Personal Statement I am an Assistant Professor at the Department of Aging and Geriatric Research at the University of Florida. After completing my undergraduate degree in Physiotherapy in Poland, I pursued my doctoral training at the Department of Rehabilitation Medicine in Rotterdam in The Netherlands under supervision of a sports physician, Dr. Stephan Praet and mentorship of the Department Chair, Dr. Henk Stam. During my doctoral fellowship I was interested in cardio-respiratory and microvascular responses to hypoxic and hyperoxic air conditions during exercise in young adults and type 2 diabetes patients (a,b,c). During that period of time, I gained a lot of experience in exercise physiology testing (spiro-ergometry) and non-invasive cardiovascular assessments (e.g. hemodynamic monitoring, near-infrared spectroscopy). Additionally, I supervised one medical student and one movement science master student, which resulted in successfully defended master theses. Moreover, I led three-month courses on exercise physiology for medical students (four academic years). I learned the Dutch Language (fluent) within 1 year for the communication and administrative purposes. At the end of my doctoral fellowship, I spent three months at the Institute on Aging at the University of Florida where I continued practicing my writing and analytical skills under supervision of Dr. Christiaan Leeuwenburgh and Dr. Stephen Anton. This collaboration resulted in three papers and sparked my interest in aging research. After completing my doctoral degree, I began my postdoctoral training at the University of Florida under the same mentoring team. My postdoctoral training consisted of gaining experience in basic science (highresolution respirometry, Western blots, ELISA, DNA damage assays, PCR) and cardiovascular clinical assessments (methods: speckle tracking echocardiography and flow-mediated dilation). I have developed collaborations with Departments of Pathology and Cardiology at the University of Florida to increase competitiveness of my translational cardiovascular research. As a faculty, my research is focused on antiinflammatory interventions to improve cardiovascular function in rather healthy older adults. I am also implementing my cardiovascular measures in critical care and developing my research niche that combines cardiovascular disease and outcomes after critical care (d), inflammation and aging science. Older critical care survivors demonstrate high levels of persistent inflammation and high risk of cardiovascular disease. My work is supported by the American Heart Association and NIH. a. Contributions to Science 1. Effect of dietary antioxidant supplementation (AS) in conjunction with exercise training on health benefits. Few recent pre-clinical and clinical reports have shown that AS in combination with endurance training may blunt the beneficial effects of exercise (e.g. mitochondrial biogenesis, insulin sensitivity, blood pressure), and therefore may impair healthy aging. I reviewed the available pre-clinical and clinical studies and concluded that the reason for inconsistent effects of AS could possibly be in different AS compositions, doses, duration time of supplementation and exercise training modalities. Our future studies are based on conclusions of this review (Mankowski et al. 2015). Findings on the effects of other nutritional compounds on tackling inflammation in aging will result in future clinical trials (Mankowski et al and Custodero et al. 2018). a. Effects of Fermented Papaya Preparation (FPP) on Safety Outcomes in Older Adults - A Short Report of a Placebo-Controlled Clinical Trial. J Frailty Aging. 2018;7(2): Physical activity is important to prevent development of the metabolic syndrome. However, extended sitting time during a day may also increase the probability of developing the metabolic syndrome in older adults. We found in a cross-sectional analysis of the multicenter clinical trial (the LIFE Study) that higher levels of objectively measured sitting time during a day was associated with higher probability of developing the metabolic syndrome and higher probability of developing diabetes (Mankowski et al. 2015). This fellowship provides full postdoctoral salary coverage, research and travel funds. The award was declined by the PI due to his transition to a faculty position. The Claude D. The central hypothesis is that resveratrol treatment will improve mitochondrial function by activating
5 key genes involved in mitochondrial biogenesis and metabolism, and that these biological/cellular changes can enhance physical performance among both low to moderately functioning older adults. NIH 1R21AG A1 (Buford) 4/1/2016-3/31/2018 Total award funding: $412,500 Role: Co-Investigator Resveratrol and exercise to treat functional limitations in late life This study will be the first to evaluate the potential of two doses of resveratrol for improving the efficacy of physical exercise in treating age-related functional limitations. As such, the study has important implications for advancing the NIA mission of developing interventions to improve the health and quality of life older adults.
