PROTECTION Against the losses Protecting or Controlling? CONTROL + PREVENTION of challenge risk (Shedding) VACCINATION + BIOSECURITY BIOSECURITY VACCINATION

Growing (drop of MDA) Infection Delivery Shedding Depletion Downtime Cleaning / Disinfection

Three types of live Gumboro vaccines Conventional Live - Intermediate - Intermediate Plus Immune-Complex IBD Recombinant (rhvt-vp2)

Live IBD vaccines and Protection All vaccines can protect against the negatives consequences of the disease (clinical protection) Protecting the bursa against the infection is the lastly characteristic that the vaccines should accomplish, but there are remarkable differences in between them.

Objectives of a live Gumboro vaccine STOP the shedding of the field virus PREVENT the challenge risk cycle after cycle BLOCK bursa s infection from any type of IBDV strains STOP The Gumboro Cycle Controlling the Gumboro Disease No buildup of existing field viruses No selection of new strains

Blocking the infection of the bursa can be proved under field conditions Laboratory investigation report Date of submission: November 11 th, 2014 Country: China Samples: Chicken bursas prints on FTA card for PCR (pool of 5 samples per age) Sample code Origin Age RESULT D2851/1 14 Negative D2851/2 China 17 Positive / W2512 D2851/3 Transmune : 1 do 21 Positive / W2512 D2851/4 23 Positive / W2512 *Samples coming from flocks with low MDA (<2000 Idexx kit)

Blocking the infection will stop the challenge virus replication in the bursa SPF chickens, vaccinated at 21 days with W2512 IBDV strain and challenged 2, 3 and 4 days later with vvibdv At Challenge 4 days post-challenge Challenge Histopathology changes (vaccine virus) Histopathology changes (challenge virus) Molecular detection (challenge virus) Molecular detection (vaccine virus) 2 days post vaccination 3 days post vaccination 4 days post vaccination 100% 0% 0% 100% 100% 0% 0% 100% 100% 0% 0% 100% V Palya, T Mato, T Tatar-Kis, Y Gardin, Dissertation at AAAP Meeting, Seatle-USA. 2009

Blocking the infection will stop the challenge virus replication in the bursa Percentage of properly vaccinated flocks after vaccination either with or one Conventional IBD Plus vaccine Live intermediate plus vaccines have the potential to block the infection, but suffered from the lack of vaccination coverage, characteristic of the drinking water technique Bursa samples from 40 flocks. Histopathology and PCR at 3, 4 and 5 weeks of age. Only vaccines that quickly replicate in the bursa can block the infection If they are applied in the Hatchery!

IBD Titer Field monitoring Serology (Spain) Field Vaccination Consistent and homogeneous results after the continuous usage of Transmune

Blocking the infection will stop the challenge virus replication in the bursa. Can rhvt-vp2 vaccines do it? Bursa are partially protected after vvibd challenge at 21 days in chickens vaccinated with a rhvt-vp2 vaccine. the on - off protection in the bursa RUSSIA, flocks vaccinated with a rhvt-vp2 vaccine MALAYSIA, flocks vaccinated with a rhvt-vp2 vaccine Z.Penzes, MDV Symposium, Townsville, Australia July-2008 CHINA, flocks vaccinated with a rhvt-vp2 vaccine

Broad spectrum of protection is necessary to avoid the likelihood for the emergence of new strains Commercial broilers vaccinated at day 1 with or a rhvt-vp2 vaccine and challenged at 28 days of age with a vvibdv strain from Turkey (TUR) or different Subclinical IBDV strains from USA (DelE, AVS-EL), Brazil (BR), Mexico (MX) or South Africa (ZA). Only vaccines applied in the hatchery that can replicate in the bursa can block the infection and stop the shedding of any field Gumboro Virus
