24 Genomics and Proteomics Analysis of Acetaminophen Toxicity in Mouse Liver Stefan U. Ruepp, et al TOXICOLOGICAL SCIENCES 65, (2002) Overdose of acetaminophen causes severe hepatic necrosis in humans and experimental animals. To explore its mechanism, we administered acetaminophen at subtoxic (150 mg/kg ip) and toxic (500 mg/kg ip) doses to overnight fasted mice. Decrease of ATP synthase subunits and beta-oxidation pathway proteins indicated a loss of energy production. Acetaminophen metabolite decreases activity of glutathione peroxidase, which increases acetaminophen toxicity by preventing a rebuilding of GSH pool. Morphology of mitochondria was also affected very early at top dose. Swollen, mega-mitochondria formed. 24

25 Methionine Adenosyl Transferase Protein Modified by NAPQI Enzyme which catalyses the synthesis of S- adenosylmethionine (SAMe) from methionine and ATP. S-adenosylmethionine is needed for transmethylation, transsulfuration and aminopropylation. Most SAMe is produced and consumed in the liver. More than forty metabolic reactions involve the transfer of a methyl group from SAM to various substrates, such as nucleic acids, proteins, lipids and secondary metabolites, and neurotransmitter metabolism. 25

38 Selenium Selenium is a powerful antioxidant, which plays an important part in the antioxidant enzyme glutathione peroxidase. 41 Milk Thistle Milk thistle flower is rich in three flavonolignins: silybin, silydianin and silychristin. silymarin, the bioflavonoid complex, responsible for liver protection and regeneration. 80% Silymarin. Silymarin acts as antioxidant, protecting the liver from freeradical damage and stimulates glutathione production. One capsule = 200mg. 1 capsule twice to three times daily 41

42 Lipoceutical Glutathione Liposome (derived from lecithin) is a microscopic fluid-filled pouch with walls identical to the phospholipids that make up the human cell membranes. The outer wall of the liposome is fat soluble, while the inside is water soluble. One teaspoon = 422mg. Suggested Dosage: For Adults: Begin with 1.5 teaspoonfuls (approx. 650 mg) twice daily for 7 to 10 days and can be lowered to one teaspoonful (approx. 430mg) daily as a "maintenance serving. For Children: Begin with 1/8 - ¼ of a teaspoon (approx mg) for every 30lbs of the child s weight, twice a day. 42

47 Chemical Toxin Exposure & Detoxification Info Average person is likely to harbor 500 to 1000X higher levels of toxic chemicals in their fat (adipose tissue, blood lipids and brain) then what is in their blood. Losing weight and reducing body fat has been shown to be an effective way of eliminating stored chemical toxins. The skin and gastrointestinal tract are the two primary ways of detoxifying toxic chemicals. Other organs can aide in detoxification too, but they are slower. Detoxification isn t just about ridding the body of xenobiotics, but also endogenously produced chemicals such as byproducts of metabolism and digestive system produced compounds. Niacin is an excellent aide in detoxification because of its ability to trigger lipolysis. 47

48 L. Ron Hubbard L. Ron Hubbard founder of Scientology. In 1979, he finalized a method for ridding the body of fatstored drugs and chemical residues. Much of his research interest was prompted by a desire to help former LSD users who often continued to feel the effects of the drug years after taking it. Current estimates are that over a 100,000 people worldwide have gone through the Hubbard Protocol, a.k.a Purification Program. Program is designed to increase mobilization of toxic compounds and prevent their re-absorption and recirculation and assist in their elimination. 48

49 Hubbard Protocol - highlights 1. Daily dosing of immediate-release Niacin: Stimulates lipid lipolysis for mobilization free fatty acids (FFA). There is an initial drop of FFA after niacin, then a marked rise within a few hours and then return to normal. 2. Aerobic exercise: To help increase circulation and distribute niacin throughout the body. Helps to increase circulation of toxins for detoxification via the kidneys, liver and digestive system. 3. Sauna (intermittent): Used to force sweating, which is the primary route of detoxification of toxins. Temperature range between 140 to 180 degrees Fahrenheit. Frequent showers and hydration. 49

50 Hubbard Protocol 1. Cold-pressed oils: Provided to help prevent mobilized toxins from being reabsorbed by the intestines. Hubbard noted that polyunsaturated oils enhance excretion of persistent chemicals without increasing fat deposits in liver. 2. Vitamin and mineral supplementation: Used to help replenish lost vitamins, minerals and electrolytes. 50

Forced sweating by sauna at F for 2.5 to 5 hrs daily, immediately following the physical exercise. The sauna was done in one period each day with short breaks for cooling shower or additional exercise. Flush Niacin (100 mg at start up to 5000 mg per day) was used in gradually increasing doses daily for days. 53

Conference on Hazardous Wastes and Environmental Emergencies, Exercise, forced sweating and niacin all act to mobilize fat, resulting in significant turnover of adipose tissue stores and consequent mobilization of the stored lipophilic toxic chemicals. Water, salt and potassium were taken as needed to avert dehydration or salt depletion, due to the concentrated sweating. Polyunsaturated (Allblend) oil, from 2-8 tablespoons daily based on individual tolerance, was used to overcome entero-hepatic recirculation because, as the fat-soluble toxic chemicals are released into the blood stream, many of them are carried to the gut where they are released intraluminally. Reabsorbed into the lipophilic bile acids and recirculated back through the liver. 54

A considerable portion (10% to 15%) of the toxic materials excreted through the body will come out through the sebaceous sweat. Calcium, magnesium and other minerals were added to replace those lost through the copious sweating from the sauna exposure. 55

57 Sauna Detoxification Suggestions Step 1 take Niacin (immediate release) at 50mg to 100mg per dose to induce vasodilation and the subsequent niacin flush If no flushing occurs at these dosages (rare) then increase by 50mg per dose. If become accustomed to niacin dose, then increase dose until flushing occurs again. Step 2 Begin exercise approximately 20 minutes after taking niacin. The exercise (preferably aerobic) should induce sweating. Exercise for 20 to 30 minutes. Stay hydrated. 57

58 Sauna Detoxification Suggestions Step 3 Begin sauna. Have sauna preheated to 130 to 140 degrees (typical for radiant heat sauna) or 100 to 110 degrees (infra-red sauna). Ideal time in sauna is one-hour: If new to sauna therapy, okay to start at shorter durations, e.g. 20 to 30 minutes. This is important for toxic and/or heat sensitive individuals. If possible, wipe sweat off body frequently with clean towel while dosing sauna. If access to warm shower do periodically during session. Drink water and/or electrolyte drink during sauna session. Coconut water, e.g. Zico. Do sauna 5 to 6 days per week for 6 to 8 weeks. Step 4 After sauna, take activated charcoal/clay product, then immediately shower. Can use polyunsaturated oil (e.g. Allblend) 2 to 3 tablespoons. 58

71 Sauna Recommendations Unable to purchase infra-red sauna. Needs to use local park and recreation facility for sauna and shower. Sauna session at least 5 times weekly (minimum 3 times weekly). Maximum 60 minutes per session with short breaks to shower every 15 to 20 minutes. Stop sauna session if feel weak, nauseous, lightheaded, etc. Prior to sauna: Niacin start at 50mg per dose. Work up to tolerable amount until flushing effect is achieved. Take Niacin approximately 20 minutes before exercise. Aerobic exercise for 30 minutes (approximately) prior to sauna. Maintain hydration via water and electrolyte drink, e.g. ZICO. Shower immediately after completing sauna session. Repeat GPL-TOX Profile in 60 days. 71
