3 Questions and Hypotheses The ability of drugs of abuse to increase brain DA concentration in limbic brain regions is considered crucial for their reinforcing effects. Drugs of abuse increase DA in naive as well as in addicted subjects. Why some people abuse drugs, whereas others do not? DA involvement in drug addiction is likely to be mediated by means of functional and structural changes in the circuits that are modulated by DA, including the frontal cortex.
