Approximately 15% of couples attempting to conceive over a period of 1 year are unable to become pregnant. A male factor is contributory in nearly 40% of cases. (Thonneau et al., 1991). Unknown Both 10% 10% Female 40% Male 40%

Chromosomal abnormality & ICSI outcomes Since its introduction in 1992 (Palermo et al., 1992), intracytoplasmic sperm injection i (ICSI)has turned out to be one of the most successful flassisted reproductive techniques. This has raised the question of the genetic consequences of ICSI, not only with regard to the karyotype of the children born, but also to a possible predisposition of the infertile couple (Silber et al., 1995; Chandley and Hargreave, 1996).
Several studies have shown a higher incidence of chromosome abnormalities in infertile males, with an overall incidence of 7.1% constitutional aberrations. Most frequently, sex chromosome aneuploidies have been reported. As a consequence, in cases of male infertility it is now common practice in most in-vitro fertilization (IVF) centres to perform a cytogenetic screening of the male partner prior to ICSI treatment.
It has previously been recognized that the outcome of ICSI might ih be ifl influenced by the occurrence of chromosome abnormalities in the female partner.
Iranian researchers reports Chromosomal Abnormalities in Iranian Infertile Males who are Candidates for Assisted Reproductive Techniques. Hamid Gourabi 2007 Chromosomal anomalies in infertile azoosperm and oligosperm men. mozdarani,2000 Chromosomal studies in infertile men. Akbari Cytogenetic analysis of sperm nucleolus components of Iranian normal and sub-fertile individuals using zona free hamster oocytes. Mozadarani 2004.
The role of sperm aneuploidy as a predictor of the success of intracytoplasmic sperm injection (J.D.M. Nicopoullos, 2008). Success of intracytoplasmic sperm injection in couples with male and/or female chromosome aberrations in lymphocytes. (P.A. Almeida1 2011).
The study of relationship between chromosomal abnormality in lymphocyte cells of infertile men with intra cytoplasmic sperm injection outcomes?
Determining the frequency of lymphocyte chromosomal abnormality of infertile men. The study of the relationship between frequency of chromosomal abnormality and ICSI outcome.
Methods In this research 80 oligosperm and 30 azoosperm patients karyotypes analyzed. To do so, heparinated blood samples were cultured in RPMI-1640 medium, then after chromosome spread preparation. At last 20 metaphases were examined by trypsin (GTG) banding. Finally, we compared the results of chromosomal analysis with the results of ICSI outcome.
Results Chromosomal abnormality was seen in 10.7% of samples. The majority of chromosome abnormalities were numerical sex chromosome changes in azoosperm samples. The frequency of oocyte failed fertilization rate in patients with chromosomal abnormality was 25% but in samples without chromosomal abnormality was 9.2% with significant difference (P<0.05).
The analysis of pregnancy frequency in patients wives showed that pregnancy frequency in patients with chromosomal abnormality was 11%, but in samples without chromosomal abnormality this frequency was 21.6% with significant difference (P<0.05).

The studies carried out so far in this area were mainly focused on chromosome abnormalities i in ICSI candidate parents. For studying Chromosome abnormalities and Intra-cytoplasm sperm injection relations there has been only some researches using FISH technique, which is a highly-cost technique
There is only one research on lymphocyte samples of infertile couples in which the relationship between Chromosome abnormalities and Intra-cytoplasm sperm injection has been studied (Tiboni GM, 2011). To initiate this line of research we started to study infertile ICSI male candidates.
The current research results show the role of aneoploidy study in blood samples of parents who intended d to use ICSI. In future studies there should be investigations on more samples and/or on infertile couples (husband & wife) who are candidates for using ICSI. In this case, it is possible to use this technique along with genetic consultant as a method to predict success rate after using ICSI.
