Sericologia, 27(2), Impact of the research conducted either on the advance of knowledge or its practical significance Studies on the influences of disturbance, light and darkness, substratum on the oviposition behavior are of practical significance. The disturbance, light and rough substratum has a negative influence on the rate of egg laying. The use of plain substratum, maintenance of total darkness without disturbance in the grainages is cost effective as it saves a day labor (Manjulakumari D., 2001; D. Manjulakumari and Geethabali. 1991). For the first time, a reduced methylene blue technique was applied to study the innervation of hair sensilla on the ovipositor of silk moth by injecting the dye into the live animal. The technique revealed that each hair sensilla is innervated by a single bipolar neuron with the dendrite ending at the base of sensillum, a characteristic of mechanoreceptors. The axons of each nerve cell innervating these mechanoreceptors form a nerve bundle that enters the 9th abdominal ganglion (D. Manjulakumari and Geethabali, 2004) Egglaying is a segmental reflex in silkmoth and the last abdominal ganglion mediates this behavior. Brain is inhibitory for the function of egg laying in silkmoth. By isolating the last abdominal ganglion from the higher centers of nervous system the virgins are made to mimic the mated moths (G Varalakshmi, Geethabali and D. Manjulakumari. 1996). Mating is not a must for egg laying in silkmoths as virgins also lay eggs, but there is lot of difference in the egg laying behaviour of mated and virgin moths. Four parameters have been identified to quantify the egg laying behaviour and they are, the initiation of egg laying, rate of egg laying, total number of eggs laid and the life span (D.Manjulakumari and Geethabali, 1996 D. Manjulakumari and Geethabali, 1993, Geethabali and D. Manjulakumari, 1987). Very few laboratories in India have undertaken work on insect peptides/proteins in relation to reproduction in general and egg physiology in particular. The occurrence of an oviposition stimulating factor which also stimulates ovulation has been demonstrated in silkmoth Bombyx mori in our laboratory. Virgin moths injected with isolated male accessory gland factor could be made to exhibit mated behavior with respect to the pre-oviposition period, rate of egg laying and the number of eggs laid. (Bali and Manjulakumari, 1987; Manjulakumari and Bali, 1991, 1996). The chemical factor is identified as a protein with a molecular weight of 45KD. Male contribution of nutrients like proteins, lipids and glycogen to the female at the time of mating is evident from our studies on silk moth Bomyx mori. These nutrients provide energy for the post mating activities of female such as oviposition. The contribution of nutrients by male to female is of highly significance in lepidopteran insects which do not feed during their adult life (Manjulakumari D and Kumar, 2007). Two peptides in H.armigera, one that stimulated egg maturation and oviposition and the other that inhibited behaviors like calling and mating in treated virgin females have been isolated in our laboratory. (Shobha et al., 2008).
5 Identification of these peptides which modify the female behaviour opens up the possibility of employing them as biopesticides by introducing these peptides to virgins and thus inhibiting pre-mating and inducing Post- mating activities in virgins. This could be achieved through recombinant DNA technology by expressing these peptides possibly in baculovirus. The gene encoding chitinase protein in Helicoverpa armigera has been isolated and the protein is successfully expressed in bacteria (E. coli) and Yeast (Pichia pastoris) system. We have demonstrated the antifeedancy, oviposition deterrence and ovicidal activity of Calendula officinalis L. extracts against Spodoptera litura a polyphagus pest on Agicultural crops. Two compounds Alpha-antiarin and Oryzanol-A responsible for oviposition deterrent and ovicidal activity against the pest have been isolated which have potential to be used as biopesticides.
