3 Place of origin South Africa Mogalakwena has an overwhelming wealth of flowering plants the vegetation in the largest parts of the country is dominated by diverse grasses, low shrubs and acacias Mogalakwena is located in the north of South Africa at the Limpopo River, embedded in the region bordering the countries Botswana, Zimbabwe and Mozambique. Here where the grass and thorn savannah gradually turns to denser bush savannah we discovered a special succulent plant. 3

4 The Plant Botanical name: Bulbine frutescens (L.) Willd. This succulent belongs to the family of Asphodelaceae Bulbine is a plant with a rosette of fleshy yellowish-green leaves The plant is 12 to 15 cm high and its leaves are thin and triangular A gel-like juice can be obtained by cutting open the leaves 4

5 Indigenous use The local inhabitants, who live in harmony with nature, have been familiar with this plant for generations and use it as a remedy for numerous ailments. Magogo the mother of grandmothers who has an extensive knowledge of the grasses, roots, herbs, leaves and bark found in the area, shared with us the secrets of the South African legends and the ancestral art of healing. 8 Efficacy To evaluate the activity profile of CremerTIMOLA the modulation of gene expression were investigated on normal human epidermal keratinocytes (NHEK) The effects were evaluated using RT-qPCR technology Tests were conducted under basal and inflammatory conditions Target genes were selected for their importance in keratinocyte differentiation and inflammation Reference for the basal condition: Calcium, well known for its cell differentiation activity Reference for the inflammatory condition: PMA (Phorbal 12-Myristate 13- Acetate) stimulating a generic inflammatory profile 8

13 Efficacy Summary of the gene expression study on epidermal keratinocytes CremerTIMOLA showed a clear pro-differentiating effect on keratinocytes as observed with the strong increased expression of many differentiation markers The increased expression of many markers involved in cell-cell interactions and lipid synthesis gives rise to the presumption that CremerTIMOLA enhances the skin barrier properties The up-regulated expression of AQP3 (Aquaporin 3), involved in the water/glycerol transport indicates a moisturization effect At least 4 markers of the PMA induced inflammatory profile have been reversed. However, more tests on different inflammatory profiles are required to verify an anti-inflammatory effect 13
