1 User Bulletin 7500 Fast Precision Plate Holder for MicroAmp Tube Strips December 19, 2007 SUBJECT: Using MicroAmp Fast 8-Tube Strips (0.1-mL) on the 7500 Fast Precision Plate Holder for MicroAmp Tube Strips (PN ) In This User Bulletin This user bulletin covers: Overview Materials Preparing MicroAmp Fast 8-Tube Strips (0.1-mL) Overview This bulletin describes how to use MicroAmp Fast 8-Tube Strips (0.1-mL) on the 7500 Fast Precision Plate Holder (PPH) for MicroAmp Tube Strips. The new 7500 Fast PPH for MicroAmp Tube Strips (Figure 1) is for use only with MicroAmp Fast 8-Tube Strips (0.1-mL). It cannot be used with MicroAmp Fast Optical 96-Well Reaction Plates or MicroAmp Fast Reaction Tubes with Caps (0.1-mL). Figure Fast Precision Plate Holder for MicroAmp Tube Strips (PN )

3 Preparing MicroAmp Fast 8-Tube Strips (0.1-mL) IMPORTANT! Please follow these instructions carefully to reduce the risk of crushed tubes when using the 7500 Fast PPH for MicroAmp Tube Strips. IMPORTANT! Carefully handle MicroAmp Fast 8-Tube Strips (0.1-mL), shown on page 2. To minimize bending or misaligning tube strips, place the tube strips in the MicroAmp 96-Well Base (shown on page 2) for capping, and use the MicroAmp Cap Installing Tool (Handle), shown on page 2, to cap the tubes. To prepare MicroAmp Fast 8-Tube Strips (0.1-mL): 1 Place the uncapped MicroAmp Fast 8-Tube Strips in the MicroAmp 96-Well Base (Figure 2). 4 2 Use the MicroAmp Cap Installing Tool (Handle), Figure 3, to attach the MicroAmp Optical 8-Cap Strips to the tubes. Slip your fingers through the tool, with the grooved side of the tool facing down. Hold the strip of caps over the row of tubes and, with a rocking motion, use the grooved side of the tool to push and seal each cap firmly in place (Figure 4). Repeat for the remaining caps in the row, then for all remaining rows. See Figure 5 on page 5. 5 Figure 5 Seven MicroAmp capped Fast 8-Tube Strips in the MicroAmp 96-Well Base 3 After capping the tubes, carefully remove the capped MicroAmp Fast 8-Tube Strips from the MicroAmp 96-Well Base by pushing up on the tubes from the underside of the 96-Well Base (Figure 6). 6 4 Verify that the MicroAmp Fast 8-Tube Strips are straight and that each tube is in line with the adjacent tube. View the tube strips from the top, side, and end and verify the tube strips are not bent or twisted. If a tube strip is bent or twisted, manually unbend or untwist it. 5 Insert two fully capped, empty MicroAmp Fast 8-Tube Strips in the far left and far right columns of the PPH (Figure 7). The empty tube strips evenly distribute the clamping load applied to the sample tube strips during processing. Carefully insert, in pairs, tube strips containing samples in the PPH, starting from the center and moving out (see Figure 8 on page 7). A minimum of two tube strips and a maximum of six tube strips can be run on the PPH at one time (Figure 9 on page 7). 6

Make sure that all the caps are lying flat. 8 Slowly close the drawer of the 7500 Fast System (see Figure 10 on page 8) to avoid shaking the tube strips and jarring them out of position before processing. 7
