NeuroTox Prenatal exposure to toxicants and childhood neurodevelopmental disorders and cognitive functions Heidi Aase, PhD NeuroTox opening seminar October 18, 2017

Background Neurodevelopmental disorders ADHD (~5%) Autism (~1%) Neurological disorders Epilepsy (~0,7%) Cerebral Palsy (~0,2%) Cognitive dysfunctions: IQ, working memory, language

Background Etiology: genetic and non-genetic factors Exposure to environmental toxins in the womb can affect child development Very little knowledge of the potential harming effects of environmental toxins on brain development and cognitive functions, or risk of neurological or neurodevelopmental disorders in children Preventable risk factors?

Questions Cd MeHg As Mn Is there a link between elevated levels of environmental toxins in the mom during pregnancy and neurodevelopmental disorders or cognitive dysfunction in the child? What about mixtures of several environmental toxins («cocktail effects»), also when they are all low level exposures? Which mechanisms are involved why may exposure during pregnancy potentially lead to developmental deviations or illness in the child?

Our study sample: Mom-Child pairs MoBa children & mothers Diagnostic groups (cases): ADHD (2258) Autism (742) Epilepsy (608) CP (248) Control group (<2000) Clinical sub-studies (2227)

Prenatal exposure PFAS Metals Outcomes ADHD, Autism, CP and Epilepsy Cognitive functions Language development Thyroid hormones in newborn

Prenatal exposure PFAS Metals Outcomes ADHD, Autism, CP and Epilepsy Cognitive ADHD, Autism, functions CP and Epilepsy Language Cognitivedevelopment functions Language development DNA methylation in newborn

Target groups Authorities New and updated knowledge to environmental- and health authorities will provide a basis for evaluating chemical regulations and regulatory advice for minimizing exposure for environmental toxicants during pregnancy Researchers and the scientific community New knowledge about mechanisms involved in how environmental contaminants affect brain development New knowledge about etiology of neurological diseases and neurodevelopmental disorders Suggest directions for future research Public New knowledge about preventable risk factors
