12 PERSONALIZED MEDICINE APPROACH Treatments for resistant cancers can be customized based on in vitro results [ Niederst MJ and Engelman JA (2013) Bypass mechanisms of resistance to receptor tyrosine kinase inhibition in lung cancer. Sci Signal 6: re6 ] The challenge is to develop accurate drug screens using the patient s own cells which have been grown in a dish. This requires the effective reproduction of the 3D tumor microenvironment, which we are accomplishing through humanderived ECM and optimal growth conditions. [ Crystal AS, et al. (2014) Patient derived models of acquired resistance can identify effective drug combinations for cancer. Science 346: ] This approach can be exploited to predict the best drug treatment regimen for patients who may have already endured previous treatments and have developed a resistant cancer. 12
