MTT assay and Cell proliferation: Treatment of cell with the various drugs and dose dependent inhibition of cell viability, and cell proliferation was performed. Flow cytometry: Flow cytometric analysis for immunophenotyping by staining membrane associated and intra cellular proteins. Apoptosis assay: Acquisition and analysis of apoptotic cells by Flow cytometer against drug treatment using AnnexinV/PI staining procedure. Cell cycle analysis: Staining of drug treated cells with PI. Cells were analyzed with a fluorescence activated cell sorter analysis (FACS), and data was analyzed by FACS Caliber software (BD, USA). Microscopy: Preparation of slides for Immunofluorescence and Fluorescence microscopy. 3

4 Gene silencing: Transfection and sirna mediated gene silencing. DNA/RNA/PCR Techniques: DNA related techniques: Genomic DNA and plasmid DNA isolation, Agarose gel electrophoresis and RNA isolation, Reverse Transcriptase-PCR and Real Time PCR. HPV screening and Genotyping using PCR from Biopsy samples of cervical cancer Primer Deigning: To check expression profiles of the genes, polymorphism, and Methyl specific PCR primer using online available software tools. MSP (Methyl specific-pcr): To study of Promoter Hypermethylation of Tumor suppressor gene, Bisulfite Conversion of the genomic DNA was performed. RFLP: Restriction digestion of DNA and Agarose gel electrophoresis to check polymorphism. Comet assay (single-cell gel electrophoresis): To check the extent of DNA damage. Protein Techniques: Western Blotting: Native and SDS-PAGE, Western blot analysis (full length and Phospho specific immunodetection of the protein), Immunoprecipitation of the protein. Immunohistochemistry: Preparation of tissue block and slides of tissue section to check the expression and localization of the protein. Spectroscopic Techniques: Quantification of Protein by Spectrophotometer and expression of protein by ELISA Microplate Reader.
