The laboratories are in the Histology Laboratory (Room 1/56) located in the Strathcona Anatomy and Dentistry Building. There are 23, 1-1/2 hour lectures and 12 laboratory periods. The first lecture will be held on September 5 th and the last lecture will be held on November 24. Labs will begin the week of September 12 th. There are no Labs the first week of class. 3 EVALUATION: 1. Written assignment - 10 marks 2. Laboratory participation 5 marks 3. Laboratory quizzes 5 marks 4. Final Examination written essay-type questions - 30 marks multiple choice-type questions - 30 marks 5. Final laboratory examination - 20 marks (spot-type identification on digitized pictures). Final laboratory examination may (sometimes) be held on the same day as the written examination (to be confirmed). Letter grades, only, are given. 4 ANAT 261: Introduction to Dynamic Histology 2017 Assignment Due Date: 10:05 a.m. Tues Oct 3, 2017 (submit paper copy at the beginning of lecture). Late penalties apply (-10 marks per day). Format (Assignments which do not respect these guidelines will be penalized) Length: max 8 pages (EXCLUDING references and title page). Additional pages will not be marked. Font: Calibri, size 11, standard margins, double spaced Title page (1 page): Include your lab day, full name, and student ID# Drawings: must be done by hand. Photo copies or images made using a computer will not be marked. All important features and layers should be clearly labelled. Artistic skill is not evaluated, but clarity is vital. Cell shapes and layers should be representative. Drawing should include a title. References (1 page): APA format. May be single spaced. Missing or incorrect citations will be penalized. (Note that Wikipedia and your professor s lecture notes do not count as references.) Section 1: Common histological methods (25 marks, max length 2 pages) a) Define H&E and explain its basic principles. Please use examples of what is stained. (5 marks) b) You receive a freshly prepared slide that a lab colleague prepared with an H&E protocol. Upon placing it under the microscope you notice that areas of the tissue section are unstained. Explain what could have gone wrong. (4 marks) c) Name three common stains other than H&E that are used in histology. What are these stains used to detect? (6 marks) d) Explain the basic principles of immunohistochemistry for fluorescence microscopy using indirect labelling. Why is direct labelling often less useful? (5 marks) e) Which microscopy technique would you use to examine structural details of cell- cell junctions in a fixed sample? Why? (5 marks) Section 2: Epithelia (40 marks, max length 3 pages) a) Draw a non- keratinized stratified squamous epithelium. Give 3 examples of anatomical regions of the body where this type of epithelium is present. (8 marks) b) Describe the composition of the basement membrane and describe a disease in which the basement membrane is compromised. Give an example of how you might stain the basal lamina. (12 marks) c) There are two types of epithelia: simple and compound. Name and draw 3 classifications of compound epithelia and give an example of where each exists in the human body. (20 marks) Section 3: Histology in research (35 marks, max length 3 pages) In biological and medical sciences, histology can be used for various purposes (observing the state of a diseased organ, assessing the structure of an organ thought to be dysfunctional, assessing the safety of a drug, searching for the manifestation of parasite contaminate, etc.). Go to a scientific search engine (e.g. PubMed) and find a primary research article published in the past year that uses histology (if it was not published in the past year, automatic zero on this 4
5 section!) Copy/paste a figure from this publication which shows histological results. Describe the protocol used for this histological preparation. Discuss how this figure fits in the publication (the results it offers, how it addresses the question the investigators were trying to answer, why use this technique if others are available, etc.). 5
