What I do: Computational Biology Computational biology uses mathematical and computational approaches to address theoretical and experimental questions in biology. Raw material (data):. DNA/RNA/protein sequences (text) and their biological annotation. Molecule abundances (numbers) Techniques:. Text manipulation. Sequence alignment algorithms. Basic statistics. Classification and clustering methods

The Central Dogma

Not that simple Orphanides 2002 Cell

mrna splicing

Alternative splicing

Alternative splicing Liver cancer 95%+ of genes undergo alternative splicing!

How is alternative splicing regulated? Interplay between protein regulatory factors and RNA sequence signals How has alternative splicing evolved? Intron-exon structures highly conserved across animals but most alternative splicing events are species-specific

The Project Survey of tissue-specific alternative splicing in vertebrates by high-throughput sequencing: finding the elements of an evolutionary splicing predictor Sequence and measure abundance mrna in several tissues (brain, liver, kidney, heart, muscle) from several vertebrate species (human, mouse, chicken, frog, lizard, fish) Model data and make predictions to be tested in the laboratory If successful, important medical applications: understanding of splicing associated disease; design of gene therapies

Why North America? In the early stages of a scientific career, not advisable to stay too long in the same place and always work with the same people (7 years in Cambridge 2 shifts ) Try different culture and lifestyle (never lived out of Europe) Try different work culture (North-Americans more pragmatic and competitive) Most of top universities in the world Resources for cutting-edge research

The 4 options

Why Toronto? Gut feeling Scientific quality of lab s recent work Congeniality with advisor (Prof. Ben Blencowe) Friendly lab environment Strongest interest in hiring me (incl. better salary) Project best suiting my scientific interests Toronto s lifestyle Bias towards welfare states Geography (closer to Europe + West Coast easy to reach)

Why the Marie Curie IOF? Prestigious and competitive award Generous fellowship (~210k over 3 years) Objective rules and evaluation process Independence (own budget for conferences, hardware, books, etc) Re-entry into Portugal/Europe Easier access to future funding Important network of scientists Excellent practice for grant writing

The Application Process

The Evaluation Process

Working in Canada so far + Congeniality with boss remains + Easy, friendly, and professional atmosphere in the lab remains + Many exciting collaborations with colleagues from very diverse scientific backgrounds + All the resources needed to succeed + More focused and objective approach to science - Little socializing at work or interest in extra work academic activities (reflected also on the infrastructure)

After Canada Jun 2013 May 2014 (return phase): Institute of Molecular Medicine, Lisbon, Portugal Jun :???
