Oleh itu, adalah amat penting untuk membasmi pembiakan mikrob pada fomit bagi mengurangkan risiko penyebaran penyakit berjangkit secara meluas. Kajian ini dijalankan untuk menguji aktiviti antibakteria asid piroligneus Rhizophora apiculata (PARa), kesitotoksikannya serta menyaring potensi PARa sebagai disinfektan. Aktiviti antibakteria PARa diuji dengan menentukan kepekatan perencatan minimum (MIC), kepekatan bakterisidal minimum (MBC) serta asai waktu-membunuh. Kesitotoksikan PARa diuji terhadap sel Vero. Manakala potensi PARa sebagai disinfektan ditentukan melalui pembasmian pertumbuhan kulat pada dinding. Nilai MIC dan MBC PARa yang ditentukan adalah 1.875% sehingga 3.75%. Asai waktumembunuh menunjukkan PARa berjaya menurunkan jumlah inokulum S. aureus dan S. typhimurium sebanyak 99.9% selepas 12 jam. Ujian sitotoksisiti mendapati PARa adalah tidak toksik kepada sel dengan nilai CC %. Kepekatan PARa 6% juga berjaya mengurangkan pertumbuhan semula kulat pada dinding yang dirawat sepertimana natrium hipoklorit (NaOCl) 5%. Kesimpulannya, PARa memiliki aktiviti antibakteria bersifat bakterisidal, selamat terhadap sel dan berpotensi sebagai bahan disinfektan. Kata kunci: Fomit; asid piroligneus Rhizophora apiculata; aktiviti antibakteria; sitotoksisiti; disinfektan Abstract: Diseases can be transmitted by fomites, i.e. inanimate objects that exist around us and can be reservoir to various microorganisms. Thus, it is very important to reduce the risk of infectious diseases widespread transmission by eradicating microbial presence on fomites. Therefore, this research was conducted to study the antibacterial activity of Rhizophora apiculata pyroligneous acid (PARa), its cytotoxicity and potential as disinfectant. The antibacterial activity of PARa was studied by determining the minimum inhibitory concentration (MIC), minimum bactericidal concentration (MBC) and time-kill assay. The cytotoxicity of PARa was tested against Vero cells and eradication of fungi on walls was performed to screen its potential as disinfectant. The results revealed the MIC and MBC values of PARa were only 1.875% to 3.75%. From time-kill assay, PARa succeeded in lowering 99.9% of S. aureus and S. typhimurium inoculums after 12 hours. The cytotoxicity test shown that PARa obtained CC 50 value of 1.889%. The treatment of PARa 6% reduced the regrowth of fungi on walls similar to sodium hypochlorite (NaOCl) 5%. In conclusion, PARa exhibit bactericidal activity, not toxic towards cell and has potentials to be used as disinfectant. 2 1. Pengenalan Penyebaran mikrob boleh berlaku antaranya melalui air, udara, tanah, dan fomit. Fomit merupakan objek bukan hidup atau permukaan keras seperti tuala, dinding, dan mangkuk tandas yang boleh menampung pelbagai jenis mikroorganisma. Menurut Lopez et al. (2014), fomit membolehkan penyebaran patogen dalam persekitaran berlaku secara terus melalui sentuhan pada permukaan fomit dengan mulut atau kulit yang luka, ataupun secara tidak terus iaitu melalui kontaminasi pada jari yang boleh menyebabkan sentuhan tangan-ke-mulut, tangan-ke-mata atau tangan-ke-hidung berlaku. Pendedahan mikrob melalui fomit juga boleh menyebabkan pencemaran silang kepada persekitaran lain, seperti patogen nosokomial yang boleh dipindahkan ke dalam rumah melalui beg tangan yang sudah terdedah kepada persekitaran di dalam hospital. Lazimnya, disinfektan digunakan bagi merencat pertumbuhan mikrob pada fomit. Penggunaan disinfektan haruslah selamat apabila terdedah kepada pengguna atau alam sekitar, juga tidak menyebabkan kerintangan mikrob terhadap bahan-bahan aktif dalam disinfektan tersebut. Tambahan pula, disinfektan tersebut haruslah mudah diperolehi, mesra pengguna serta menggunakan kos yang efisien (CDC, 2008). Oleh itu, disinfektan yang diperolehi daripada sumber semula jadi amat penting sebagai alternatif kepada disinfektan yang dihasilkan secara sintetik. Mikrob yang bersifat patogen nosokomial didapati boleh hidup di atas permukaan fomit dalam pelbagai keadaan dan jangkamasa (Kramer et al. 2006). Oleh itu, penggunaan disinfektan bagi membersihkan permukaan fomit adalah penting bagi mengurangkan pertumbuhan dan mengawal penyebaran mikrob. Asid piroligneus (PA) atau cuka kayu, merupakan fraksi cecair akuas yang diperolehi daripada kondensasi wap proses pirolisis, iaitu proses termokimia yang menyebabkan penguraian komponen biojisim tumbuhan iaitu selulosa, hemiselulosa dan lignin berlaku tanpa kehadiran oksigen. PA yang dihasilkan melalui pirolisis perlahan biojisim tumbuhan adalah berwarna kuning-coklat atau coklat gelap dengan ph yang rendah serta terdiri daripada pelbagai komponen, antaranya ialah asid asetik, fenol dan alkohol (Mathew & Zakaria 2015). Terdapat pelbagai kegunaan PA hasil daripada pirolisis pelbagai jenis sumber seperti kulit kelapa, kayu buluh dan herba. Di Malaysia, sumber PA boleh diperolehi daripada penghasilan arang kayu menggunakan Rhizophora apiculata, atau bakau minyak. PA daripada R. apiculata didapati memiliki aktiviti antioksida (Loo 2008; Loo et al. 2007), aktiviti antibakteria (Darah et al. 2014) serta aktiviti antiyis terhadap Candida albicans (Darah et al. 2013). Asid piroligneus R. apiculata (PARa) dikatakan boleh merawat luka-luka kecil serta menghalau serangga perosak. Namun begitu, produk ini sering dilupuskan sebagai bahan buangan kerana dianggap tidak mempunyai kepentingan. Oleh hal yang demikian, kajian yang dijalankan memberi fokus kepada aktiviti antibakteria PARa, kesitotoksikannya dan potensi PARa untuk dijadikan bahan disinfektan daripada sumber semula jadi dan memenuhi ciri-ciri disinfektan yang diinginkan. 2. Bahan dan Kaedah 2.1 Asid piroligneus Rhizophora apiculata Sampel asid piroligneus R. apiculata (PARa) diperolehi dari daerah Kuala Sepetang, Perak, Malaysia. Sampel dituras melalui membran selulosa dengan liang bersaiz 0.22 µm untuk mendapatkan cecair PARa yang steril. Pencairan ganda dua PARa dilakukan dengan menggunakan kaldu Mueller-Hinton (MHB, Oxoid, UK) dan ph bagi setiap pencairan diukur serta direkodkan. 2.2 Penyediaan inokulum bakteria ujian Bakteria ujian yang digunakan terdiri daripada empat spesies bakteria Gram positif iaitu E. faecalis, S. aureus, B. subtilis serta MRSA dan empat spesies bakteria Gram negatif iaitu S. marcescens, Shigella spp., S. typhimurium, dan E. coli. Kesemua bakteria diperolehi dari Makmal Mikrobiologi Patogen, Pusat Pengajian Biosains dan Bioteknologi, Fakulti Sains dan Teknologi, Universiti Kebangsaan Malaysia. 143

3 Koloni tunggal bakteria diperolehi dengan kaedah piring coretan pada agar nutrien (NA, Oxoid, UK) dan dieramkan selama 24 jam pada suhu 37 C. Pencilan koloni tunggal yang terhasil diinokulasikan ke dalam MHB dan dieram dalam suhu 37 C dengan goncangan 150 psm selama 24 jam. Kepekatan inokulum ditetapkan kepada CFU/mL, berdasarkan piawaian kekeruhan 0.5 McFarland. 2.3 Penentuan kepekatan perencatan minimum (MIC) MIC dilakukan dengan menggunakan kaedah pencairan kaldu (Grare et al. 2007). Pencairan ganda dua PARa dilakukan dengan menggunakan MHB, bermula daripada kepekatan isipadu per isipadu 15% sehingga %. Sebanyak 100 µl daripada setiap pencairan PARa dan 50 µl inokulum piawai bakteria ditambah ke dalam piring mikrotiter 96 telaga. Piring dieram selama 24 jam pada suhu 37 C. Larutan 3-(4,5-dimetil-2-tiazolil)-2,5-difenil-2H-tetrazolium bromida (MTT, Calbiochem, USA) ditambah sebanyak 20 µl ke dalam setiap telaga setelah pengeraman dilakukan dan piring mikrotiter dieram lagi selama dua jam. Pencerapan pertukaran warna MTT daripada kuning kepada ungu menunjukkan terdapatnya pertumbuhan bakteria. Ujian dilakukan secara triplikat dan nilai MIC diambil pada kepekatan PARa yang paling minimum tanpa berlaku sebarang perubahan warna MTT. 2.4 Penentuan kepekatan bakterisid minimum (MBC) Sebanyak 20 µl campuran daripada telaga yang mempunyai nilai MIC, 2 MIC dan 4 MIC diambil, dipindahkan ke atas agar Mueller-Hinton (MHA, Oxoid, UK) dan dicoretkan menggunakan putik kapas steril. MHA kemudiannya dieram selama 24 jam pada suhu 37 C. Pertumbuhan bakteria kemudiannya dicerap. Nilai MBC ditentukan dengan kepekatan yang paling rendah dimana agen antibakteria membunuh lebih daripada 99.9% daripada inokulasi awal selepas 24 jam pengeraman (Aboaba et al. 2006). 2.5 Perbandingan aktiviti PARa dengan disinfektan Natrium hipoklorit 5.25 % (NaOCl, Clorox, Malaysia) digunakan dalam ujian MIC dan MBC bagi membandingkan aktiviti PARa dengan disinfektan yang lazim digunakan dalam pembersihan kawasan rumah dan peralatan di fasiliti-fasiliti kesihatan. Semua parameter yang diaplikasikan adalah sama dengan parameter ujian MIC dan MBC bagi PARa, kecuali kawalan positif yang digunakan dalam ujian ini adalah NaOCl dengan kepekatan 0.26%, yang termasuk dalam julat kepekatan NaOCl yang berkesan sebagai disinfektan terhadap bakteria iaitu 0.02% sehingga 0.5% (Mazzola et al. 2009), serta nilai MIC ditentukan melalui kekeruhan campuran kaldu dalam telaga yang menunjukkan terdapatnya pertumbuhan bakteria. 2.6 Asai waktu-membunuh (Time-kill assay) Ujian ini dilakukan mengikut kaedah CLSI (1999) dengan sedikit perubahan bagi menentukan keberkesanan masa tindakan PARa sebagai bahan antibakteria. Kultur bakteria S. aureus dan S. typhimurium ditetapkan kepada CFU/mL. Sebanyak 1 ml kultur bakteria dan 9 ml PARa dengan nilai MIC bagi kedua-dua bakteria tersebut dicampurkan ke dalam kelalang kon 100 ml dan dieram dalam penggoncang digital pada suhu 37 C dengan goncangan 150 psm. Pada selang masa setiap empat jam (0, 4, 8, 12, 16), pemiringan sebanyak 25 µl daripada campuran MIC PARa dan kawalan pertumbuhan dilakukan ke atas MHA dan dieram selama 24 jam pada suhu 37 C. Pengiraan koloni yang hidup serta penentuan CFU/mL dilakukan. Akhir sekali, graf log 10 CFU/mL melawan masa diplotkan. Kesan bakterisidal boleh dilihat dengan terdapatnya penurunan jumlah koloni kepada 3 log 10, atau sebanyak 99.9% daripada jumlah asal pada masa yang ditetapkan. 144

4 2.7 Penentuan kesitotoksikan dan analisa CC 50 Sel Vero, iaitu sel epitelium ginjal monyet hijau Afrika, Cercopitheus aethiops, diperolehi daripada penyimpanan stok kultur Makmal Virologi, Pusat Pengajian Biosains dan Bioteknologi, Fakulti Sains dan Teknologi, UKM. Pemiringan sebanyak 100 µl ampaian sel Vero ditambahkan ke dalam piring mikrotiter 96 telaga dan dieram pada suhu 37 C dengan kehadiran CO 2 selama 48 jam bagi membenarkan pembentukan sel ekalapis berlaku. Kemudian, medium pertumbuhan yang lama digantikan dengan 100 µl PARa yang mempunyai kepekatan isipadu per isipadu 100% sehingga %. Piring mikrotiter kemudiannya dieram pada suhu 37 C dengan kehadiran CO 2 selama 48 jam bagi membenarkan PARa bertindak terhadap sel Vero. Pewarnaan sel dilakukan dengan menggunakan asai MTT (Silva et al. 2006). Setelah pengeraman 48 jam tamat, semua medium dan PARa dikeluarkan dan diganti dengan 100 µl MTT. Piring dieram selama dua jam pada suhu 37 C dengan kehadiran CO 2. Kemudian, MTT digantikan dengan 100 µl DMSO yang ditambah ke dalam setiap telaga bagi melarutkan kristal formazan. Piring mikrotiter digoncang dengan perlahan selama lima minit. Nilai serapan optik (OD) kemudiannya dibaca menggunakan spektrofotometer pada jarak gelombang 540 nm. Ketoksikan PARa diukur dengan nilai CC 50 iaitu kepekatan PA yang boleh menyebabkan gangguan kepada kekuatan integriti membran sebanyak 50% daripada populasi sel kultur. Peratus kemandirian sel boleh dikira dari nilai penjerapan yang diperolehi menggunakan formula berikut: Kemandirian Sel (%) = (OD ujian - OD pengosong / OD sel OD pengosong) x 100 Graf peratus kemandirian sel melawan kepekatan PARa diplotkan dan analisis regresi tidak linear dilakukan menggunakan perisian Graphpad Prism 7 versi Demo bagi mendapatkan nilai CC 50 PARa. 2.8 Pembasmian kulat pada dinding menggunakan asid piroligneus Rhizophora apiculata Kaedah ini dilakukan dengan menyediakan kuadrat (20 cm 20 cm) pada dinding yang berkulat berhampiran Bilik 3139, Aras 3, Bangunan Biologi, FST, UKM. Kajian keberkesanan PARa pada kepekatan 2%, 4% dan 6% bagi merencat pertumbuhan semula kulat pada dinding tersebut dilakukan dengan menggunakan NaOCl 5% sebagai kawalan positif dan air paip sebagai kawalan negatif. PARa dan NaOCl dicairkan menggunakan air paip bagi memimik kaedah pembasmian dalam situasi sebenar. Kuadrat pada dinding yang terlibat dibersihkan sebanyak dua kali dengan menyemburkan bahan pembasmi yang terlibat dan dilap menggunakan tisu steril. Pertumbuhan semula kulat diperhatikan selepas sebulan dengan menyebarkan putik kapas steril yang telah disapu pada dinding kepada agar dekstros kentang (PDA, Oxoid, UK). Piring PDA kemudiannya dieram pada suhu 30 C selama 5 hari. 3. Hasil dan Perbincangan 3.1 Pengukuran ph asid piroligneus Rhizophora apiculata PARa lazimnya ditemui dalam bentuk cecair dan mempunyai ph yang rendah. Manakala, PARa yang dipekatkan menggunakan rotavapor sehingga mencapai isipadu 1:10 daripada isipadu asal serta bebas daripada air dan komponen meruap mempunyai ph 1.8 (Darah et. al. 2014). Dalam kajian ini, ph PARa yang telah dicairkan diukur dan hasilnya direkodkan dalam Jadual 1. Kepekatan PARa yang paling sedikit masih mempunyai sifat berasid namun lemah dengan nilai ph menghampiri ph 7. Oleh itu, sifat PARa yang berasid boleh menyumbang kepada aktiviti antibakteria kerana kebanyakan bakteria bermandiri pada ph neutral. 145

Nilai MIC ditentukan oleh perubahan warna MTT, dimana ia merupakan telaga terakhir yang kekal berwarna kuning selepas pengeraman piring mikrotiter dilakukan. Telaga yang mengalami perubahan warna MTT kepada warna biru-keunguan membuktikan bahawa terdapat kehadiran bakteria kerana wujudnya aktiviti pemotongan garam tetrazolium pada MTT kepada formazan yang berwarna biru dan tak larut dalam air oleh mitokondria yang aktif (Grare et al. 2007). Nilai MIC dan MBC serta kepekatan PARa yang mempunyai sifat bakterisidal ditunjukkan dalam Jadual 2. Kesemua nilai MIC PARa terhadap bakteria sama ada Gram positif atau Gram negatif adalah sama iaitu 1.875%, kecuali bagi MRSA dan S. typhimurium iaitu 3.75%. Seperti yang dijangka, kepekatan PARa yang lebih tinggi diperlukan bagi merencat MRSA berbanding S. aureus kerana ia sifatnya rintang metisilin. Namun, nilai MIC PARa yang diperoleh dalam kajian ini adalah tinggi berbanding nilai MIC PARa dalam kajian Darah et al. (2014), iaitu 6.25 mg/ml atau bersamaan dengan 0.625%. Walaupun terdapat perbezaan nilai kepekatan, tetapi pola perencatan adalah sama dimana kepekatan PARa yang lebih tinggi diperlukan untuk merencat pertumbuhan MRSA berbanding bakteria lain. Berdasarkan Jadual 2, kepekatan PARa sebagai MBC dapat ditentukan setelah berjaya menurunkan lebih daripada 99.9% jumlah inokulum daripada inokulasi awal selepas 24 jam pengeraman seperti yang dinyatakan oleh Aboaba et al. (2006). Dengan menggunakan maklumat nilai MIC dan MBC, maka aktiviti PARa sama ada ia bersifat bakteriostatik atau bakterisidal boleh ditentukan melalui nisbah MBC kepada MIC (MBC:MIC), dimana nilai nisbah yang bersamaan dengan 1 atau 2 menunjukkan bahawa kepekatan PARa yang digunakan adalah bersifat bakterisidal manakala nilai nisbah yang bersamaan dengan 4 atau 16 menunjukkan bahawa kepekatan PARa yang digunakan hanya mempunyai aktiviti bakteriostatik (Konaté et al. 2013). Hasilnya, kepekatan PARa yang diperolehi dalam Jadual 2 menunjukkan PARa mempunyai aktiviti bakterisidal terhadap kesemua bakteria ujian yang digunakan. Nisbah MBC:MIC yang diperolehi bagi bakteria B. subtilis, S. aureus, MRSA dan S. typhimurium, adalah 1 yang menunjukkan bahawa kepekatan MIC terhadap bakteria-bakteria ini bukan sahaja boleh merencat pertumbuhan, malah boleh membunuh bakteriabakteria tersebut. Bagi E. faecalis, E. coli, Shigella spp. dan S. marcescens pula, nilai MBC yang diperolehi iaitu 3.75% adalah dua kali ganda lebih tinggi daripada nilai MIC yang ditentukan iaitu 1.875%. Nisbah MBC:MIC bagi bakteria-bakteria ini adalah 2, dimana ia menunjukkan bahawa PARa bersifat bakterisidal namun kepekatannya perlu dinaikkan sebanyak dua kali ganda daripada nilai kepekatan yang mempunyai aktiviti perencatan bagi membunuh bakteria yang telah dinyatakan. 6 Aktiviti antibakteria PARa dibandingkan dengan NaOCl yang merupakan disinfektan lazim digunakan dalam pembersihan kawasan rumah serta peralatan di fasiliti-fasiliti kesihatan (CDC, 2008). Menurut Mazzola et al. (2009), NaOCl digunakan sebagai disinfektan asas dan mempunyai spektrum aktiviti antimikrob yang luas. Dalam kajian ini, NaOCl 5.25% digunakan dalam ujian MIC dan MBC dan hasilnya direkodkan dalam Jadual 3. Nilai MIC NaOCl adalah lebih rendah berbanding PARa dan semua nilai MBC yang diperolehi menunjukkan NaOCl memiliki sifat bakterisidal. Oleh itu, aktiviti antibakteria NaOCl adalah lebih berkesan berbanding PARa. Jadual 3. Hasil daripada asai ini jelas menunjukkan keberkesanan PARa sebagai bahan antibakteria yang bersifat bakterisidal. A B Rajah 1. Manakala dengan menggunakan perisian Graphpad Prism, nilai CC 50 PARa telah berjaya diperolehi iaitu 1.889%. Menurut Zihiri et al. (2005), sebatian yang mempunyai nilai CC 50 melebihi 0.02 mg/ml, atau 0.002% adalah tidak toksik. Oleh itu, nilai CC 50 PARa yang diperolehi menunjukkan bahawa ia adalah tidak toksik dan selamat untuk digunakan. 147

Dengan nilai CC 50 dan nilai MIC kepekatan PARa yang telah berjaya diperolehi, maka indeks selektif (SI) bagi sesuatu agen antibakteria, yang mana dalam hal ini ialah PARa, boleh ditentukan dengan menggunakan rumus SI = CC 50 / MIC. SI digunakan untuk menentukan sama ada bahan yang dikaji sesuai dijadikan sebagai agen terapeutik atau sebaliknya (Fontanay et al. 2008). Sebatian yang mempunyai nilai SI melebihi 10 adalah tidak bertoksik dan selamat digunakan sebagai agen terapeutik. Berdasarkan Jadual 4, ia menunjukkan bahawa PARa hanya sesuai dijadikan sebagai bahan antibakteria serta mempunyai nilai CC 50 yang tidak toksik kepada sel. Namun, ianya tidak sesuai dijadikan sebagai agen terapeutik terhadap manusia atau haiwan kerana kesemua nilai SI yang diperolehi hanya daripada 0.5 sehingga 1 sahaja. Jadual 4. Secara kasarnya, bahagian dinding yang terlibat masih kelihatan bersih dari kali pertama pembersihan kuadrat dijalankan. Namun, selepas pengeraman PDA dilakukan, maka terdapat pertumbuhan pelbagai spesies kulat seperti yang ditunjukkan pada Rajah 3. PARa pada kepekatan 6% mengalami pertumbuhan kulat yang lebih sedikit berbanding NaOCl 5%. Ini menunjukkan bahawa kepekatan PARa yang lebih tinggi diperlukan bagi membasmi pertumbuhan semula kulat berbanding kepekatan yang disyorkan oleh syarikat pengeluar NaOCl yang terlibat iaitu 5% bagi membersihkan kawasan dinding dan lantai dalam rumah. 148

Kesimpulan Asid piroligneus Rhizophora apiculata (PARa) mempunyai aktiviti antibakteria yang bersifat bakterisidal serta tidak toksik kepada sel dan berpotensi untuk dijadikan bahan disinfektan yang mesra pengguna. PARa hanya memerlukan nilai kepekatan yang rendah bagi merencat aktiviti dan membunuh bakteria. Kepekatan PARa yang tidak toksik dan selamat kepada sel juga telah berjaya ditentukan namun didapati PARa tidak sesuai dijadikan sebagai agen terapeutik terhadap haiwan dan manusia. Simulasi kepada situasi pembasmian kulat pada permukaan dinding juga menunjukkan kepekatan PARa yang lebih tinggi diperlukan berbanding kepekatan disinfektan yang disyorkan bagi kegunaan dalam kawasan rumah.
