Abstract of Work: 200 individuals (100 Patients, 50 Family member of patients and 50 Normal individuals with no family history) were screened from Eastern India for mutations throughout the entire coding region of both genes. The main focus behind the study is to provide reliable hospital based estimates of genetic influence, and to characterize the nature and prevalence of BRCA1and BRCA2 germline mutations in early-onset and familial breast cancer cases. Mutation detected by PCR-SSCP, Direct Sequencing, Multiplex/duplex PCR. Haplotype analysis done for detecting the source of the mutation. 2 Supervisor: Dr. Jayasri Basak, Senior scientist, Dept of Molecular Biology, Netaji Subhas Chandra Bose Cancer Research Institute, 16 A Park Lane, Kolkata, India. ncri.molecularbiology@gmail.com Abstract of Work: The majority of breast cancer patients found to have BRCA1 or BRCA2 also reveal a family history of breast and/or ovarian cancer in at least one first-degree relative. To assess the contribution of BRCA1 and BRCA2 mutations to breast cancer in young Indian women, a hospital-based study become undertaken. We are analyzing a sample of young breast cancer patients who are treated at our cancer research Institute. Patients are selected solely because they had a diagnosis of breast cancer at a young age and were observed in our clinics, and were not selected on the basis of family history. Complete sequencing for germline mutations in BRCA1 and BRCA2. We also establish a correlation between the different hormone receptor status with the breast cancer patients with BRCA mutation along with the nodal status and other pathological factor of the tumor. In West Bengal till date, no such supporting data of BRCA1 and BRCA 2 gene causing mutations along with the correlation between the BRCA mutation status with clinical characteristics are available. From our hospital record, we have observed that there are many breast cancer patients whose molecular level diagnosis and family level survey has not yet been done. We expect association of mutation(s) with the risk of breast cancer among Eastern Indian population. We also study the immune-pathological status among the BRCA mutated patients. Using our data, we hope to develop a model in the future to identify patients with the lowest risk of tumor metastasis and give them better counselling with clear information about the disease and its treatment. Project Experience: Project Title: Status of Prevalence and Awareness Among Thalassemia Population: Special Reference to Various Tribes Belonging in Different District of West Bengal Funded by: Dept. of Biotechnology, Govt. of India. (2009 to present) Supervisors: Dr. Ashis Mukhopadhyay, Medical director, NCRI, Kolkata, India Abstract of Work: Dr. Jayasri Basak, Senior scientist, Dept of Molecular Biology, NCRI, Kolkata, India Thalassemia is the most common monogenic disorder in the world. In developing countries like India thalassemia increases the burden of health care. Every year 10,000 children with thalassemia major are born in India that constitutes 10 percent of the total number in the world. There is about 10-15% thalassemia carrier in West Bengal and it increases by per year. We are 2

3 undertaking mainly four strategies to prevent thalassemia. i) Thalassemia awareness and screening camp followed by post awareness camp; ii) Genetic counseling of the parents of thalassemia patients ; iii) Bone marrow transplantation of the thalassemia patient after selection of HLA matched donor ; iv) Promoting prenatal diagnosis (PND) of the fetus of pregnant mothers where both the parents are thalassemia carrier PEER-REVIEWED PUBLICATIONS 1.
