2 BioSiteHisto Nordic BioSite wants to make diagnostic work easier. For this purpose, we have selected a number of markers, which are frequently needed for diagnostics in the routine laboratory. We have selected antibodies with the characteristics that they work well for immunohistochemistry (IHC) and have adapted the format and size to meet the needs of most diagnostic laboratories. In this catalogue, you will find the Nordic BioSite selection of CE/IVD certified antibodies for IHC. They have all been tested and approved for IHC on formalin-fixed paraffin-embedded tissue. The continuous control and the high lot-to-lot consistency make them ideal for diagnostic and routine use. We provide them in a concentrated format in standard sizes of 0.5 ml and 1 ml. Most of the antibodies are also available in a 6 and 16 ml ready-to-use format. Other sizes are available upon request. Standard storage is at 4 C and expiration date at least 1 year from reception. 16 Detection Detection Systems BioSite Histo lus detection systems are characterised by outstanding sensitivity and are recommended for use in both automated and manual immunohistochemistry. BioSite Histo lus reagents are mainly IVD/CE certified. You can choose between polymer systems and streptavidin-biotin systems. Both kit systems are available with alkaline phosphatase (A) or horseradish peroxidase (HR). BioSite Histo lus olymer Detection Systems (High Sensitivity) Our BioSite Histo lus olymer Kits provide high sensitivity and ready-to-use reagents. Since the enzyme is directly linked to a polymer, background staining due to endogenous biotin is avoided. erfectly synchronized components guarantee brilliant staining results. You can choose between alkaline phosphatase or horseradish peroxidase kit systems. 20 Reagents Epitope retrieval retreatment of tissue after paraffin embedding can be divided into different parts. One is to get rid of the paraffin hiding the epitope of interest and retrieve antigen so antibodies can detect it. Nordic Biosite has unique products with this focus. Our Omnirep Solution can be used for heat induced epitope retrieval (HIER). The working solution will have a ph 9 and will increase epitope expression in the tissue. Depending on selected primary antibody epitope unmasking can be performed with HIER with different buffer solutions with different ph or enzymatic digestion. Often works ph6 and for increased binding potentially ph9. The lower ph with focus on antibody pi which is close to ph6. At ph 6 will the antibody in theory bind with it s antigen specific part. With higher ph will the antibody be more charged and can add other binding possibilities in it s interacting with tissue. All protocols has to balance between maximal specific binding of antibody and epitope retrieval. Each producer of antibodies test their antibodies with at least one protocol and for receiving the same result this protocol must be followed. Every pathology laboratory tests a lot of antibodies to select which one s that deliver the correct staining pattern according to this laboratory procedure. Several laboratories have different protocols and different preferred clones that works with their specific protocol. Most laboratories has selected Citrate buffer as pre-treatment and we have the same as standard protocol. 23 Control Cell Blocks Control Cell Blocks Nordic BioSite is now able to offer Control Cell Blocks for immunohistochemistry and in situ-hybridisation. Our Control Cell Blocks provided by the company multiblock are homogenous paraffin blocks with different numbers of embedded cell lines. These micro arrays are so small that they can be mounted together with the sample of interest e.g. patient specimen on one slide. The cell lines are fixed in buffered formalin under controlled conditions. The paraffin block can be cut as any regular paraffin block on a standard microtome without any further equipment. Red dyed paraffin was used for better orientation during cutting and mounting. Sections can be mounted on normal or coated slides following standard methods. A tissue core of dog heart muscle is included for rapid and easy orientation of the micro array on the glass slide. All Control Cell Blocks are labelled for research use only. Nordic BioSite offers four different control blocks The Receptor Control Cell Block contains 4 types of cores: a negative control cell line, ER positive cell line, R positive cell line and a cell line with HER2 overexpression. The cell lines will positively react to IHC staining of estrogen receptor, progesterone receptor and HER2 antibodies. Each block consists of a set of low and a set of normal expressing cell lines. Thus, a differentiation between high and low staining intensity is possible, indicating the sensitivity of the stains.
