11 The Culprit: Inadequate Kit Storage New kits originally stored at a satellite lab Kits stored in a frost-free freezer for months Should be stored at -80 (ideal) Not discovered for ~1 month Precipitate in solution 3 19 samples affected Obtained new kit from PTC Data was BEAUTIFUL again!
12 Types of Profiles Obtained (Overall) 55 samples tested to date (110 fractions) Various substrates and times post-coital Sperm fractions: mostly single-source male Non-sperm fractions: male, female, and mixtures
13 Breaking Down the Data: Sperm Fractions (Overall) Profiles obtained from 62% of samples tested 79% of these were single-source male 36% of samples stopped at quant 13 different cases affected (20 samples) All but 4 samples could be explained PSA cards too sensitive (no sperm cells detected) Multiple samples in same case stopped at quant (vasectomy?) Extracted with the problem kit
14 Types of Profiles Obtained (Without Bad Kit Data) 36 samples tested (72 fractions) Sperm fractions: predominately single-source male Non-sperm fractions: male, female, and mixtures (no change from before)
