Modulation of gut microbiota for wellbeing Dr. Jelena Vulevic Clasado Research Services United Kingdom

What is wellbeing? State of being comfortable, healthy or happy

Health in 21 st century 225 = 45% of World overweight 25 = 4% of World > 65 years old Increased chronic disease (CVD, T2D, GI disorders, some cancers and dementias) = major societal and financial concern Health conscious consumers Natural depletion of nutrients from foods

Health model in 21 st century Has to comprise of both preventative life style and therapeutic entities, with diet playing a principal role Functional foods = properties over and above nutritional value

Gastrointestinal tract Microbiome the most metabolically active organ in the body ~ 6% of all the cells in the human body are bacteria Last 2 years symbiotic relationship Today one of the main root causes of all diseases GALT - ~ 7% of the immune system Innate and adaptive defences Discriminates between harmful and innocuous antigens Managed by microbiota

Great news for us! Microbiota = link between lifestyle/diet and wellbeing Appropriate dietary management = a route to an overall wellbeing and healthier ageing

Prebiotics = Functional Food Theory = any dietary material that enters the colon Practice = NDO s - low in natural ingredients but produced commercially Criteria escape enzymatic digestion in the upper gut enter the ceacum without change to their structure confer the degree of selective fermentability Currently more than 2 prebiotics on the market - FOS and GOS with most evidence in humans

BIMUNO B-GOS (Mode of action) Prebiotic Tailored towards bifidobacteria = positive microbiota modulation Mixture of galactose based oligosaccharides produced from lactose through the transgalactosidic activity of b- galactosidase Anti-adhesion (eg Salmonella and E. coli) Improved GI function (eg discomfort) Immuno modulating effect (eg antiinflammatory) Metabolic effect (eg lipid metabolism) Gut Brain Axis (eg. behaviour, stress)

PI scores and % bifidobacteria In 3 healthy volunteers (RDPC), Bimuno was twice as effective as V-GOS in increasing bifidobacteria and PI score (Depeint et al, 28) B-GOS (1.35g/d) B-GOS (2.65g/d) V-GOS (2.78g/d) Prebiotic index Bifidobacteria

Mean weeklly records of gut function parameters In 83 adults (RDPC) who suffer with GI discomfort, significantly reduced flatulence, bloating and abdominal pain (Vulevic et al, 218) 3 - Preventing activities 2 - Not preventing activities 1 - Tolerated start week 1 week 2 - None.5 B-GOS Placebo B-GOS Placebo Bimuno-GOS Placebo Bloating Flatulence Abdominal pain

Fecal bifidobacteria copies/g Bifidobacterium copies/g Supplementation with Bimuno to FGD patients (n=4, RDPP), results in effect on symptoms comparable to LFD while also preventing the decline in bifidobacteria (Huaman et al, 218) B-GOS LFD fecal bifidoba cteria copies/ g B-GOS Bimuno FODMAP LFD flatulence 1 pain 2 distension 3 borborygmi 4 bloating 5 pre-treatment (white), treatment (black) and post-treatment phase (grey)

Cortisol difference at 4 wks from start (nmol/l) Vigilance (ms) at 4 wks In healthy students (n=4, RDPP), 4 weeks of supplementation resulted in reduced cortisol and improved emotional processing (Schmidt et al, 215) 5 Placebo FOS B-GOS 4 Placebo B-GOS FOS positive negative

Want to know more? Come along for a chat to stand 798 to learn more about Bimuno
