3 PROSTAQUINONA ESTUDOS CLÍNICOS Evaluation of a Therapeutic Alternative for Telogen Effluvium: A Pilot Study Background/Aim: Telogen effluvium (TE) is a scalp disorder characterized by the thinning or shedding of hair resulting from the early entry of hair in the telogen phase. Nigella sativa (NS) is a dicotyledonous belonging to the Ranunculacae family. It has been shown that its major constituent, tymoquinon (TQ), exerts anti-oxidant and antiinflammatory effects by inhibiting proinflammatory mediators, such as cyclooxygenase and prostaglandin D2. The aim of this study is to evaluate the efficacy of NS essential oil as a potential treatment for TE, a pathology characterized by a significant inflammatory component. Study Design/Methods: Twenty patients affected by TE for this double-blind, placebo controlled and randomized study were enrolled. Ten of these patients were treated with a scalp lotion containing 0.5% NS, daily for 3 months, while the other ten patients were treated with placebo daily for 3 months. Videodermatoscopic analysis (Trichoscan Dermoscope Fotofinder ) and evaluation of three independent dermatologists were performed before treatment (T0), after 3 months of treatment (T3) and at the 6 months follow-up (T6). Results: The results showed a significant improvement in 70% of patients treated with NS. Videodermatoscopic analysis showed a significant increment of hair density and hair thickness in patients treated with NS. NS was also able to reduce the inflammation observed in the majority of patients affected by TE. Conclusions: The results of this study suggest that NS can be considered potentially useful for the treatment of TE. The anti-inflammatory, analgesic and antipyretic activity of Nigella sativa. The aqueous extract of Nigella sativa (N. sativa) was investigated for anti-inflammatory, analgesic and antipyretic activities in animal models. The extract has an anti-inflammatory effect demonstrated by its inhibitory effects on Carrageenan induced paw edema. It also produced significant increase in the hot plate reaction time in mice indicating analgesic effect. However, N. sativa crude suspension had no effect on yeast induced pyrexia. This study therefore, supports its use in folk medicine both as analgesic and anti-inflammatory agent and calls for further investigations to elucidate its mechanism of action.
4 Fixed oil of Nigella sativa and derived thymoquinone inhibit eicosanoid generation in leukocytes and membrane lipid peroxidation. Samples of the expressed fixed oil from different sources of Nigella sativa seeds were examined by thin-layer and gas chromatography for content of fixed oils and thymoquinone, and these substances were tested as possible inhibitors of eicosanoid generation and membrane lipid peroxidation. The crude fixed oil and pure thymoquinone both inhibited the cyclooxygenase and 5-lipoxygenase pathways of arachidonate metabolism in rat peritoneal leukocytes stimulated with calcium ionophore A23187, as shown by dose-dependent inhibition of thromboxane B2 and leukotriene B4, respectively. Thymoquinone was very potent, with approximate IC50 values against 5-lipoxygenase and cyclooxygenase of < 1 microgram/ml and 3.5 micrograms/ml, respectively. Both substances also inhibited non-enzymatic peroxidation in ox brain phospholipid liposomes, but thymoquinone was about ten times more potent. However, the inhibition of eicosanoid generation and lipid peroxidation by the fixed oil of N. sativa is greater than is expected from its content of thymoquinone (ca. 0.2% w/v), and it is possible that other components such as the unusual C20:2 unsaturated fatty acids may contribute also to its anti-eicosanoid and antioxidant activity. These pharmacological properties of the oil support the traditional use of N. sativa and its derived products as a treatment for rheumatism and related inflammatory diseases. Formulation and finding out the efficacy of the herbal hair oil over simple coconut oil (purified)-a formulation and clinical study in Bangladesh Hair fall is a very common phenomenon and a matter of concern within whatever young or aged. This study is formulated a herbal hair oil and made a clinical trial over selected person under the research and development unit at Routi Herbal Limited. To see the efficacy of the experimental oil made up of Kala Jera Toil (Nigella sativa) Narkal Toil (Cocos nucifera) Amloki (Emblica officinalis) Henna (Lawsonia alba) Durba Ghas (Cynodon dactylon) Mathi (Trigonella foenum-graecum) in different concentration over the simple Coconut Oil (purified). Total 90 volunteers man and women were selected for 90 days and arranged in 3 groups, one for experimental oil recipient, one for control group another for the simple Coconut Oil (purified) recipient. And it is found that hair fall reduced to 76%, 72%, 67%, 59%, 32%, 0% on 15days, 30 days, 45 days, 60 days, 75 days, and 90 days by using of this experimental herbal hair oil. Aims of the study: To find out the efficacy of test herbal hair oil over simple Coconut Oil (purified) to reduce the hair falls. Nigella sativa oil, nigellone and derived thymoquinone inhibit synthesis of 5-lipoxygenase products in polymorphonuclear leukocytes from rats. In the present study, Nigella sativa oil (NSO), nigellone (polythymoquinone) and derived thymoquinone were studied to evaluate their effect on the formation of 5-lipoxygenase (5-LO) products from polymorphonuclear leukocytes (PMNL). NSO produced a concentration dependent inhibition of 5-LO products and 5-hydroxy-eicosa-tetra-enoic acid (5-HETE) production with half maximal effects (IC50) at 25±1 μg/ml, respectively 24±1 μg/ml. Nigellone caused a concentration-related inhibition of 5-HETE production (IC50: 11.9±0.3 μg/ml). Moreover thymoquinone, the active principle of NSO inhibited the production of 5-LO products (IC50: 0.26±0.02 μg/ml) and 5-HETE production (IC50: 0.36±0.02 μg/ml) in a similar way. The effects are probably due to an antioxidative action. The data may in part explain the effect of the oil, its derived thymoquinone and nigellone in ameliorating inflammatory diseases.
5 Black cumin seed essential oil, as a potent analgesic and antiinflammatory drug. The steam-distilled essential oil of Iranian black cumin seed (Nigella sativa L.) was investigated for its composition and analgesic and antiinflammatory properties. After oil analysis by GC/MS, 20 compounds were identified in the oil, obtained in 0.4% (v/w) yield. Among them, para-cymene (37.3%) and thymoquinone (13.7%) were the major components. Acetic acid-induced writhing, formalin and light tail flick tests were used for assessment of analgesic activity. Antiinflammatory activity was evaluated using carrageenan-induced paw oedema in rats and croton oilinduced ear oedema in mice. Black cumin seed essential oil (BCSEO) was found to produce a significant analgesic effect in acetic acid-induced writhing, formalin and light tail flick tests. Naloxone, an opioid antagonist, could not reverse the analgesic effect observed in the formalin test. Although oral administration of BCSEO at doses of 100, 200 and 400 µl/kg did not exert a significant antiinflammatory effect in the carrageenan test, i.p. injection of the same doses significantly (p < 0.001) inhibited carrageenan-induced paw oedema. BCSEO at doses of 10 and 20 µl/ear could also reduce croton oil-induced oedema. It seems that mechanism(s) other than opioid receptors is (are) involved in the analgesic effect of BCSEO since naloxone could not reverse this effect. Both systemic and local administration of BCSEO showed antiinflammatory activity.
