4 ( Abstract) ULTRASTRUCTURE OF DIGESTIVE GLAND OF DISK ABALONE ( HAL IO TIS DISCUS HANNAI) L I Tai2Wu XU Ji2Lin DIN G Xin L IU Yan ( Key L aboratory of Mari ne Biotechnology, Ni ngbo U niversity, Ni ngbo , Zhejiang, China) The digestive gland of Disk abalone ( Haliotis discus hannai) t ransmission elect ron microscopy. ( Gast ropoda : Haliotidae) was st udied wit h The specimen of Haliotis discus hannai ( shell lengt h 7 cm) were collected f rom Dalian coast in Portions of t he digestive gland were dissected out and fixed in 215 % glutaraldehyde (011 mol/ L phosphate buffered containing 10 % sucrose ; p H 712) at 0 4 for two hours. After glutaralde2 hyde fixation, tissues were rinsed thoroughly in buffer (sucrose2adjusted, 0 4 ) and osmicated for 80 min2 utes (1 % osmium tetroxide ; phosphate buffered ; sucrose2adjusted, 0 4 ). After final rinsing, tissues were dehydrated in an et hanol series and embedded in Epon812 medium. Thin sections were cut using an L KB28800 Ultrotome and stained with uranyl acetate and lead citrate. Specimen grids were examined with J EM22000EX transmission electron microscopy ( TEM) at 100 kv. The epit helial layer of t he digestive gland of Haliotis discus hannai consisted of t hree types of cells. The first cell type was the digestive cells, which displayed two characteristic membrane structures : apical granules and cisternae with electron2dense cores, and there were many lipid droplet in the base part of the cells. The size and density of electron2dense cores in the cisternae with electron2dense cores was decreased gradually from apical to basement of t he cells. There are abundant t ubules or cisternae rough endoplasmic reticulums around t he nucleus. The nucleus, round or curved shapes, is very elect ron dense in some sections, block2like chromatin scattered in the nucleus. The second cell type is excretory cells which contain a great deal of large vacuoles in t heir cytoplasm displaying high elect ron2dense cores. There are many cisternae rough endoplasmic reticula and one Golgi s body near the nucleus. The third cell type is calcium cells which are pyramidal in shape and usually filled with concentric rings of calcium granules. In the cytoplasm of the three types of cells there are rough endo2 plasmic reticulum, f ree ribosomes, numerous mitochondria and nucleus wit h nucleoli.
