No. R6872) is a sandwich enzyme immunoassay for the quantitative determination of cashew or parts of cashew in food like cereals, pastries, ice cream and chocolate. The test has a detection limit of 0.19 mg/kg cashew and a limit of quantification of 2,5 mg/kg cashew. Please also note our application note More sensitive analysis with RIDASCREEN FAST Peanut (Art. No. R6202) or RIDASCREEN FAST Cashew (Art. No. R6872). In this application note the procedure for a more sensitive quantification (> mg/kg) is specified for these both products. Therefore, a more sensitive analysis for cashew is possible. At present RIDASCREEN FAST Cashew is the most sensitive ELISA for the detection of cashew on the market. 2 R-Biopharm AG New products RIDASCREEN Streptomycin Streptomycin is one of the most frequently used antibiotics in veterinary medicine, predominantly in dairy animals. Application errors (such as noncompliance with the prescribed withdrawal period after application) can result in residues of the drug in foods of animal origin. The EU has introduced maximum residue limits (MRL) to protect consumers against potential health risks caused by inhibitor residues. These are 200 µg/l in milk and 500 µg/kg in beef, pork, and rabbit meat or livers. In addition, streptomycin is also used to fight blight in pomaceous fruit trees (caused by Erwinia amylovora) and can cause residues, e.g. in honey. The newly developed RIDASCREEN Streptomycin (Art. No. R3104) test replaces the previous test (Art. No. R3103) and provides some improvements compared to it: Sample testing is clearly faster and easier due to incubation periods reduced to 45 minutes and supply of all reagents needed for performing the tests as ready-for-use solutions. No. R3104 The wide range of analyzable matrices (milk and milk powder, honey, meat, liver, kidney, shrimp, and apple juice) and the improved sensitivity of the test (also suitable for dihydrostreptomycin) allow safe testing of a large variety of samples. You have the option to test both RIDASCREEN Streptomycin tests in comparison. This can be done until the current stock of the old RIDASCREEN Streptomycin test (Art. No. R3103) is sold. Yeasts and molds in the Compact Dry portfolio: Compact Dry YMR to detect yeast and mold in hours! As of fall 2016, the new rapid detection test for yeast and mold: Compact Dry YMR (detection in 48 to 72 hours) is available from R-Biopharm! Like the previous Compact Dry plates, this parameter is available in two package sizes: 100 tests (Art. No. HS9801) and 40 tests (Art. No. HS9802). This easy to use chromogenic culture medium plate in a dry format, which can not only be used for food analyses but also for beverages and for surface sampling (in combination with wet swabs) and is based on the same color detection system as the Compact Dry YM (blue/blue-green colonies). Results can be read off after just 48 hours, which saves up to 5 days on the incubation period! The new Compact Dry YMR lets a food manufacturer determine the total status of the extremely critical parameter of microbial fungi in the food considerably faster. Particularly, foods with a short shelf life can be released much faster, or countermeasures can be initiated in time before the production process ends. Official validations compared to the respective standard detection methods (according to AOAC-RI, MicroVal, and NordVal) are being carried out. page 2

3 Our products RIDASCREEN Gliadin AOAC Official Methods of Analysis from First Action to Final Action The Expert Review Panel (ERP) has recommended the AOAC Official Method (corresponding to the ELISA RIDASCREEN Gliadin, Art. No. R7001) for Final Action during the AOAC Annual Meeting in Dallas (Texas) in September. The Official Methods Board will finally vote in December 2016 on Final Action. The supporting documentation submitted to AOAC showed the excellent precision, robustness and accuracy of our R5 Mendez method for gluten analysis. RIDASCREEN Gliadin, Art. No. R7001 RIDA QUICK Soya no cross reactivities The RIDA QUICK Soya (Art. No. R7103) dip stick can be used for swabbing and for food analysis. Approximately 80 compounds were tested and no cross reactivity was observed. Food samples are extracted with the environmental friendly RIDA QUICK Soya Extraction buffer (Art. No. R7113). The detection limits are as follows: Soya flour in wheat flour 0.5 mg/kg soya protein Processed food approx. 10 mg/kg soya protein (depending on the matrix) Surfaces approx. 0.5 µg soya protein/100 cm 2 The antibodies utilized for dip stick RIDA QUICK Soya and for the ELISA RIDASCREEN FAST Soya (Art. No. R7102) detect especially highly processed soy proteins. In food samples the soy proteins are present in their native form or they may be still denatured. To ensure a complete detection of soy proteins by the antibody all samples have to be cooked at 100 C to convert potentially present native proteins into their denatured form. 4 New Application for histamine in wine RIDASCREEN Histamine (enzymatic) (Art. No. R1605) is an enzymatic test in microtiter plate format for the quantitative determination of histamine in fresh fish, canned fish, fish meal, wine (with RIDA Sample Decolorant) and cheese. The application note describes how the measurement range for wine analysis can be lowered to mg/l histamine. The limit of detection of an extracted sample is 0.05 mg/l histamine and the limit of quantification is 0.3 mg/l histamine. The molecular biological real-time PCR method, based on specific DNA amplification, detects highly specific DNA and DNA fragments of the relevant species. The sensitivity is based on the detection system, the food matrix and processing grade. A relative quantification is possible by determining Ct values of the real-time PCR run. However, systems are calibrated on raw muscle tissue. The German reference office for food proficiency testing and reference materials (DRRR) is offering a proficiency test end of 2017, in which stable gelatin is used as a matrix for qualitative detection of low amounts of present porcine (sus scrofa) and bovine (bos taurus) DNA. 5 News from our cooperation partner GEN-IAL GmbH Fast analysis of beverage spoiling bacteria, particularly beer spoiling bacteria in the brewing industry, is meeting with increased interest. On the one hand, globalization and international shipping and storage of well-known brand name beers are becoming more and more important. On the other hand, the crafts brewer movement, which is going global as well, brew a large number of new beer styles. They use methods like cold hopping or no filtration, which are at a higher risk of introducing undesirable bacteria and yeasts. Brewers increasingly rely on fast real time PCR to ensure brand reputation and reduce quarantine periods. GEN-IAL is offering two new kits in addition to its kits of the First-Beer series that have proven useful for years and the new QuickGEN kits: GEN-IAL QuickGEN P1 Screening TaqMan (Art. No. QPP1HR 0050): This test is suited for routine testing of beer samples or for in-process controls. It detects Lactobacillus spp. and Pediococcus spp., bacteria that are harmful to beer, as well as the anaerobic bacteria Megasphaera and Pectinatus. As it also detects the hora and horc hop resistance genes, the risk that there are bacteria that can harm the beer can then be estimated. GEN-IAL QuickGEN First-Biofilm TaqMan (Art. No. QTPBF 0050): This test is suited for the specific DNA detection of the early biofilm forming bacteria Lactococcus lactis, Leuconostoc mesenteroides and yeast Pichia anomala. The formation of biofilms can have severe consequences because they may not be removed in the typical routine CIP cleaning process. Early detection in suspect cases therefore is of great interest. Both test kits are multiplex kits with internal amplification control. These kits are designed for 50 reactions and can be used in all common thermocyclers with the FAM, VIC/HEX, ROX and Cy5 detection channels. The QuickGEN DNA preparation kits CSE 0100 or FSE 0100 can be used for sample preparation. Therefore, the analysis can be performed at the user s discretion from the preconcentration, from direct centrifugation, or from a filtration sample. Although the illegal use of banned substances such as antibiotics in seafood products has been well-known for some years, the problem has not completely disappeared. The EU Rapid Alert System for Food and Feed (RASFF) shows some 19 alerts so far in 2016, with drugs such as nitrofurans and chloramphenicol continuing to be detected in imported products. Fortunately, screening for residues can easily be accomplished for specific target drugs using the range of RIDASCREEN ELISA kits available from R-Biopharm. Of particular interest is a recent publication in Journal of Food Protection by Edward Jester and co-workers from the US Food & Drug Administration, who have conducted a performance assessment of RIDASCREEN Chloramphenicol (Art. No. R1505) for detecting residues in crab and shrimp. At levels from 0.1 to 0.6 ppb for chloramphenicol in spiked shrimp and crab meat, recoveries by ELISA were quantitative and there was close agreement with spiking levels and with an LC-MS/MS confirmatory method. These authors concluded that their findings support the use of this ELISA kit for screening purposes and, when used in conjunction with validated instrumental methods, for regulatory monitoring of chloramphenicol in these species. For validation purposes R-Biopharm also supply an EASI-EXTRACT CHLORAMPHENICOL immunoaffinity column (Art. No. RBRP300/P300B) which have been shown in a 2013 JAOAC International publication to be as an effective clean-up technique for analysis of chloramphenicol. The detection limit was reported to be 0.05 ppb for prawns and 0.16 ppb for honey, both well below the minimum required method performance limit. The accuracy of the immunoaffinity column and LC-MS/MS method was demonstrated by participation in proficiency testing and the method was shown to be applicable to a wide range of matrices, including milk, egg, Royal Jelly, meat, and seafood products. With ELISA kits being commercially available for screening for many drug residues, and immunoaffinity columns available for sample clean-up, food control authorities are well placed to monitor imports into the EU. Consumers can therefore have confidence that efficient and effective testing methods are available for analysis of these products for illegal use of drugs being detectable at very low levels. page 6

7 New product: EASI-EXTRACT MULTI-VIT B (LGE) B vitamins are a class of water soluble vitamins which play important roles in cell metabolism. These vitamins must be obtained from the diet as the body excretes excess vitamins and does not store them within the body, except vitamin B12. As a result many food products are fortified with vitamins to ensure infants and the elderly are consuming the recommended daily allowance of the required vitamin. Consumers are becoming more aware of what they eat and food labelling plays an important part in informing customers, enabling them to make informed choices. Foods may contain natural levels of vitamins however in many cases certain foods like infant formula, baby food and milks are fortified with vitamins such as biotin, folic acid and vitamin B12. Fortification is an expensive process and manufacturers are required to check that the food has been fortified with the correct level of vitamin and to ensure that it is still present at the end of the product shelf life. Traditional microbiological methods can be very tedious, time consuming and lack specificity. These methods are no longer capable of meeting the new challenges requiring low-level determination of vitamins with a high specificity. In addition, current trends in analysis are taking a multi-analyte approach, which is consistent with the strategy being implemented by R-Biopharm Rhône. R-Biopharm are pleased to extend their vitamin product range to include a new and innovative immunoaffinity column for the simultaneous analysis of biotin, folic acid and vitamin B12 EASI-EXTRACT MULTI-VIT B (LGE) (Art. No. RBRP183/P183B). Biotin, folic acid and vitamin B12 are extracted from the sample using a single extraction. The column is designed with an integrated reservoir and the top frit is absent enabling the sample extract to mix directly with the antibody gel where binding takes place between the antibody and the vitamins. The extract is passed through the column before washing which removes any unbound material. The vitamins are released from the antibody following elution before injection onto the UV-HPLC system. Column performance is demonstrated through the extraction of vitamins from a commercial infant milk formula, an adult nutritional drink and the NIST 1849a standard reference material.
