14 Balanced brain function requires balanced action of neurotransmitters axon Communication of information between neurons, involves the movement of chemical neurotransmitters across the synapse The neurotransmitter exerts its effect until it is removed from the synapse by: Diffusion Degradation / Metabolism Re-uptake / re-absorbtion Slowing these processes, increases the residence time of the neurotransmitters in the synapse & so increases their effect synapse signal Page 13
15 DSM s pure compound and extract library An oregano-extract was one of those identified through research to find new ingredients from food with positive effects on cognition and mood. This oregano-extract (orévida) was found after profiling food extract libraries for in-vitro activity in assays for serotonin-re-uptake, noradrenaline-reuptake, dopamine re-uptake and monoamine-oxidase (MAO) A&B activity. Sources of food plant extracts? > 4000 pure compounds (individual phytochemicals) > 1200 food plant extracts > fractions from extracts Feb, 2011 Page 14
16 Project progress Preclinical The active components in the extract have been identified The mechanisms of action are well elucidated In vivo, the levels of the active components in the extract increase in the brain after oral administration After administration the levels of neurotransmitter are increased in the brain Pre-clinical studies in validated animal models have shown beneficial effects of the extract on behavior Safety No genotoxicity No reprotoxicity No adverse effect level has been determined in 13w toxicity trial The effective dose is safe and exhibits no adverse side effects No interference with drug metabolizing CYP enzymes orévida is well tolerated (in rodents and humans) Clinical The extract improved attention and vigilance in a first in human trial as measured by waking electroencephalograph (EEG) and Event Related Potentials (ERP) Page 15
