REMEDIATE Linking bioavailability and toxicity changes of complex chemicals mixture to support decision making for remediation endpoint of contaminated soils Cipullo S., Negrin I., Claveau L., Snapir B., Tardif, S., Pulleyblank C., Prpich G., Campo P., Coulon F. 20 th September 2018, Remediation and Land Management: Research Meets Practice Session D1 Site Modelling, Risk Assessment, and Risk Communication II

Experimental Design Soil 1 Soil 2 PHC 9000 mg kg -1 HM 450 mg kg -1 PHC 2000 mg kg -1 HM 500 mg kg -1 Un-treated Un-treated 15% w/w compost 15% w/w compost Former gaswork sites 5% w/w biochar 5% w/w biochar Sampling time 0, 30, 90, and 180 days

Methodological approach CHEMISTRY ORGANICS: Total extractable : Dichloromethane :Hexane (1:1) Bioavailable: β-cyclodextrin Readily available: Methanol INORGANICS: Gas Chromatography Mass Spectrometry Total extractable (GC-MS) : Aqua regia Bioavailable: Sequential extraction BIOLOGICAL INDICATORS Bacteria count Soil respiration Microbial community fingerprint (PLFAs) TOXICITY BIOASSAYS Seeds germination (4 days) Earthworms survival (14 days) Bioluminescent bacteria Providing an integrated approach combining chemical, biological and toxicological analyses

Behaviour and distribution changes overtime Total petroleum hydrocarbons Bioavailable hydrocarbons TPH decrease 46% (Soil 1 + Compost) and 30 % (Soil 2 + Compost) Bioavailable content decreased by 92 % and 59 % (EC aromatic) and by 27 % and 62 % (EC aliphatic) in Soil 1+ Compost and Soil 2 + Compost respectively

HM and metalloids solid phase distribution Carbonates Pore water Oxides Elements (concentration) Extractions Illustrative heatmap and associated clustergram for CISED extraction data for the Soil 1 (treated with compost, biochar, or un-amended) HM and metalloids were almost entirely found in the non-exchangeable fraction No major changes in their distribution were observed with incubation time

Bioassays (I) Biological indicators For both soils : Incubation time and treatment (p 0.001) significantly influenced the PLFA composition Culturable bacteria were significantly higher in compost amended samples

Bioassays (II) Ecotoxicological assays EARTHWORMS LETHALITY SEEDS GERMINATION MICROTOX BIOASSAY Soil 1 + Compost Condition index for increased across sampling time Soil 2 No significant effect (p > 0.05) of treatment and time on Condition index Soil 1 +40% germination at 90 days for mustard and rye grass Soil 1 and Soil 2 Compost amended 90%germination across all seeds types Soil 1 + Compost 14 times less toxic after 180days Soil 2+ Biochar slightly less toxic after 180days Different bioassays often yields contradictory results (sensitivity) (1) Respiration, (2) E. fetida CI at 14 days, and (3) Seeds germination were the best at discriminating between the amended and un-amended soils (in particular for Soil 1). These assay are expected to be more reliable to be used in the risk assessment

Correlation between bioavailability/bioassays Soil 1 + Compost Soil 2 + Compost Linear correlation (Spearman) bioavailable concentrations and toxicological responses in multiple bioassays. Positive correlations were displayed in blue and negative correlations in red colour, were intensity were proportional to the correlation coefficients (scale). Significant relationship (p < 0.05) between the bioavailable/readily available fractions of the chemical mixtures and the ecotoxicological bioassays When bioavailable/readily available concentrations decrease, the toxicity also decreases

IMPACT Take home message 15% w/w compost Both biotic and abiotic degradations occurred; Degradation in Soil 1 > Soil 2 Bioavailable fractions significantly decreased over time in particular for EC aromatic Bioassays were highly correlated with the decrease of the bioavailable hydrocarbons Improve risk evaluation Provide multiple line of evidence bioavailable fraction rather than total concentration linking bioavailability and toxicity to end point remediation Refining guideline values Avoiding unnecessary remediation Reducing remediation costs Delivering a sustainable approach

REMEDIATE Thanks For Listening This project has received funding from the European Union s Horizon 2020 research and innovation programme under the Marie Skłodowska-Curie grant agreement No Project coordinated by the QUESTOR Centre at Queen s University Belfast
