1 Advanced Materials Research Online: ISSN: , Vols , pp doi: / Trans Tech Publications, Switzerland Effects of Motherwort total alkaloids on prostatitis model of mice Mingsan Miao, Lin Guo,Shuo Tian Henan University of Traditional Chinese Medicine, Henan, , china Keywords: Motherwort total alkaloids; Prostatitis; Mice model Abstract Objective: To investigate the effect of Motherwort total alkaloids on prostatitis model of mice.methods:different groups of mice for Motherwort total alkaloids are given high,medium,low suspension.observe the number of white blood cells, lecithin corpuscle density groups in prostatic fluid and the morphological changes of prostate,testis, epididymis.results:compared with the model group, the Motherwort total alkaloidss high, medium dose group can significantly reduce the number of white blood cells of prostate in mice model of prostatitis and increase lecithin corpuscle number (P<0.01); the Motherwort total alkaloidss high dose group significantly reduce the pathological changes of prostatitis (P<0.01); the Motherwort total alkaloidss medium dose group significantly reduce the pathological changes of prostatitis (P<0.05). the Motherwort total alkaloidss high dose group significantly reduce the pathological changes of testis (P<0.05)and it can significantly reduce the pathological changes of the epididymis (P<0.01).Conclusion:Motherwort total alkaloids has a therapeutic effect on prostatitis model of mice. Introduction Motherwort is the fresh or dried aerial part of Leonurus japonicus houtt, with the flavor of bitter and pungent and the property of somewhat cold,belonging to the main and collateral channels of liver,pericardium,bladder.the main effective components of Motherwort is total alkaloids,including stachydrine hydrochloric acid and so on[1].modern pharmacological research shows that Motherwort can improve microcirculation and myocardial ischemia.[2] Prostatitis is a common disease among adult males, accounting for about 25% ~ 30% of male patients admitted to department of urology,and occurs in 20 ~ 40 year-old young adults[3]. As a high incidence disease,related research of prostatitis is particularly important.this paper reported on the effects of Motherwort total alkaloids on prostatitis model of mice. Materials Animals.KM mice, SPF grade, Weight 28~30g, supplied by Experimental Animal Center of Henan Province, Animal permit number: Instruments and reagents drugs.fa (N)/JA (N) electronic balance (supplied by Shanghai Minqiao Precise Science Instrument Co., Ltd.).Motherwort total alkaloids, provided by the Henan University of Traditional Chinese Medicine Chemistry Laboratory of traditional Chinese medicine (in 58.23%).Qianliekang(supplied by Zhejiang Conba pharmaceutical Limited by Share Ltd, batch number: ).Xiaozhiling injections(supplied by Beijing double crane pharmaceutical Limited by Share Ltd, batch number: )sodium pentobarbita(supplied by China Pharmaceutical Group Shanghai Chemical Reagent Company, batch number: F ). 2 1050 Progress in Applied Sciences, Engineering and Technology Methods 60 male KM mice, male, weighing 28~30g, were randomly divided into blank group(bg), model group(mg), positive control group(this group can also be called Qianliekang),the Motherwort total alkaloids in high,medium,low dose group(these groups can also be called the Motherwort total alkaloids in HD,the Motherwort total alkaloids in MD,the Motherwort total alkaloids in LD).BG were given sham operation and the rest groups were made model according to the literature method[5-7].from the eighth day of modeling, BG and MG were given distilled water (0.2ml/100g), positive control group were given Qianliekang capsule suspension (1.5g/kg, distilled water 0.075g/ml, dose equivalent to 15 times of the clinical dosage) (0.2ml/100g), the Motherwort total alkaloids in HD,MD,LD were given in the high,medium,low dose(37.5mg/kg,18.8mg/kg, 9.4mg/kg, respectively, equivalent to about 15g crude drug /kg, 7.5g crude drug /kg, 3.75g crude drug /kg) of Motherwort total alkaloids suspension (0.2ml/10g), 1 time per day by gavage, for 21 consecutive days. 1h after the last administration (fasted for 12 hours).the mice were killed,the rapid laparotomy, and then under microscope count the number of white blood cell in prostate massage fluid from 10 µ L; another drop of prostate fluid smear, lecithin corpuscle density were observed under microscope. Score standard: full view for 4; 3P4 view for 3; 1P2 view for 2; 1P4 view for1. Then took the part of prostate tissue and testis, epididymis and fixed in 10% formalin, embedded in paraffin, sliced, HE staining, morphological changes of prostate, testis, epididymis were observed under light microscope. Methods Statistical Analysis SPSS 11.5 statistical analysis with for Windows software package was used for data analysis, measurement data with the mean ± standard deviation ( ±s) said, variance analysis between group (LSD method) Results Influence on the white blood cells and lecithin corpuscle density in prostate massage fluid from the model mice of prostate.the results show in Table 1. Compared with the model group, Qianliekang and the Motherwort total alkaloidss group in high, medium dose group can significantly reduce the number of white blood cells in mice model of

Motherwort total alkaloids low dose group of prostatitis model mice effect on lecithin corpuscle number is not obvious, but can significantly reduce the number of white blood cells in mice (P<0.01). The experimental results show that, Motherwort total alkaloid improve prostatitis in the prostatitis model mice. Influence on prostatitis model mice organs morphology Influence on prostate tissue and the results show in table 2. Table 2: Effect of Motherwort total alkaloids on prostate tissue of the model mice of prostatitis group n Blank Model Qianliekang Motherwort total alkaloids in HD Motherwort total alkaloids in MD Motherwort total alkaloids in LD The prostate gland, glandular epithelium and stroma are normal; + The prostate gland less expansion, glandular epithelium appear flat, fiber hyperplasia and a small amount of inflammatory cell infiltration around small glands; ++ The prostate gland hyperplasia, Ming glandular cavity expansion, glandular epithelial flat, interstitial fibrous proliferation and inflammatory cell infiltration; +++ The prostate gland hyperplasia, luminal dilatation, glandular epithelial flat, interstitial fibrosis and inflammatory cells infiltration significantly. Table 2 shows that, by Ridit test, compared with the blank group, pathological changes significantly in prostatitis model group (P<0.01), compared with model group, the pathological changes of Motherwort total alkaloid HD can significantly reduce the prostatitis (P<0.01), pathological changes in the medium dose group can significantly reduce (P<0.05). Influence on testicular tissue and the results show in table 3. Table3 shows that,by Ridit test,compared with the blank group,testicular pathological changed evident in model group (P<0.01), compared with model group, the Motherwort total alkaloids in HD can obviously reduce the pathological changes of testis (P<0.05).

Table 4 shows that, by Ridit test, compared with the blank group, in the epididymis, pathological changed of the model group were significant (P<0.01), compared with model group, the pathological changed of Motherwort total alkaloids in HD can significantly reduce the epididymis (P<0.01). Summary White cell stacks or clustered distribution often suggests the presence of inflammation of the prostate, prostatitis lecithin corpuscle can be significantly reduced, and the piles of tendency[4]. The pathological features of chronic prostatitis is mainly interstitial cell infiltration, fibroblast hyperplasia, gland duct obstruction, glandular secretion decrease, so the observation of prostate tissue is exploring visual indicators prostatitis. Testicular discomfort is sensory index prostatitis. Observation in the testis and epididymis pathological changes has the important effect on prostatitis research. No records of prostatitis disease in Chinese medicine, generally attributed to drench card, turbid sperm, leucorrhagia, gonorrhea. The basic pathogenesis of the disease is damp heat and blood stasis[5].motherwort has the inflammation of stasis, diuretic swelling,heat clearing and detoxifying.it coincides with basic disease machine function and is the main treatment of traditional Chinese medicine "certificate".the experimental results show that the Motherwort total alkaloids has good effects on treating prostatitis, and can significantly reduce the pathological changes in related organs.the experiment provides support for clinical treatment of prostatitis by Motherwort, also provides new ideas and methods for the prevention and cure of prostatitis.
