Media used in plant tissue culture generally consist of mineral salts, sugars, vitamins, amino acids, growth regulators, and a semi-solid support of agar or gelrite. The minerals are needed by plant cells as building blocks for the synthesis of organic molecules, or as catalysts for enzymatic reactions. The ions in the media play an important role in the transport of molecules across the membranes, in osmotic regulation and in maintaining the electrochemical potential across the cell membranes. Mineral Elements The minerals in plant tissue culture media are divided into macro- and micro-elements. Ca, P, S, Mg, N and K are considered to be macro-elements while Fe, Cu, Mn, Co, Mo, B, I, Ni, Cl and Al are considered micro-elements. This categorization of elements into macro- and micro- is based on the quantities of the elements necessary to support healthy cell growth. The quantity of micro-elements (or trace elements) in the media is not a representation of the importance of these elements to the plant. For example, an iron deficiency can have catastrophic effects on the growth and development of a plant. The necessity of micro-elements such as cobalt and aluminum is not very clear but certainly their presence in trace amounts in the medium display some growth benefits. It is necessary that the plant cells receive all nutrients in balance thus making cations and anions equally available to support plant cell growth. For example, of potassium nitrate, both (K) and (NO) are essential. Within the group of macroelements the nitrogen containing salts, such as potassium-, ammonium- and calcium nitrate, are among the most plentiful. Ammonium ions in excess may inhibit growth. All macro and micro elements must be supplied in a pure form, free of other elements and heavy metal contaminants. Vitamins Vitamins are needed in the plant media in small concentrations and must be supplied in natural forms. These compounds are essential for many biochemical reactions. All media for plant cell and tissue culture contains thiamine (vitamin B1). Linsmaier and Skoog, after a thorough revision of the vitamins present in the medium, concluded that Thiamine is essential to plant growth. Inositol significantly stimulates the growth and development of plants but it does not appear to be essential for growth. The effect of vitamins on the development of the cell in vitro differs from species to species, and therefore the concentration of vitamins needs to be adjusted for each explant.
