This product is classified as flammable liquid Category 3 (Warning, flammable liquid and vapor) because more than or equal to 10%of ethanol is contained in this product and 0.04% Phenolphthalein ethanol solution (40) manufactured by Wako Pure Chemical Industries Ltd., which has a similar composition to R-1 reagent, is classified as Category 3 in MSDS No. JW (revised ). 10. Stability and reactivity Avoid leaving in a place where high temperature, humid or under direct sunlight. No data on flash point, ignition point, explosion limits and dangerous/hazardous decomposition product is available. Although ethanol vapor generates explosive mixed gas with air, stable under normal handling conditions and no dangerous reaction is expected to occur under specific conditions. 11. Toxicological information No data on mixture is available. Data on R-1, R-2, R-2 (C) and R-2 (D) reagents are shown below. R-1 reagent Phenolphthalein (no data on solution): Acute toxicity (Oral): Classification is not possible because of data lack; Oral-rat LD 50 > 1,000 mg/kg (RTECS, Access in Aug 2007) and no other data is available. Skin corrosion/ irritation: Classified as Category 2 based on data; it is reported that severe edema in palpebra and echymotic hemorrhages in conjunctiva in humans (HSDB, 2004). Respiratory or skin sensitization Classified as Category 1 because it is reported that Causes allergic skin reaction in humans (HSDB 2004, IARC ). Germ cell mutagenicity: Classified as Category 2 based on data; Negative in vitro reverse mutation study and in vitro sister chromosome exchange study (HSDB 2004, NTP TR465, 1996). Although in an Ames and in vitro sister chromosome exchange studies are negative, in vivo mouse micronucleus study in peripheral blood and in vitro chromosome aberration studies are positive (IARC ). Carcinogenicity: Classified as Category 2 because of facts that IARC Group 2B (Possibly carcinogenic to humans) (IARC 76, 2000) and NTP-R (NTP, 2005). Reproductive toxicity: Classified as Category 2 based on data; In a mouse reproductive toxicity study by oral exposure, decreased weight of testes and decreased sperms number, decreased

4 4/7 pregnancy dates and number of new born child, decreased body weight in child animals were observed where no maternal toxicity was reported (IARC ). Specific target organ toxicity (repeated exposure): Classified as Category 1 (digestive organs, kidneys and nervous system) based on data; stomach ache and abnormality in digestive organs in humans after chronic exposure, effects on kidneys and central nervous system were caused by chronic exposure in humans (IARC ). Ethanol (no data on solution): Acute toxicity (oral): Rat-oral LD 50 = g/kg-bw, 5g/kg-bw (DFGOT vol.7 (1996, p148) and Patty 5th, (2005, p385)). Acute toxicity (inhalation: vapor): Rat-inhalation LC 50 = 20,000ppm/10H (RTECS (2004)) on which data can be considered as a vapor inhalation test because saturated vapor concentration of ethanol at 20 ºC is 56,580ppm. Furthermore, 20,000ppm/10H * 10/ 4 = 31,600ppm/4h > 12,500 ppm (gas 5,000 ppm (category 4) * 2.5). Acute toxicity (inhalation: dust-mist): Rat-inhalation LC 50 (4h) = ca 63,000ml/m 3 = 63,000ppm (definition by DFG, DFGOT (1996)) exceeds saturated vapor concentration of ethanol at 20 ºC of 56,580ppm and is classified as a mist inhalation test. Skin corrosion/ irritation: Not irritating in an OECD TG 404 and American Guideline tests (DFGOT (1996)). Serious eye damage/ eye irritation: Classified as moderate according to OECD TG 405 and Draize tests (DFGOT (1996), and it is reported that disorder of cornea epithelium and conjunctival congestion in human have been recovered in 1-2 days (ACGIH (2001)). Germ cell mutagenicity: Positive in rats and mice dominant lethal tests and induction of heteroploidy in mice germ cell (DFG (1999) and IARC (1988)). Carcinogenicity: IARC classifies as a Group 1 alcohol beverages is human carcinogen. This is based on the fact that there is a relationship of cancer in liver and esophagus and alcohol beverages which can be concluded from large numbers of epidemiological studies regarding regular intake of alcohol beverages (DFGOT (1996)). On the other hands, ACGIH classifies ethanol A4 (not classifiable as a human carcinogen) as a toxicant in occupational settings (ACGIH, 1996). In this MSDS, it is evaluated that not classified based on the classification made by ACGIH and according to the technical guidance manual because classification should not be made on hazards of alcohol beverages but focused on hazards of ethanol. Reproductive toxicity: Large number of teratogenicity in human child or other adverse effects are reported which are caused by heavy drinking of alcohol beverages (DFGOT (1996)). Specific target organ toxicity (single exposure): Ingestion of ethanol causes disorder of central nervous system, headache, tiredness, loss of concentration in humans (ICSC (2000)) and acute poisoning may cause death (DFGOT (1996)) and inhalation of 5,000ppm (9.4 mg/l) caused respiratory irritation, lethargy and sick sleep in humans (ACGIH (2001)). Specific target organ toxicity (repeated exposure): Classified as Category 1 (liver system) based on data; Chronic and large amount of consumption of alcohols cause systemic toxicity in almost all organs in humans and the target organ is liver. Disorder may initiates from fatty degeneration and results in necrosis, fibrosis and liver cirrhosis. (DFGOT (1996)) Classified as Category 2 (nervous system) based on data; Withdrawal symptoms of alcoholism: tremor, epilepsia and distraction. (HSDB (2003)) Water: Acute toxicity: Oral-rat LD 50 >90 ml/kg (used 90g/kg for the calculation of ATEmix below) R-2 (C) and R-2 (D) reagents Sodium hydroxide (no data on solution): Acute toxicity: Interperioneal-mouse LD 50 = 40 mg/kg, Oral-human LDLo = 1.57 mg/kg, Oral-rabbit LDLo = 500 mg/kg Skin corrosion/ irritation: 0.5% and higher solution is irritating (SIDS, 2002), severe corrosion to human skin (DFGOT vol.12, 1999). 8% and higher solution causes corrosion to pig skin (SIDS, 2002). 5% and 4 hours administration causes severe necrosis to rabbit skin (ACGIH 7th, 2001;

5 5/7 PATTY 5th 2001). Serious eye damage/ eye irritation: Human severe, serious hazard (ACGIH 7th 2001, DFGOT vol , PATTY 5th 2001). Rabbit corrosive 1.2% and higher (SIDS 2002). Respiratory or Skin sensitization: Not classified because it is reported no skin sensitization in a human sensitization test (SIDS 2002; IUCLID). Germ cell mutagenicity: Negative in vivo mouse bone marrow micronucleus study and in vitro Ames study (SIDS 2002, DFGOT vol ). Not Classified. Specific target organ toxicity (single exposure): Irritating to respiratory organs and airways in humans. Causes lung edema. (SIDS 2002, ACGIH 7th 2001, DFGOT vo , PATTY 5th 2001). GHS classification results of R-1, R-2 (C) and R-2 (D) reagents as mixtures are shown below.
