Neurocognitive, Neurophysiological, Structural, Cytoarchitectonic, and Molecular Measures in Williams Syndrome Narrative Report of Research: My research interest in schizophrenia has focused on brain abnormalities in the superior temporal gyrus (STG) of the temporal lobe. The STG, the auditory-language-related region of the brain, has a long history of being implicated in schizophrenia and conceptually linked with its positive symptoms. Given the complexity of schizophrenia and approaches that can identify and examine more than one level of dysfunction, I have used three technologies to investigate the STG: in living subjects, (1) Electrophysiology (P300 evoked-potential voltage measures) and (2) MRI (tissue volume measures); and in postmortem brains, (3) Histology (neuronal-cell measures). In addition, I have investigated the differentiating effects of handedness and gender in studies of schizophrenia and found that indeed, handedness and gender do show differentiating differences at electrophysiological and MRI levels. More specifically, P300 in right-handed schizophrenic males showed left-sided P300 voltage reductions whereas in left-handed schizophrenic males, there was a right-sided P300 voltage reduction. With regard to gender and P300 in right-handed schizophrenic females, results did not show lateralized asymmetric reductions, rather, voltages were lower at the central sites. Moreover, there were fewer left-handed schizophrenic females compared to schizophrenic males suggesting that left-handedness may occur as an anomaly more frequently in schizophrenic males. At a brain structural level, the MRI data of left-handed schizophrenic males show right-lateralized tissue volume reduction in the STG. In concert, these structural data support the findings of right-lateralized P300 temporal region voltage reductions in the left-handed schizophrenic males. And finally, at a third level, a microscopic level, preliminary histological data show cell differences in the STG in postmortem schizophrenic male brains, namely decreased cell packing density and cell size differences compared to the postmortem control brains. These STG cell alterations not only support electrophysiological voltage reductions, but also support data from other researchers reports of STG tissue volume reductions in right-handed schizophrenic males. Because of the numerous neuropathological abnormalities in schizophrenia reported in the STG, an area not only specialized for hearing and language, but also conceptually related to its classic symptoms, my interest in the importance and uniqueness of STG research in schizophrenia has been threefold. First, my data show that gender and handedness have differentiating effects on neurophysiological and gross anatomical abnormalities in the living brain in schizophrenia. Second, neurodevelopmental factors related to these effects may have a critical influence on the STG. Third, the preliminary histological data of cellular differences contribute a new level of STG analysis, one heretofore lacking, to the existing database of STG abnormalities in schizophrenia. In sum, the congruent findings of electrophysiological, structural, and histological anomalies in the temporal region, specifically the STG, in schizophrenia, show that gender and handedness have differentiating effects within schizophrenia, and also a histological basis for brain-behavior abnormalities reported in the STG in schizophrenia. The significance of this research is the elucidation of the neuropathology of schizophrenia by identifying neural correlates at three different

4 Page 4 levels of brain organization and function, and demonstrating the differentiating effects of gender and handedness within this debilitating disorder. Extending my interest in the STG, my recent research efforts focused on measurements of postmortem neurons in both primary auditory and visual areas in Williams Syndrome. Williams is a mental retardation syndrome affecting 1 in 25,000 births and associated with genes that are hemideleted on Chromosome 7. This syndrome is a compelling neurodevelopmental disorder because of the juxtaposition of cognitive strengths and weaknesses. Williams subjects show relative preservation of language, have excellent face recognition, and often have remarkable hearing and musicality. Yet they show severe visuospatial deficits, deficits that co-exist with cognitive and auditory strengths suggesting that the dorsal part of the brain, especially area 7 in the parietal lobe, is more affected than ventral auditory and language areas. In contrast, the cognitive strengths, language preservation, and auditory acuity in Williams point to primary auditory cortex, area 41, as being less affected. This hypothesis was supported in my finding that area 41 was relatively preserved in Williams postmortem brains compared to controls. Earlier work showed neuronal anomalies in primary visual cortex in Williams. The results in primary visual cortex show significant differences in the opposite direction than the more recent evidence found in primary auditory cortex, all of which point to a neuronal substrate underlying visual spatial impairment and one underlying the relatively preserved auditory-language function in Williams subjects. My most recent endeavor was teaching a course at the Harvard Summer School. The course, Brain and Behavior: Research Methods and Technologies, was an introduction to various research methods and technologies used to investigate the relationship between brain and behavior. From an introduction to how the brain begins in embryogenesis to primary and association cortices in the adult brain, the focus was the study of selected brain and behavior abnormalities using electrophysiological, neuroanatomical, and neuronal measuring tools to address research questions in selected abnormalities, such as schizophrenia and Williams Syndrome. To augment the importance and excitement of the research in brain and behavior, several investigators presented their research to the students. In addition, students visited clinical and research laboratories, and also observed a postmortem brain dissection. The aim was to integrate the knowledge obtained from exploring various levels of analysis into a coherent understanding of brain and behavior.
