No. R5304 The contamination with the mold toxins T-2 and HT-2 toxin has been discussed as a serious health risk for human and animals for many years. As a result scientific EFSA committees advise EU-wide guidance levels for T-2 / HT-2 toxin in food and feed. T-2 and HT-2 toxin enter the food chain by grain containing food or feed and affect livestock conditions. Especially oats but also further grains like corn, wheat and barley may contain considerable concentrations of these two important mycotoxins. The new test system RIDA QUICK T-2 / HT-2 RQS (Art. No. R5304) increases the product portfolio of R-Biopharm with a fast and quantitative lateral flow test, which is characterized by its simple handling and its very rapid test procedure. This makes it particularly suitable for the time-sensitive quantitative grain acceptance analysis. The usage of organic solvents during toxin extraction is not necessary. A water based toxin extraction is sufficient and reduces the handling with organic solvents to a minimum. The test strips are quantitatively analyzed using the well-established RIDA QUICK SCAN reader (Art. Replicas can be created on all flat surfaces within the production area, and on solid foodstuffs. Our products Consistent sample preparation for milk proteins Efficient extraction of samples is the key to correct and reliable results. Milk proteins are difficult to extract, especially from processed breads and cookies. A consistent sample preparation is now available with RIDA Extractor 2, to ensure a better extraction. The extract can be used within the three different RIDASCREEN milk sandwich ELISAs. With the RIDASCREEN FAST Casein test system, a total of three different sample preparations are available for the user. Along with the RIDA Extractor 2, there is also the opportunity to further use the RIDA Extraction Solution (colorless), Art. Information from R-Biopharm Rhône, Scotland Launch of new immunoaffinity column for the analysis of sterigmatocystin R-Biopharm have launched a new and innovative immunoaffinity column, EASI-EXTRACT STERIGMATOCYSTIN (Art. No. RBRP125/RBRP125B). Sterigmatocystin is a precursor in the metabolic pathway for aflatoxin formation and is therefore structurally related. Like aflatoxin, sterigmatocystin is produced by over 50 fungal species including the Aspergillus fungi. Sterigmatocystin has been found in grains, grain based products, cheese and other commodities like green coffee and spices even though information on its occurrence in foods is limited. In 2013, the EFSA Panel on contaminants in the food chain were asked to assess the risk continued on page 5 page 4

5 continued from page 4 R-Biopharm AG to public and animal health related to the presence of sterigmatocystin in food and feed. Commodities to be tested included: grains including wheat, barley, rye, oats and rice grain based products including flour, bread and rolls, pasta, cereal flakes and muesli nuts and beer Proposed limits of quantification of 1,5 ppb have been recommended by EFSA. This low level of quantification can be difficult to achieve with UV-HPLC due to background noise and analysis of difficult commodities like cheese, coffee and spices can also be difficult with LC-MS/MS. Therefore, the use of an immunoaffinity column for sample clean-up and concentration of the toxin is required to ensure accurate quantification. The product has been tested on a range of cereals with work currently on-going for the development of additional application notes. Rejections by EU: Spices containing aflatoxins and ochratoxin A Spices contaminated with either aflatoxins or ochratoxin A continue to be rejected by the EU. Regulatory limits are set at low levels and confidence in measurement must involve proper sampling and accurate determination. Since January 2012, the EU has rejected a total of 86 consignments of spices which have been tested at the border inspection posts and found to contain levels of either aflatoxin or ochratoxin A, which have exceeded regulatory limits. In total some 15 different countries were involved in these export rejections, the majority (around 60 %) of the rejected consignments continued to originate from India, with 10 % from Indonesia and the remaining countries only having between one and four rejected consignments. A large number of rejections were for chilli pepper (paprika) and nutmeg, but also black pepper, garlic powder, ginger, curry powder, turmeric and liquorice were implicated in having unacceptable levels of these two mycotoxins. Spices are high value commodities, and therefore rejections by the EU has severe financial implications for the exporters, who not only have to bear the material costs of the spices themselves, but also pay the costs of sampling/analysis, storage at the port and ultimately the costs of destruction, if this course of action is taken by the authorities. However, these rejections can be avoided if spices are sampled and analysed strictly following EU regulations before they are exported. Only consignments in compliance with EU limits should be considered for export. Commission Regulation (EU) No. 165/2010 sets limits of 5 µg/kg for aflatoxin B1 and 10 µg/kg for total aflatoxins in Capsicum spp. (chillies, chilli powder, cayenne and paprika), Piper spp. (white and black pepper), Myristica fragrans (nutmeg), Zingiber officinale (ginger), Curcuma longa (turmeric) as well as mixtures containing one or more of any of these individual spices. Commission Regulation (EU) No. 105/2010 sets a limit of 15 µg/kg for ochratoxin A in the same specified spices, and a limit of 20 µg/kg for liquorice (Glycyrrhiza glabra, Glycyrrhiza inflate and other species). Methods of analysis for aflatoxins and ochratoxin A in spices have received less attention than other foodstuffs. Although methods have been published and single laboratory validation conducted, there are no official methods e.g. AOAC International or CEN. Spices are complex matrices and good sample clean-up as provided by an immunoaffinity column is essential prior to HPLC determination. Immunoaffinity columns supplied by R-Biopharm such as AFLAPREP, OCHRAPREP and AFLAOCHRA PREP have been successfully used for the analysis of aflatoxins and ochratoxin A in a wide variety of spices. A strong EU focus will remain on mycotoxins in spices and collaborative studies leading to new official methods are planned to be conducted during 2014/15. page 5
