Research Over the last fourteen years my research has been focused on different aspects of assisted reproductive technologies such as oocyte collection, evaluation, in vitro maturation (IVM), in vitro fertilization (IVF) using epididymal and frozen spermatozoa, parthenogenetic activation, somatic cell nuclear transfer (SCNT), Intracytoplasmic Sperm Injection (ICSI), embryo culture and 1

2 embryo evaluation in different mammalian species include cattle, camel, buffalo, sheep, goat, pigs, mice and rats and humans. Also my research focused on cryopreservation of oocytes at different stage (germinal vesicle (GV) or metaphase II (MII)) in cattle, sheep, mice and rats using different techniques such as slow freezing or vitrification by Cryoloop, solid surface vitrification (SSV) or open pulled straw (OPS) trying to establish an oocyte and embryo banks for widespread application of ART in different species. I studied the effect of different vitrification techniques of immature oocytes on viability, in vitro maturation (nuclear and cytoplasmic maturation), MPF/MAPK activities, sperm penetration, in vitro fertilization, in vitro embryo development following IVF and SCNT, embryo evaluation (morphological or by different stains such as Hoechst stain and TUNEL assay for total cell numbers and apoptosis in blastocyst stage embryos) or using karyotyping technique. In addition to evaluation the effects of vitrification on oocyte spindle and chromosome configuration using immunostaining technique, fluorescence microscopy and image analysis system. I am also interested in molecular biology, cell biology and tissue culture. I have been worked with embryo cryopreservation in cattle, sheep, mice, rats and human. Current Research My current research focuses on improvement the efficiency of oocyte cryopreservation as a tool for fertility preservation in women and for genetic preservation and wide spread application of ART in animals by using mouse model. The goal of my studies is to evaluate the effect of L-carnitine (known to play an essential role in fat metabolism and energy production, and also acts as an antioxidant) supplementation during vitrification and IVM on viability, nuclear maturation, spindle configuration, ATP levels, mitochondrial activity and subsequent development of mouse oocytes vitrified at GV-stage.
