As gallbladder stones affect a significant percentage of the population in many countries throughout the world and the major of cause of abdominal morbidity. There is no effective drug for gallstone disease now a day. Removal of gallbladder with gallstones will solve the gallstone disease problem permanently. It affects more than 20 million Americans and causes hospitalized each year, 10-15% adults in Europe and USA prevalence in gallstone disease. Gallbladder surgery is one of the most common operations with direct cost of more than $2 billion. The present study deals with morphological characterization of human gallbladder tissue with different (Brown, Black and Mixed or white) types of gallstones problem. Introduction Key words : gallbladder, SEM-EDS, FTIR Gallbladder stones affect a significant percentage of the population in many countries throughout the world and the major cause of abdominal morbidity [Heaton et al., 1984]. It affects more than 20 million Americans and are hospitalized each year [National institute of Diabetes and Digestive and Kidney Diseases., 1995], making gallbladder surgery one of the most common operations with direct cost of more than $2 billion [National Center for health statistics., 1994]. Although many studies on calculi have been undertaken hitherto, most of them are dealing with etiological or clinical problems and with all those studies no perfect explanation in the field has been made. Gallstones are primarily composed of cholesterol[1-9]. Human gallstones may be generally classified into three types (1) cholesterol stones, (2) brown pigment or calcium bilirubinate stones, and (3) black pigment or pure pigment stones [Kaufman et al., 1994; Chijiikawaet al., 1995]. Its classification was[10-16], proposed at the NIH International 7069

2 Gallstone Workshop [Solowayet al., 1990]. Although, other types such as combined (cholesterol and pigment) are also found[17-24]. Gallstones are composed of a mixture of hard-to-separate substances of different chemical natures, mainly cholesterol with admixture of higher fatty acids, bile acids, phospholipids, bilirubin, protein etc [Tretyakovet al., 2007]. Materials and method Subject and methods The randomly selected 15 (5 brown stone, 5 black stone and 5 mixed or white stone) patients and 5 (no stone in gallbladder) controls are included in this study. The study period was between May 2007 and June 2009[25-32]. Diagnosis of gallstone disease The diagnosis of gallstone disease was verified on the basis of medical records showing either a history of cholecystectomy or a diagnosis of gallstone disease based on cholecystectomy or ultrasound investigation[33-39]. All patients fasted at least eight hours before examination. SEM analysis The morphology of the gallbladder tissues were characterized by Scanning Electron Microscope and qualitative elemental concentration were studied by Energy Dispersive Spectrum attached with SEM instrument (SEM JSM 840A JEOL, Japan). Obtained gallbladder tissue samples were lyophilized at 6 hours to remove water content, the tissue samples were grained to form powder using pestle and mortar. The homogenous powders were subjected to SEM analysis[40-45]. 7070

3 FTIR analysis Powdered gallbladder tissue samples were mixed with KBr in the ratio of 1:40 to form pellet, and FTIR is recorded in the range of cm -1 at Nicolet Avatar instrument. Results and discussion All the gallbladder tissue samples with various (normal (no stone), brown, black and mixed or white) gallstones patients were subjected to SEM and FTIR analysis. Morphological structure of human gallbladder tissues (normal 1(a-d), brown 2(a-d), black 3(a-d) and white or mixed 4 (a-d) ) with various magnifications (100 X, 1000 X, 3000 X) and EDS of the gallbladder tissue samples respectively are shown. Table 1 shows the SEM-EDS analysis of human gallbladder tissues (normal (no stone), brown, black and mixed or white) gallstone patients. Fig 2 represents the relative elemental distribution of all gallbladder tissue samples. FTIR spectrum of all gallbladder tissue samples are show in the figures 1d, 2d, 3d and 4d. Table 2 is the visual intensity estimation and tentative assignment of gallbladder tissue samples. Change in elemental concentration are due to many of the reasons i.e. Native bile chemical composition, absorption of metals and so on. Conclusion There is no appreciable chemical changes were found in the normal and other gallstone patients gallbladder tissue samples. Some morphological and elemental changes were found in 7071

4 SEM image and EDS data. Some of the elemental concentrations were coincide with gallstone and bile samples.
