2 The power of two: fusion DNA polymerases Fusion DNA polymerases are genetically engineered enzymes comprising two constituent proteins: a DNA polymerase and double-stranded DNA binding protein. These chimeric proteins are powerful enzymes that have significantly improved modern PCR by enhancing its efficiency and enabling a variety of new applications. The best-known and most widely used fusion PCR enzyme is the Thermo Scientific Phusion High-Fidelity DNA Polymerase. Since its introduction in 2003, Phusion High-Fidelity DNA Polymerase has set a high benchmark for high-performance PCR. Phusion products continue to be referenced in thousands of publications and have become some of the most chosen DNA polymerases for a multitude of applications due to their robustness, high fidelity (52x over Taq DNA polymerase), speed and inhibitor tolerance. The green format of Thermo Scientific DNA polymerases is a combination of DNA polymerase and green reaction buffer. The green buffer includes a density reagent and two tracking dyes, and enables direct loading of reaction mixtures on gels, thus simplifying PCR workflows. The buffer does not interfere with the performance of DNA polymerases and is compatible with downstream applications including DNA sequencing, phosphorylation, ligation and restriction digestion.

Proofreading DNA polymerases are unable to incorporate dutp into a growing DNA strand, and therefore to amplify uracil-containing templates. This is because they have a so-called uracil-binding pocket, which by binding to uracil residues inhibits polymerase activity and as a consequence PCR.
