4 A Brief Overview of MODcol s Product Lines MODcol pioneered Axial Compression Spring Columns, a novel concept for preparative HPLC chromatography that features a powerful self-contained axial compression mechanism. As the volume occupied by the media gradually contracts, this mechanism expands to maintain a high density in the packed column, yielding high efficiency and increasing column lifetime. The MultiPacker is an instrument for on-site slurry packing of Spring Columns. This instrument packs Spring Columns in only a few minutes, start to finish. The MultiPacker and Spring Column are designed as separate units. Once a column is packed, the MultiPacker is immediately ready to pack another column while the packed Spring Column is relocated to the point of use. Used together, the Spring Column and MultiPacker provide a dramatic cost savings over conventional dedicated packing systems. MODcol s Patented Fixed Bed Flanged Chromatography Columns are widely accepted as the standard for high performance prep-lc. MODcol s patented flow distribution system provides uniform flow across the top of the media bed at the column inlet and outlet, yielding analytical level performance in a prep-scale column. 6 Product Spotlight: Axial Compression Spring Columns The Spring Column products are MODcol s highest performance chromatography columns. Spring Columns have an integrated axial compression system that delivers high performance in a small and portable package. Axial Compression Spring technology separates the axial compression that is used during column packing (requiring a large displacement) from the axial compression used during column use (requiring a small displacement). This results in a lighter, smaller and less expensive axial compression system that is fully contained within the body of the Spring Column. Fail-safe and fault-tolerant operation is ensured by the uncomplicated, purely mechanical design of the axial compression system. Some products available today from other manufacturers are continually powered by bulky hydraulic systems that must be mounted on a cart and some require tedious manual actuation during packing while providing only marginal or unreliable performance. In contrast, MODcol s Axial Compression Spring system provides industry-leading chromatographic performance in an easy-to-use package. The Technology Behind the Spring Column All axial compression systems compensate for void formation that can occur under certain circumstances commonly found in preparative-scale chromatography columns. Typically, a movable piston defines the end of the column at which the inlet flow is located. A force is continuously used to push the piston against the bed of the packed column. The force used to push the piston against the bed may be active or passive, mechanical or hydraulic. This action allows the piston to maintain contact with the bed when it consolidates or when particles are physically degraded and washed out of the column. In traditional columns designed with fixed bed geometry, bed consolidation can lead to an internal void or an uneven bed at the inlet side causing peak tailing, broadening or splitting. Such severe performance degradation renders the column useless for high performance separations. In general, axial compression systems must compensate for void formation as discussed earlier and a careful consideration of the void volume can be used to show that a spring driven system is well suited for this task. Most columns used in modern preparative chromatography are 25cm and shorter in length. Even the worst modern chromatographic resin will not consolidate more than five to ten percent in a packed bed. At these high levels of bed settling, other problems such as frit blockage or extensive media collapse would render the column unusable. It is therefore sufficient to compensate for voids on the order of one centimeter in length, roughly equal to five or ten percent of the bed length. Over this length, a spring can be used to provide the force necessary for axial compression. A spring-driven axial compression system does not operate in the same manner as a hydraulic powered one. In a spring, the restorative force increases from zero as the spring is compressed. In order for the spring to perform useful work it must be under a minimum amount of compression at all times. To advance the piston, the force generated by the spring must be greater than the static frictional forces. The spring force must also be sufficient to counteract the hydrostatic force that is generated by the pressure of the liquid within the column acting upon the piston head. 7 The limits discussed in the previous paragraph define a window of operation for the spring. Consider the figure below. Illustration of force vs. compression relationship for a spring. When the spring is fully compressed, the force is greatest indicated by Lmin, Fmax. In the spring column design, one end of the spring is locked in place while the other is free to push against the piston. As the spring uncompresses to push the piston along the column, the force it is generating decreases. When the force is equal to the hydrostatic force in the column plus the static frictional force, the spring can no longer advance the piston and the axial compression action is lost. This is indicated by L0,F0 on the graph. If the hydrostatic pressure is reduced to zero, i.e. the pump is shut off, the piston could advance but when the pump was restarted the piston could be pushed back to L0. This situation is easily overcome by returning the column to the MultiPacker instrument and applying hydraulic pressure to recompress the spring back to Lmin, Fmax. Experience has proven that, under most real-world conditions, the Spring Column provides dramatically improved performance over similar fixed bed columns. This improvement occurs in both performance and longevity. In addition, Spring Column performance is very reproducible from column to column when compared to fixed bed columns. This translates to dependable high performance at little additional cost. MODcol Spring Columns are available for use with the MultiPacker column-packing instrument for users who would like to pack the columns on site using their own media and/or procedures. The Spring Column is also available as a ready-to-use packed column, a feature that is unique among axial compression columns on the market today. The columns used above were 50mm ID Spring Columns packed to a bed depth of 15cm using 20µ Chiracel OD. Each column was packed only once. 3

8 Product Spotlight: Fixed Bed Flanged Columns MODcol s high quality fixed bed flanged column hardware has been awarded several patents for its design. All of our flanged column products are constructed from a high quality 316 stainless steel alloy that is electro-polished to provide the highest possible corrosion protection and chemical inertness. The flanged construction is designed to be rugged and is very conservatively rated at 2000psi. The column closure is achieved using a set of highstrength bolts and two heavy-duty endplates that are engineered to provide a long product lifetime. The simplicity of this design, without threads or other parts that can wear out, makes the column hardware practically infinitely reusable and repackable. Central to the ability to provide near-analytical levels of performance in preparative scale columns is the flow distribution system. MODcol s column designs promote homogeneous flow across the boundaries of the media bed to reduce band broadening. Pure Teflon seals provide maximum inertness and solvent compatibility under all conditions. All frits used in MODcol columns are made by sintering stainless steel powder into a porous disc. These critical parts are sourced from suppliers who provide this material with a high open area (area available for flow) because this ensures a negligible pressure drop across the frit. Special care is taken to passivate the internal surfaces of the frits so that they are inert towards biological materials. MODcol fixed bed columns are offered in a variety of inner diameters and lengths to suit a wide range of needs. 10 Bulk Media DaisoGel Daiso s SP-P (spherical ultra pure) series is based on a spherical and totally porous silica gel with a low level of metal impurities (specified to be less than 10 ppm each for Al, Fe, Ti and Zr). This packing material also features a narrow particle size distribution, high loading capacity and enhanced mechanical strength. DaisoGel is a workhorse, production-grade media for large-scale applications. Even with such advanced features it represents a very cost effective media for all preparative applications. We offer 10µm, 15µm and 20µm particle sizes and 120Å pore diameters, however other particle sizes and porosities are available upon request. The selection of 300Å pore is due to the fact that larger polypeptides must enter the pore interior to interact with the majority of the stationary phase. This allows separations that are not possible when separating large polypeptides on media having 100Å pores. At the same time, wide pore silica are well-suited for the separation of small peptides. 28 Application-Specific Prepacked Columns Columns for Chiral Chromatography Kromasil s Chiral phases are covalently bonded to the same high performance spherical silica used throughout their product line. The covalent attachment of the bonded phase to the support material increases the chemical stability above the level achieved by other manufacturer's chiral materials. Kromasil chiral phases feature the same high mechanical strength, chemical purity, stability and well-defined pore structure found in other Kromasil materials. This media features a narrow particle size distribution for high efficiency, low-pressure drop and best total economy in chromatographic purification. 30 Application-Specific Prepacked Columns Columns for Rapid and High-Throughput Separations MODcol delivers the highest quality columns for rapid LC applications by combining 20 years of know-how and expertise in column hardware, packing procedures and media to create a product line that is specifically designed for this demanding application. A new proprietary packing process has been developed that delivers outstanding performance with short columns. MODcol is offering both flanged and threaded columns in its high-throughput separations product line. To improve the sample throughput, a common solution is to increase the flow of the mobile phase. High flow produces very high shear forces within the bed. Shear-driven media degradation generates fine debris that clogs the outlet frit and increases backpressure, eventually making the column unusable. 33 Application-Specific Prepacked Columns Columns for Purification of Peptides and Proteins The separation of proteins and polypeptides has become one of the largest applications in semi-preparative and preparative HPLC. With almost 20 years of experience, MODcol has created a complete solution for the protein chemist wanting to quickly and effectively purify proteins and peptides. MODcol has selected two media for this demanding application: the Vydac 300 Å and the Daiso 300 Å, both exhibiting very good separations and complementary selectivity. The selection of 300 Å pore is due to the fact that larger polypeptides must enter the pore interior to interact with the majority of the stationary phase. This allows separations that are not possible when separating large polypeptides on media having 100 Å pores. At the same time wide pore silica are well-suited for the separation of small peptides. 35 MultiPacker Instruments Series 2002 MultiPacker Instruments MultiPacker instruments are high-performance systems designed to pack MODcol Spring Columns. Their ease of operation and low cycle time provide a self-contained and versatile column packing solution. Spring Columns offer outstanding convenience and flexibility when used with the MultiPacker Instrument. The ability to continuously vary bed length up to 30cm and the advantages of its spring-driven axial compression mechanism provide a high performance, adaptive solution for both the product development and the demanding process chromatography arenas. Furthermore, compared to traditional column packing processes, MODcol Spring Columns require significantly less media per packing because the bed is formed from the entire quantity of media that is introduced into the column without wastage. More columns can be packed from the same batch of media, reducing concerns regarding lot-to-lot variations. Also, by reducing media requirements, the cost of the MultiPacker may be recouped in as little as a few months time. Unlike other axial compression systems, the MODcol Spring Column is selfcontained. Once packed, the Spring Column is independent of the MultiPacker Instrument. Subsequently, one MultiPacker can be used to maintain an entire fleet of Spring Columns. For the first time, the chromatographer has the ability to pack columns on demand in order to quickly respond to in-house column needs. Series 2002 Multipacker Instruments. The MultiPacker provides several unique advantages to the chromatographer. You will find peace of mind by being independent from column packers and manufacturers. In addition, you have the ability to pack columns when and where they are needed. Money is saved with every column packed because less media is required. You have control of bed length and column volume and columns can be unpacked and repacked in a matter of minutes. Configure the Spring Column to use the optional axial compression subsystem when you wish to extend column lifetime and eliminate voids or use the MultiPacker to pack spring columns without the axial compression subsystem in order to maintain a constant bed volume. Because axial compression systems are able to pack columns at a lower pressure the performance of less robust or economical media is superior when the MultiPacker is used. Less mechanical damage is imparted to media when packed using the MultiPacker making it a perfect match to delicate media that is not bonded to silica. Unlike other column packing instruments that use an awkward hand pump and can require hours for column packing, from beginning to end, Spring Column packing takes only minutes and operates with the push of a button. The MultiPacker uses modular construction techniques that make it lightweight and easily assembled or disassembled. Many key components use advanced aluminum casting techniques that reduce manufacturing costs and provide high strength. The MultiPacker Series 2002 instruments are easy-to-use packing stations specifically designed to be used with the MODcol Spring Columns.
