centers on role of obesity in relation in metabolic abnormalities in PCOS women. The primary aim of this pilot study is to determine whether women with polycystic ovarian syndrome (PCOS) have reduced number of subcutaneous adipocytes compared with women without PCOS. It is postulated that reduced cellularity is an underlying defect of the metabolic 2
3 syndrome, a syndrome that is characterized by central obesity, dyslipidemia, impaired glucose tolerance, hypertension, and prothrombotic and inflammatory state, in PCOS. This protocol has been approved by the IRB and by Moss Heart Foundation for funding. We are in the process of recruitment for the study; so far, we have attained 80% of the targeted sample size. Wrote a research project titled Body Fat content and distribution and Metabolic Risk Factors in Polycystic Ovarian Syndrome (PCOS). The purpose of this project is to examine the relative distribution of truncal-tolower body fat and of subcutaneous- to- intraperitoneal + retroperitoneal fat in PCOS cases and their age-and BMI-matched normal cycling controls. The design is cross-sectional study of a nested cohort from a multi-ethnic population-based sample (Dallas Heart Study). Measurements of body composition by dual x-ray absorptiometry (DXA), abdominal fat by MRI, lipids and lipoprotein cholesterol, NMR lipoprotein, insulin resistance (HOMA-IR) as well as leptin and adiponectin levels will be used to achieve the purpose of the project. The Reynolds Cardiovascular Clinical Research Center has provided us with the database. I have performed the statistical analyses associated with the manuscript and I am currently involved in the write-up of the manuscript. Wrote a review PCOS and its Complications: Is it Visceral Fat, Upper-tolower Body Fat Ratio or BOTH. The purpose of this review is to examine the evidence, in the past 10 years, regarding the association of each truncal-to-lower body fat distribution and visceral fat to PCOS. Wrote another review Adipocyte size and number in PCOS. The purpose of this review is to examine the number and size of adipose cells reported in the PCOS population. Started writing a grant titled Relation of Body Composition and Fat Distribution to Insulin Resistance in Adolescents of high risk ethnicity. The purpose of this grant is to examine differences in insulin resistance according to body composition and fat distribution in adolescents of high risk ethnicity (African Americans, Asian Indians and Hispanics). Strengthened oral and written communication skills by lecturing nutrition students and writing grants and manuscripts, and by presenting orally at scientific meetings University of Pittsburgh, Graduate School of Public Health, Pittsburgh, PA Graduate Research Assistant (PhD program). Carried out a Research Study Funded by a Grant From the National Cancer Institute (NCI) titled Molecular Epidemiology of Prostate cancer, Tobagonians. Trained in Dr Bunker s laboratory to carry out the following. I used organic solvents to extract antioxidants from serum collected in Trinidad 3
4 and Tobago. I then separated and quantitated individual antioxidants (alpha- and gamma-tocopherol; retinol; and the carotenoids: alpha- and beta-carotene, cryptoxanthin and lycopene) by HPLC. The components were monitored using a photodiode array detector and the internal standards were retinol acetate and tocopherol acetate. The results were evaluated in relation to prostate cancer. Developed and carried out a Research Study Funded by a Grant from the National Institute of Child Health and Human Development (NICHD) titled Genetics of Insulin Resistance: The PPAR Pathway Graduate Research Assistant (M.P.H program), AUB, Beirut, Lebanon Worked, under the supervision of Dr. Mary Deeb, on a research study titled The Mental Health Status of Sensorially Adolescents in the aftermath of War. Worked, under the supervision of Dr. Shaar, K., on a research study titled Prevalence of Gynecological Disease in Greater Beirut. OTHER WORK EXPERIENCE Network Director, MedNet Liban, Beirut, Lebanon Controlled and supervised the implementation of contractual agreements involving health care providers (Hospitals, Laboratories and pharmacies) and third party payers (Insurance Companies and Mutual Societies). Conducted a meticulous selection of network providers according to need. 6 June 2004-June 2006 Genetics of Insulin Resistance: The PPAR Pathway ; PI - Evelyn Talbott, Dr.PH ( NICHD R03 HD ). As an investigator in this grant, I developed the protocol to explore the relationship between PCOS genotype and insulin resistance by examining a well-established population of polycystic ovary syndrome cases and their first and second-degree family members. Specifically, I developed new markers of insulin resistance and inflammation that are specific to my doctoral thesis and proposed new scientific hypotheses. To test my hypotheses, I worked on recruiting families with polycystic Ovary syndrome, extracting DNA from collected blood samples and genotyping analyzing data and writing the manuscripts. Oct.2008-Present. Adipose Tissue Cellularity in Polycystic Ovarian Syndrome ; PI- Gloria L. Vega PhD/Grundy Scott, M.D., Ph.D (Funding Source: Moss Heart Foundation). As an investigator in this grant, I developed the protocol to determine whether women with polycystic ovarian syndrome (PCOS) have reduced number of subcutaneous adipocytes compared with women without PCOS. The hypothesis to be tested is that PCOS women have a deficiency of subcutaneous adipose tissue cellularity. It is postulated that reduced subcutaneous adipose tissue cellularity is an underlying defect of the metabolic syndrome in PCOS. The long-term goal of this project is to determine whether hyperandrogenemia in women causes reduced adipose tissue cellularity. GRANTS SUBMITTED I and Dr Jessy El Hayek developed and submitted to the CNRS a research project titled Prevalence of Pre-hypertension and Primary Hypertension among Lebanese School Students: a Cross-Sectional Study. The purpose of the project is to assess the prevalence of pre-hypertension and primary hypertension among Lebanese School Students as well as examine the association between elevated blood pressure and its anthropometric, dietary and lifestyle risk factors. IN PROGRESS I am in the process of writing a grant titled Relation between Body fat Composition and Distribution and Metabolic Syndrome in Lebanese Adolescents. The purpose of this grant is to examine differences in prevalence of metabolic syndrome between adolescents with upper-body fat and lower-body fat distributions. I am writing a grant titled Epidemiological Differences in Breast Cancer Incidence Rates between Female Muslims and Christians in Beirut. The specific goal in this grant is to develop the existing national cancer registry in Lebanon through implementing standardized guidelines for cancer registries; this will allow us to collect incidence data over a five year period and test our hypothesis that in Beirut, the age-standardized incidence rate of Breast Cancer in Female Muslims is lower than that in Female Christians (For Breast Cancer, ASIR Muslims<ASIR Christians). 6
